Background:An increase in life expectancy has led to an increased elderly *** turn,this aging population is more likely to develop health conditions,such as pelvic floor disorders(PFDs).This study aimed to assess the prevalence of these disorders and the associated quality of life in institutionalized and noninstitutionalized elderly *** and methods:A cross-sectional study was conducted with 80 female participants older than 60 years,divided into 2 groups:institutionalized and noninstitutionalized *** Pelvic Floor Distress Inventory Short-Form and a sociodemographic questionnaire were used.A chi-squared test was used to assess the differences in prevalence between ***:There was no statistically significant difference between the groups in the prevalence of PFDs or quality of *** this study,the prevalence of PFDs was higher than that reported *** institutionalized women,a higher prevalence of PFDs and impaired quality of life were expected,although not ***:There was a higher prevalence of pelvic disorders and impaired quality of life due to these disorders in elderly women.

Exploring MIA-QSARs for farnesyltransferase inhibitory effect of antimalarial compounds refined by docking simulations

Two series of farnesyltransferase (FTase) inhibitors were grouped and their antimalarial activi-ties modeled by means of multivariate image analysis applied to quantitative structure-activity relationship (MIA-QSAR). A reliable model was achieved, with r2 for calibration, external prediction and leave-one-out cross-validation of 0.96, 0.87 and 0.83, respectively. Therefore, biological activities of congeners can be estimated using the QSAR model. The bioactivities of new compounds based on the miscellany of substructures of the two classes of FTase inhibitors were predicted using the MIA-QSAR model and the most promising ones were submitted to ADME (absorption, distribution, metabolism and excretion) and docking evaluation. Despite the smaller interaction energy of the two most promising, predicted compounds in comparison to the two most active compounds of the data set, one of the proposed structures did not violate any Lipinski’s rule of five. Therefore, it is either a potential drug or may drive synthesis of similar, improved compounds.

Objective: To evaluate the alpha-defensin (α-DF) genes polymorphism in women with vulvovaginal candidiasis and recurrence. Methods: This observational study included clinical vaginal secretion samples collected over four years from 88 women, ranging in age from 18 to 65 years, from medical centers of Sao Paulo and Mogi das cruzes, Brazil. Thirty-six of these women were asymptomatic (control group) and 52 presented clinical condition compatible with vulvovaginitis (38 primary or episodic as non-recurrent forms, and 14 recurrent vulvovaginal candidiasis). A portion of each sample was plated on Sabouraud dextrose agar with chloramphenicol and grown on cHROMagar candida for presumptive characterization. The identification of the species was obtained by sequencing of the ITS1 region of rDNA. α-DF genes were amplified for subsequent evaluation of polymorphisms by endonuclease restriction assay. Results: From 88 samples were isolated 60 candida albicans and 28 non-albicans candida spp. Resistant c. albicans strains and non-albicans candida spp. were more prevalent in recurrence. In all groups, the number of resistant non-albicans candida spp. was most high than susceptible strains. α-DF1, α-DF3 and α-DF1/α-DF3 genotypes were found in 32 (36.4%), 17 (19.3%), 6 (6.8%) vaginal samples, respectively. About 33 samples were not amplified. Recurrence and severe disease were more observed in homozygous population. conclusions: Non-albicans candida spp. and homozygotic α-DF genotipes (α-DF1 and α-DF3) were more related with severe clinical signs and recurrence. Further studies about vulvovaginal candidiasis and α-DF genes are necessary to access the more comprehensive role of defensins in clinical manifestations.

AIM: To investigate the effects of bariatric surgery on metabolic parameters, incretin hormone secretion, and duodenal and ileal mucosal gene ***: Nine patients with type 2 diabetes mellitus(T2DM), chronic serum hyperglycemia for more than2 years, and a body mass index(BMI) of 30-35 kg/m2 underwent metabolic surgery sleeve gastrectomy with transit bipartition between May 2011 and December2011. Blood samples were collected pre and 3, 6 and12 mo postsurgery. Duodenal and ileal mucosa samples were collected pre- and 3 mo postsurgery. Pre- and postoperative blood samples were collected in the fasting state before ingestion of a standard meal(520kcal) and again 30, 60, 90, and 120 min after the mealto determine hemoglobin A1c(HbA1c) levels and the lipid profile, which consisted of triglyceride and total cholesterol levels. Intestinal gene expression of p53 and transforming growth factor(TGF)-β was analyzed using quantitative reverse-transcription PcR. Gastric inhibitory polypeptide(GIP) and glucagon-like peptide-1(GLP-1)were quantified using the enzyme-linked immunoassay method and analyzed pre- and ***'s t test or repeated measurements analysis of variance with Bonferroni corrections were performed as ***: BMI values decreased by 15.7% within the initial 3 mo after surgery(31.29 ± 0.73 vs 26.398± 0.68, P ced improved T2 DM, with 7 patients(78%)achieving complete remission(HbA1c chieving improved diabetes(HbA1ccemic agents). Medicinal properties of Angelica archangelica root extract: cytotoxicity in breast cancer cells and its protective effects against in vivo tumor development

Objective: Although Angelica archangelica is a medicinal and aromatic plant with a long history of use for both medicinal and food purposes, there are no studies regarding the antineoplastic activity of its *** study aimed to evaluate the cytotoxicity and antitumor effects of the crude extract of A. archangelica root(cEAA) on breast ***: The cytotoxicity of cEAA against breast adenocarcinoma cells(4T1 and McF-7) was evaluated by a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay. Morphological and biochemical changes were detected by Hoechst 33342/propidium iodide(PI) and annexin V/PI staining. cytosolic calcium mobilization was evaluated in cells staining with FURA-4 NW. Immunoblotting was used to determine the effect of cEAA on anti-and pro-apoptotic proteins(Bcl-2 and Bax, respectively).The 4 T1 cell-challenged mice were used for in vivo ***: Using ultra-high-performance liquid chromatography–mass spectrometry analysis, angelicin, a constituent of the roots and leaves of A. archangelica, was found to be the major constituent of the cEAA evaluated in this study(73 mg/mL). The cEAA was cytotoxic for both breast cancer cell lines studied but not for human fibroblasts. Treatment of 4 T1 cells with the cEAA increased Bax protein levels accompanied by decreased Bcl-2 expression, in the presence of cleaved caspase-3 and cytosolic calcium mobilization, suggesting mitochondrial involvement in breast cancer cell death induced by the cEAA in this cell line. No changes on the Bcl-2/Bax ratio were observed in cEAA-treated McF7 cells. Gavage administration of the cEAA(500 mg/kg) to 4T1 cell-challenged mice significantly decreased tumor growth when compared with untreated ***: Altogether, our data show the antitumor potential of the cEAA against breast cancer cells in vitro and in vivo. Further research is necessary to better elucidate the pharmacological application of the cEAA in breast cancer therapy.
