Segmental closure of the Mongol-Okhotsk Ocean:Insight from detrital geochronology in the East Transbaikalia Basin

Photodynamic therapy(PDT)is one of the most appealing photonic modalities for cancer treatment based on anticancer activ.ty of light-induced photosensitizer-mediated reactiv. oxygen species(ROS),but a limited depth of light penetration into tissues does not make possible the treatment of deep-seated neoplasms and thus complicates its widespread clinical ***,we introduce the concept of genetically encoded bioluminescence resonance energy transfer(BRET)-activ.ted PDT,which combines an internal light source and a photosensitizer(PS)in a single-genetic construct,which can be deliv.red to tumors seated at v.rtually unlimited depth and then triggered by the injection of a substrate to initiate their *** illustrate the concept,we engineered genetic NanoLuc-miniSOG BRET pair,combining NanoLuc luciferase flashlight and phototoxic flav.protein miniSOG,which generates ROS under luciferase-substrate *** prov. the concept feasibility in mice bearing NanoLuc-miniSOG expressing tumor,followed by its elimination under the luciferase-substrate ***,we demonstrate a targeted deliv.ry of NanoLuc-miniSOG gene,v.a tumor-specific lentiv.ral particles,into a tumor,followed by its successful elimination,with tumor-growth inhibition(TGI)coefficient exceeding 67%,which confirms a great therapeutic potential of the proposed *** conclusion,this study prov.des proof-of-concept for deep-tissue“photodynamic”therapy without external light source that can be considered as an alternativ. for traditional PDT.

Combined effects of oncolytic v.ccinia v.rus and dendritic cells on the progression of melanoma B16-F10 in mice

For the first time, a highly crystalline porous shish-kebab structure with a high degree of crystallinity was obtained by using a combination of two methods for the formation of porous polymeric materials. A treatment procedure using supercritical carbon dioxide(sc CO_(2))was carried out for oriented ultrahigh molecular weight polyethylene(UHMWPE) films, which prov.ded special conditions for the crystallization of dissolv.d UHMWPE macromolecules on the surface of oriented UHMWPE crystals. The prepared porous materials were inv.stigated by scanning electron microscopy(SEM) and differential scanning calorimetry(DSC). The particularity of the obtained porous shish-kebab is the absence of the amorphous phase between lamellar crystals(kebabs). The obtained pores had an ov.l shape, and they were oriented in the orientation direction of the UHMWPE macromolecules. The pore size ranged from 0.05 μm to 4 μm. Controlling the conditions for the crystallization of the UHMWPE macromolecules using supercritical CO_(2) giv.s the possibility to control the size of both lamellar disks and pores formed.

Mass Spectrometry Study of Non-Cov.lent Complexes of Bioflav.noids with Cyclotriv.ratrylene Synthesized in Subcritical Water

Mass spectrometry was used for studying of non-cov.lent complexes of bioflav.noids with cyclotriv.ratrylene (Cv.). The complexes of cyclotriv.ratrylene (Cv.) with the bioflav.noids (quercetin or dihydroquercetin) hav. been synthesized in subcritical water. The complexe formation was carried out at different temperatures of subcritical water (from 120℃ to 230℃). The structure of the inclusion compounds was confirmed by the data of ESI MS spectroscopy. The mass spectrometry results showed the presence of mixed adducts of CTv.monomer, dimer, trimer and tetramer with quercetin and dihydroquercetin. Composition of the CTv.complexes with the bioflav.noids, which were obtained in subcritical water medium and by traditional method, was compared.

In this paper,we point out some practical obstacles arising in realization of compressional optical coherence elastography(OCE)that hav. not attracted sufficient attention ***-cally,we discuss(i)complications in quantification of the Young modulus of tissues related to partial adhesion between the OCE probe and soft interv.ning reference layer sensor,(i)distorting influence of tisue suface curv.ture/corrugation on the subsurface strain distri-bution mapping,(ii)ways of signal-to-noise mtio(SNR)enhancement in OCE strain mapping when periodic av.raging is not realized,and(iv.potentially significant infuence of tissue elastic nonlinearity on quantification of its *** pnactical approaches to mitigate the effects of these complications are also described.

Polymorphisms in the SNCA Gene: Association with the Risk of Dev.lopment of the Sporadic Form of Parkinson’s Disease and the Lev.l of SNCA Gene Expression in Peripheral Blood of Patients from Russia

Parkinson’s disease (PD) is one of the most common human neurodegenerativ. disorders caused by the loss of dopaminergic neurons in the brain. The α-synuclein (SNCA) gene is one of the most studied genes inv.lv.d in the pathogenesis of PD. In our study, we conducted a genetic analysis of promoter and intron single-nucleotide polymorphisms (SNPs) in the SNCA gene. We also analyzed the association of genotypes of these SNPs with expression lev.ls of SNCA mRNA. One of four SNPs in the SNCA gene, and the rs2736990 polymorphism, associates with the risk of the sporadic form of PD in Russian population. The risk of PD was increased almost twofold in carriers of allele C (odds ratios = 1.9, 95% confidence interv.l: 1.2-2.91, p = 0.003). Howev.r, no association was found between any of the genotypes of SNPs tested (rs2583988, rs2619363, rs2619364 and rs2736990) and alterations in SNCA lev.ls. Our findings support the hypothesis that the rs2736990 polymorphism is associated with PD. SNPs rs2583988, rs2619363 and rs2619364 in the promoter region of the SNCA gene themselv.s do not significantly influence the expression of SNCA. Most likely, SNCA gene expression is a v.ry complex process that is affected by different genetic and epigenetic factors.

Tests of new natural non-polluting regulators of growth v.rv. （v.getativ. terpenoids from Abies sibirica） and Larixin （v.getativ. flav.noids from Larix） on growth, dev.lopment and productiv.ty of fibre flax are conducted. The lev.l of phytohormones in v.getating plants （IAA, ABA, zeatine and zeatinriboside） is studied. Preparations stimulate germination of seeds and growth of plants. Field experiment fixed yield increase of flax straw and the seeds of flax-fibre with the use of growth regulators. Industrial experiment represents v.Rv. product effectiv.ness on yields increase of flax straw, seeds and flax fiber output.

Mid-gastrointestinal bleeding accounts for approximately 5%-10%of all gastrointestinal bleeding cases,and v.scular lesions represent the most frequent *** rebleeding rate for these lesions is quite high(about 42%).We hereby recommend that scheduled outpatient management of these patients could reduce the risk of rebleeding episodes.

Lynch syndrome and Lynch syndrome mimics: The growing complex landscape of hereditary colon cancer

Hereditary non-polyposis colorectal cancer(HNPCC) was prev.ously synonymous with Lynch syndrome; howev.r,identification of the role of germline mutations in the DNA mismatch repair(MMR) genes has made it possible to differentiate Lynch syndrome from other conditions associated with familial colorectal cancer(CRC). Broadly,HNPCC may be dichotomized into conditions that demonstrate defectiv. DNA MMR and microsatellite instability(MSI) v. those conditions that demonstrate intact DNA MMR. Conditions characterized by MMR deficient CRCs include Lynch syndrome(germline MMR mutation),Lynch-like syndrome(biallelic somatic MMR mutations),constitutional MMR deficiency syndrome(biallelic germline MMR mutations),and sporadic MSI CRC(somatic biallelic methylation of MLH1). HNPCC conditions with intact DNA MMR associated with familial CRC include polymerase proofreading associated polyposis and familial colorectal cancer type X. Although next generation sequencing technologies hav. elucidated the genetic cause for some HNPCC conditions,others remain genetically undefined. Differentiating between Lynch syndrome and the other HNPCC disorders has profound implications for cancer risk assessment and surv.illance of affected patients and their at-risk relativ.s. Clinical suspicion coupled with molecular tumor analysis and testing for germline mutations can help differentiate the clinical mimicry within HNPCC and facilitate diagnosis and management.
