targeting multiple immune mechanisms may overcome therapy resistance and further improve cancer immunotherapy for ***,we describe the application of virus-like vesicles(VLV)for delivery of three immunomodulators alone and in combination,as a promising approach for cancer *** vectors were designed to deliver single chain interleukin(IL)-12,shorthairpin RNA(shRNA)targeting programmed death ligand 1(PD-L1),and a dominant-negative form of IL-17 receptor A(dn-IL17RA)as a single payload or as a combination *** delivery of the VLV vector expressing IL-12 alone,as well as the trivalent vector(designated CARG-2020)eradicated large established ***,only CARG-2020 prevented tumor recurrence and provided long-term survival benefit to the tumor-bearing mice,indicating a benefit of the combined *** abscopal effects of CARG-2020 on the non-injected contralateral tumors,as well as protection from the tumor cell re-challenge,suggest immune-mediated mechanism of protection and establishment of immunological ***,CARG-2020 potently activates th1 immune mechanisms and inhibits expression of genes related to t cell exhaustion and cancer-promoting *** ability of CARG-2020 to prevent tumor recurrence and to provide survival benefit makes it a promising candidate for its development for human cancer immunotherapy.

AIM： tO determine the possible modulating effect of the COX-2 polymorphisms, -765G→C and -1195A→G, on the risk of colorectal cancer （CRC） in a Dutch population. MEtHODS： this case-control study includes 326 patients with CRC and 369 age- and gender-matched controls. Genotypes of the COX-2 polymorphisms -7dEG→C and -1195A→G were determined by polymerase chain reaction-based restriction fragment length polymorphism. COX-2 genotypes and haplotypes were analyzed and odds ratios with 95% confidence intervals were estimated by logistic regression. RESULtS： the -765GG genotype was associated with an increased risk of developing CRC （OR, 1.45; 95% CI, 1.03-2.04）. No significant difference was observed in the genotype distribution of the -1195A→G polymorphism between patients and controls. the GG/AC haplotype was present significantly less often in patients than in controls （OR 0.44; 95% CI, 0.22-0.85）. When the AC, AG and GG haplotypes were investigated separately, the AC haplotype showed a tendency to be less frequent in patients than in controls （OR（AG/AC） 0.78; 95% CI, 0.57-1.06）. CONCLUSION： the -765GG genotype is associated with an increased risk of developing CRC and the G6/ AC haplotype seems to protect against CRC. these findings suggest a modulating role for the COX-2 polymorphisms -765G→C and -1195A→G in the development of CRC in a Dutch population.

In plants, RNA-directed DNA methylation （RdDM） plays an essential role in silencing transposable elements that would otherwise have a deleterious effect on genome integrity. RdDM is an important pathway that establishes and maintains de novo DNA methylation in all three sequence contexts： CG, CHG, and CHH （where H is A, C, or t）, and the methylation is targeted by 24-nt small interfering RNAs （siRNAs）（Law and Jacobsen, 2010）.

transmasculine individuals,considering whether to undergo total hysterectomy with bilateral salpingectomy,have the option to have a concomitant *** studies have evaluated hormone changes following testosterone therapy initiation,most of those patients have not undergone *** are currently limited to support health outcomes regarding the decision to retain or remove the *** performed a retrospective chart review of transmasculine patients maintained on high-dose testosterone therapy at a single endocrine clinic in Vancouver,British Columbia,*** transmasculine individuals who underwent bilateral oophorectomy and had presurgical and postsurgical serum data were *** identified 12 transmasculine subjects as controls,who were on testosterone therapy and did not undergo oophorectomy,but additionally matched to the first group by age,testosterone dosing regimen,and body mass *** was a statistically significant decrease in the estradiol levels of case subjects postoophorectomy,when compared to presurgical estradiol levels(P=0.02).there was no significant difference between baseline estradiol levels between control and case subjects;however,the difference in estradiol levels at follow-up measurements was significant(P=0.03).total testosterone levels did not differ between control and case subjects at baseline and follow-up(both P>0.05).Our results demonstrate that oophorectomy further attenuates estradiol levels below what is achieved by high-dose exogenous testosterone *** clinical outcomes,such as impacts on bone health,were not *** clinical implications of oophorectomy versus ovarian retention on endocrinological and overall health outcomes are currently limited.

this paper addresses men＇s perspectives on the relationship between female genital mutilation/cutting （FGM/C） and women＇s sexuality in Egypt with the purpose of studying how men and women see a relationship between FGM/C and sexual life, the study used qualitative methods and was conducted in three sites. Men saw FGM/C as the key to ensuring women＇s sexual morality, and considered FGM/C as the key to controlling sexual relationships. Men were reported to increasingly take part in the decision-making about daughters＇ FGM/C. In men＇s view, FGM/C reduces female sex drive and thereby keeps girls chaste before marriage and women faithful during marriage. Men wholeheartedly support FGM/C for these reasons. However, others are ambivalent due to a concern that the practice might have negative consequences on their own sexuality as a consequence of a negative effect on their wives＇ sexual feelings. the study concludes that men＇s role is important and that they should be involved in community activities against the practice. It also concludes that without comprehensive sexuality education, men will continue to hold on to the misconceptions that FGM/C has a positive effect on women＇s sexual morality through reducing their sexual desire.
