Advanced mid-infrared plasmonic waveguides for on-chip integrated photonics

Long-wave infrared(LWIR, 8–14 μm) photonics is a rapidly growing research field within the mid-IR with applications in molecular spectroscopy and optical free-space communication. LWIR applications are often addressed using rather bulky tabletop-sized free-space optical systems, preventing advanced photonic applications,such as rapid-time-scale experiments. Here, device miniaturization into photonic integrated circuits(PICs) with maintained optical capabilities is key to revolutionize mid-IR photonics. Subwavelength mode confinement in plasmonic structures enabled such miniaturization approaches in the visible-to-near-IR spectral range. However,adopting plasmonics for the LWIR needs suitable low-loss and-dispersion materials with compatible integration strategies to existing mid-IR technology. In this paper, we further unlock the field of LWIR/mid-IR PICs by combining photolithographic patterning of organic polymers with dielectric-loaded surface plasmon polariton(DLSPP) waveguides. In particular, polyethylene shows favorable optical properties, including low refractive index and broad transparency between ~2 μm and 200 μm. We investigate the whole value chain, including design, fabrication, and characterization of polyethylene-based DLSPP waveguides and demonstrate their first-time plasmonic operation and mode guiding capabilities along S-bend structures. Low bending losses of ~1.3 d b and straight-section propagation lengths of ~1 mm, pave the way for unprecedented complex on-chip mid-IR photonic devices. Moreover, DLSPPs allow full control of the mode parameters(propagation length and guiding capabilities) for precisely addressing advanced sensing and telecommunication applications with chip-scale devices.

Combined Endoscopic Transanal Vacuum-Assisted Rectal Drainage:A Novel Therapy for Colorectal Anastomotic Leak after TME for Cancer

Introduction: Low colorectal and colo-anal anastomosis leakage after anterior resection of rectum occurs in up to 24% of cases. Leak is the main cause of morbidity and mortality in postoperative period and can result in a permanent stoma in up to 25% of cases. Loop ileostomy or colostomy does not reduce the percentage of fistulas, but reduces flow rate. Conservative management imply long times to heal. Endosponge®system (b. braun Aesculap AG, Germany) was recently acquired in clinical practice. This system accelerates healing process through vacuum creation, removal of fluids and edema, infection control, local increase of blood flow and stimulation of granulation tissue formation. The device consists of a cylindrical polyurethane sponge, with 400 to 600 micron pores, standard dimension of 7 cm, which can be cut to fit the estimated size of the abscess cavity. The drain tube is connected to a Redyrob Trans Plus®bottle which exerts a constant suction during the therapy. Materials and Methods: From November 2012 and March 2014 four patients were treated with Endosponge®system, all of them underwent operation of anterior resection of rectum for cancer and developed anastomotic leakage during postoperative course. All patients had loop ileostomy, none presented signs of sepsis. The Endosponge®system was then placed endoscopically. In all cases, after the first placing, the authors utilized sterile foam dressing from negative pressure wound therapy kit, tailored to better fill the abscess cavity and connected with Redyrob Trans Plus?bottle. Device changes were performed every 5 - 7 days, according to the size of the cavity and the amount of secretions produced. The treatment was started during hospitalization first and then continued as outpatient. Results: There was no mortality. The amount of secretions was related to the initial size and the degree of contamination of the abscess cavity. Antibiotic therapy was discontinued after an average of 21 days,

Thymidine Glycol: The Effect on DNA Structure and DNA binding by Site-Specific Proteins

Thymidine glycol (5,6-dihydroxy-5,6-dihydrothymidine, Tg) is a major type of oxidative damage in DNA. During chemical oligonucleotide synthesis, Tg residue was incorporated in the different positions of 17 b.p. DNA duplexes, which differ in one base pair in the internal part. According to UV-melting curves, Tg destabilizes the double helix in a sequence independent manner. In contrast, the localized alterations in duplex structure were shown by CD spectroscopy to depend on the type of base pairs flanking the Tg lesion. Molecular dynamics simulations demonstrate that Tg is partially out of the double helix. For the first time, Tg impact on several site-specific DNA-binding proteins is studied, namely p50 and p65 subunits of nuclear factor kappa-b (NF-κb) and DNA methyltransferase SsoII (***). Our results show that p50/p50 and p65/p65 homodimers of NF-κb can tolerate a single Tg residue in the binding site quite well. Nevertheless the homodimers have different affinities to the oxidized κb site depending on the Tg position. *** can act as a transcription repressor when bound to the regulatory site. *** demonstrates decreased affinity and lowered methylation efficiency when its methylation site contains Tg in the central position. Single Tg in one half of the regulatory site decreases *** affinity to the oxidized DNA, whereas Tg presence in both half-sites prevents *** binding to such ligand.

Preserving microbial diversity has become a strategic undertaking. Thus, ex situ microalgal culture conservation results in strategic and functional resource in both biodiversity protection and application domains. Cryopreservation of microalgae has been practiced since the 1960s and is now considered the optimal preservation strategy. Furthermore, the overall monitoring during growth of cultures after freezing/thawing protocols was hardly investigated and there is poor evaluation related to preserve especially the photosystem apparatus. The present study focuses on Stichococcus bacillaris as case study for short-term cryopreservation at −80 °C storage. Various freezing pretreatments using cryoprotective agents, and two thawing methods were compared introducing a novel variable to evaluate viability recovery and assessing growth kinetics of cultures immediately after thawing and after a series batch cultivation. Photosynthetic rate and pigments assessment were proposed to evaluate hidden metabolic cell damage. Results underline cryoprotective agents can increase the kinetic recovery of preserved cells in terms of reduction of lag phase during batch cultivation tests: the use of dimethyl sulfoxide and glycerol granted a growth comparable to unpreserved cells when sudden thawing occurs after 24 hours of storage, but recovery after preservation is less sensitive to cryoprotective agents when gradual thawing and 1 month of storage is considered. However, cells are always able to restore their physiological pathways even without agents, so their kinetic effect has been proved and quantified. Interestingly, both the photosynthetic efficiency and the ratio between total chlorophyll and carotenoids are comparable (0.75 Fb>vb>/Fb>mb>, 2.2 ± 0.25 g/g) to unpreserved cells and they are unsensitive to chosen agents, but the ratio between chlorophyll a and chlorophyll b was clearly altered (up to 10 times), suggesting that photoactive pigments relative proportions

Liver transplantation for viral hepatitis in 2015

Liver transplantation(LT) is a life-saving treatment forpatients with end-stage liver disease and for patients with liver cell cancer related to liver disease. Acute and chronic liver diseases related to hepatitis viruses are between the main indications for liver transplantation. The risk of viral reinfection after transplantation is the main limiting factor in these indications. before the availability of antiviral prophylaxis, hepatitis b virus(HbV) recurrence was universal in patients who were HbV DNA-positive before transplantation. The natural history of recurrent HbV was accelerated by immunosuppression, and it progressed rapidly to graft failure and death. Introduction of post-transplant prophylaxis with immunoglobulin alone first, and associated to antiviral drugs later, drastically reduced HbV recurrence, resulting in excellent long-term outcomes. On the contrary, recurrence of hepatitis C is the main cause of graft loss in most transplant programs. Overall, patient and graft survival after LT for hepatitis C virus(HCV)-associated cirrhosis is inferior compared with other indications. However, successful pretransplant or post transplant antiviral therapy has been associated with increased graft and overall survival. Until recently, the combination of pegylated interferon and ribavirin was the standard of care for the treatment of patients with chronic hepatitis C. Highly active antiviral compounds have been developed over the past decade, thanks to new in vitro systems to study HCV entry, replication, assembly, and release.

神经外科作为高度专业化领域，常涉及对精细结构的手术操作，其中大脑牵拉术常用于触及大脑深层病灶，但目前尚无可靠的方法预防系列牵拉并发症的发生。本研究拟设计一种适用于经颅内镜手术的新技术，以保证周围脑组织的安全牵拉。新技术设计将可扩展腔插入颅内病灶点附近，置于最佳位置后即可被打开，保证了神经外科手术中可观的操作空间。本研究涉及使用力传感器评估牵拉阶段施加在脑组织上的压力值试验。翼点开颅术后，在牵拉过程中使用不同倾角的传统金属抹刀测量压力，尽管牵拉和施加在抹刀上的力相同，但抹刀边缘以不同倾角施加在实质脑组织上的局部压力变化却很大。综上，本研究通过设计一种腔式牵开器，用于大脑牵拉，避免由于边缘超压引起牵拉并发症，以保持施加在实质脑组织上的压力值适宜。

Photodynamic inactivation of microorganisms known as antibacterial photodynamic therapy(APDT)is one of the most promising and innovative approaches for the destruction of pathogenic *** the photosensitizers(PSs),compounds based on cationic porphyrins/metalloporphyrins are most successfully used to inactivate *** of meso-substituted cationic pyridylporphyrins and metalloporphyrins with various peripheral groups in the third and fourth positions of the pyrrole ring have been synthesized in *** aim of this work was to determine and test the most e®ective cationic porphyrins and metalloporphyrins with high photoactivity against Gram negative and Gram positive *** was shown that the synthesized cationic pyridylporphyrins/metalloporphyrins exhibit a high degree of phototoxicity towards both types of bacteria,including the methicillinresistant *** *** complexes of porphyrins are more phototoxic than metal-free porphyrin *** e®ectiveness of these Zn–metalloporphyrins on bacteria is consistent with the level of singlet oxygen *** was found that the high antibacterial activity of the studied cationic porphyrins/metalloporphyrins depends on four factors:the presence in the porphyrin macrocycle of a positive charge(+4),a central metal atom(Zn2þÞand hydrophobic peripheral functional groups as well as high values of quantum yields of singlet *** results indicate that meso-substituted cationic pyridylporphyrins/metalloporphyrins cannd wider application in photoinactivation of bacteria than anionic or neutral PSs usually used in APDT.

Cryopreservation by Vitrification of Vitis vinifera cv. ＂Red Globe＂ Zygotic Embryos and Effect on the Expression of DNA Methyltransferase Genes

Mexico is a large producer of table grape （Vitis vinifera L.） and therefore it is important to develop protocols to store the grape varieties germplasm. The objective of the present work was to design a protocol for the cryopreservation by vitrification of zygotic embryos of V. vinifera cv. ＂Red Globe＂ and evaluate possible epigenetics changes. The plant vitrification solution 2 （PVS2） was utilized before the utilization of liquid nitrogen （LN）. The effect of this protocol on embryo viability was tested by the triphenyl-tetrazolium chloride solution, as well as by the in vitro development of grape embryos into plantlet. A cDNA expression library of grape zygotic embryos was created to isolate expressed sequence tags of several DNA methyltrasferases. Gene expression of domains rearranged methyltransferase type 1 （DMR1） and DNA （cytosine-5）-methyltransferase 1 （MET1-2） isozymes was analyzed by quantitative reverse transcriptase PCR. The optimal conditions for vitrification were 10 min in 50% PVS2, followed by I0 min in 100% PVS2. Under these conditions, about 30% of plantlet was obtained from embryos after cryopreservation. It was recorded a reduction in the MET1-2 gene expression, which plays a role in the maintenance of DNA methylation. It is possible to cryopreserve viable grape zygotic embryos, although the treatment seems to induce alterations in the normal DNA methylation pattern of the zygotic embryo genome.
