AIM: To determine the role for the intermediate filament protein nestin in glioma invasion. MeTHODS: We examined the expression and function of nestin in gliomas(Grades Ⅱ-Ⅳ as defined by the World Health Organization). We determined nestin expression using Immunohistochemical methods. To elucidate nestin's biological function(s), we reduced m RNA levels by 61% and 87% in two glioblastomaderived neurosphere lines using short hairpin RNAs and determined the effect of reduced nestin expression on glioma cell proliferation and invasion using MTS and matrigel migration assays, respectively. We also utilized quantitative real time polymerase chain reaction assaysto determine the effect of reduced nestin expression on the expression of other markers associated with glioma stem cells and their differentiated progenies. ReSULTS: We found a significant correlation between nestin immunoreactivity and astrocytoma tumor grade, with 36% of grade Ⅱ, 75% of grade Ⅲ, and 100% of grade Ⅳtumors expressing significant levels of the protein when assessed using immunohistochemistry. Reduction in nestin expression had no effect on cell growth in culture, but did retard the capacity of one line to migrate in-vitro on matrigel. Interestingly, in the line whose migration was not affected, m RNA levels of a second intermediate filament, synemin(also knowns as desmuslin), were elevated following introduction of sh RNA targeting nestin. As synemin was not induced in the line which required nestin for migration, it is a possibility that synemin may compensate for the loss of nestin in this process. CONCLUSION: Nestin expression is prominent in high-grade astrocytomas. Nestin is not required for cell growth but it may, however, be required for cell motility.

Species delimitation based on diagnosis and monophyly, and its importance for advancing mammalian taxonomy

A recently proposed taxonomic classification of extant ungulates sparked a series of publications that criticize the Phylogenetic Species Concept （PSC） claiming it to be a particularly poor species *** opinions reiteratively stated that （1） the two fundamental elements of the ＂PSC＂,i.e.,monophyly and diagnosability,do not offer objective criteria as to where the line between species should be drawn;and （2） that extirpation of populations can lead to artificial diagnosability and spurious recognitions of *** sudden eruption of criticism against the PSC is *** attributed to the PSC are common to most approaches and concepts that modern systematists employ to establish species *** controversial taxonomic propositions that sparked criticism against the PSC are indeed highly problematic,not because of the species concept upon which they are based,but because no evidence （whatsoever） has become public to support a substantial portion of the proposed *** herein discuss these topics using examples from *** areas of biological research rest upon taxonomic accuracy （including conservation biology and biomedical research）;hence,it is necessary to clarify what are （and what are not）the real sources of taxonomic inaccuracy.

Group actions on treelike compact spaces

We show that group actions on many treelike compact spaces are not too complicated *** first observe that an old argument of Seidler(1990)implies that every action of a topological group G on a regular continuum is null and therefore also *** every local dendron is regular,one concludes that every action of G on a local dendron is *** then use a more direct method to show that every continuous group action of G on a dendron is Rosen thal represent able,hence also *** resul ts are obtained for median *** a related result,we show that Helly's selection principle can be extended to bounded monotone sequences defined on median pretrees(for example,dendrons or linearly ordered sets).Finally,we point out some applications of these results to continuous group actions on dendrites.

Possible therapeutic role of Ig e blockade in irritable bowel syndrome

Omalizumab is a humanized monoclonal antibody that binds to the high-affinity type-I Ig e Fc receptors on mast cells(MCs) and basophils, inhibiting the Ig e immune pathway. Irritable bowel syndrome(IBS) is the most common functional gastrointestinal disorder, and dysregulation of the immune system likely contributes to its etiology and/or symptomatology. Colonic biopsies from patients with IBS demonstrate considerable increase in the number of degranulating MCs releasing histamine in proximity to nerves, and this event may underlie the common IBS symptom of abdominal pain. Pharmacologic control of MC activation and mediator release is a current area of active interest in the field of IBS research. Recently, we and Pearson et al described 2 cases of patients with IBS with diarrhea(IBS-D) showing positive clinical response to omalizumab. In both cases, the female patients had severe, long-lasting IBS-D and achieved an almost complete resolution of IBS symptoms. Both patients were also able to discontinue all IBS medications after commencing the anti-Ig e therapy. For both patients, the omalizumab treatment showed a relatively rapid onset of action, resembling the efficacy observed in and previously reported for patients with chronic spontaneous urticaria. In this editorial, we discuss the possible biological mechanisms that may underlie the clinical efficacy of omalizumab in IBS. We suggest that there is a need for a well-designed prospective study to investigate the therapeutic effects of anti-Ig e in IBS.

What do we know about biological nitrogen fixation in insects? evidence and implications for the insect and the ecosystem

Many insects feed on a low‐nitrogen diet,and the origin of their nitrogen supply is poorly *** has been hypothesized that some insects rely on nitrogen‐fixing bacteria(diazotrophs)to supplement their *** fixation by diazotrophs has been extensively studied and convincingly demonstrated in termites,while evidence for the occurrence and role of nitrogen fixation in the diet of other insects is less ***,we summarize the methods to detect nitrogen fixation in insects and review the available evidence for its occurrence(focusing on insects other than termites).We distinguish between three aspects of nitrogen fixation investigations:(i)detecting the presence of potential diazotrophs;(ii)detecting the activity of the nitrogen‐fixing enzyme;and(iii)detecting the assimilation of fixed nitrogen into the insect *** show that although evidence from investigations of the first aspect reveals ample opportunities for interactions with potential diazotrophs in a variety of insects,demonstrations of actual biological nitrogen fixation and the assimilation of fixed nitrogen are restricted to very few insect groups,including wood‐feeding beetles,fruit flies,leafcutter ants,and a wood *** then discuss potential implications for the insect's fitness and for the ecosystem as a *** suggest that combining these multiple approaches is crucial for the study of nitrogen fixation in insects,and argue that further demonstrations are desperately needed in order to determine the relative importance of diazotrophs for insect diet and fitness,as well as to evaluate their overall impact on the ecosystem.

Hospital costs, length of stay and prevalence of hip and knee arthroplasty in patients with inflammatory bowel disease

To examined the prevalence of hip and knee arthroplasty in patients with inflammatory bowel disease (IBD) by comparing the diagnostic codes for these procedures in patients with IBD and a control group of patients. MeTHODSThe National Inpatient Sample database (NIS) is part of the Healthcare Cost and Utilization Project (HCUP), the largest publicly available inpatient healthcare database in the United States. The NIS samples about 20% of discharges from all community hospitals participating in HCUP, representative of more than 95% of the United States population, with approximately 7000000 hospitalizations reported annually. NIS contains data on diagnoses, procedures, demographics, length of stay (LOS), co-morbidities and outcomes. ICD-9-CM diagnostic codes for primary hospitalizations for arthroplasty of the hip or knee with a co-diagnosis of IBD [combining both Crohn’s disease (CD) and ulcerative colitis (UC)] were used to identify study subjects for cost and LOS analysis for NIS from 1999-2012. Statistical analysis: 1: 2 propensity score matching between IBD vs a control group based on following factors: Patient age, gender, race, total co-morbidities, # of procedures, admission type, insurance, income quartiles, and hospital bed size, location and hospital teaching status. Categorical variables were reported as frequency and compared by χ2 tests or Fisher’s exact tests. Individual 1:3 matching was also performed for patients carrying diagnostic codes for CD and for patients with the diagnostic code for UC. After matching, continuous variables were rcompared with Wilcoxon signed rank or Paired T-tests. Binary outcomes were compared with the McNemar’s test. This process was performed for the diagnosis of hip or knee arthroplasty and IBD (CD and UC combined). Helicobacter pylori infection in patients with selective immunoglobulin e deficiency

AIM: To investigate the prevalence and clinical characteristics of Helicobacter pylori(***)-infected dyspeptic patients with selective immunoglobulin e deficiency(Ige d).MeTHODS: All individuals who underwent serum totalimmunoglobulin e(Ig e) measurement at the Leumit Healthcare Services(Israel) in 2012 were identified in an electronic database search(n = 18487).From these,selected case group subjects were ≥ 12 years of age and had serum total Ig e e control group was selected from a random sampling of the remaining subjects ≥ 12 years of age to obtain a case-control ratio of 1:20(n = 3160).Dyspeptic diseases,diagnosed no more than 5 years before serum total Ig e testing,were identified and retrieved from the electronic database using specific International Classification of Diseases diagnostic *** of C13-urea breath tests were used to identify subjects infected with *** variables between case and control subjects were analyzed using Fisher's exact tests,whereas continuous variables were analyzed using χ2 ***: Dyspepsia was present in 27.2%(43/158) of case subjects and 22.7%(718/3160) of *** these,significantly more case subjects(32/43,74.4%) than controls(223/718,31.1%) were positive for ***(P esophagogastroduodenoscopy was performed in 19 case and 94 control subjects,revealing that gastritis was more prevalent in Ige d case subjects than in controls(57.9% vs 29.8%,P ermore,a significantly greater proportion of case subjects presented with peptic duodenal ulcers(63.2% vs 15.9%,P examination showed marked chronic inflammation,lymphoid follicle formation and prominent germinal centers,with polymorphonuclear cell infiltration of gastric glands,that was similar in case and control biopsy ***,Ig ed case subjects that underwent esophagogastroduodenoscopy were more likely to exhibit treatment-refractory *** infections that require seco
