Advances in small animal mesentery models for in vivo flow cytometry,dynamic microscopy,and drug screening

Using animal mesentery with intravital optical microscopy is a well-established experimental model for studying blood and lymph microcirculation in vivo. Recent advances in cell biology and optical techniques provide the basis for extending this model for new applications, which should generate significantly improved experimental data. This review summarizes the achievements in this specific area, including in vivo label-free blood and lymph photothermal flow cytometry, super-sensitive fluorescence image cytometry, light scattering and speckle flow cytometry, microvessel dynamic microscopy, infrared (IR) angiography, and high-speed imaging of individual cells in fast flow. The capabilities of these techniques, using the rat mesentery model, were demonstrated in various studies; e.g., real-time quantitative detection of circulating and migrating individual blood and cancer cells, studies on vascular dynamics with a focus on lymphatics under normal conditions and under different interventions (e.g. lasers, drugs, nicotine), assessment of lymphatic disturbances from experimental lymphedema, monitoring cell traffic between blood and lymph systems, and high-speed imaging of cell transient deformability in flow. In particular, the obtained results demonstrated that individual cell transportation in living organisms depends on cell type (e.g., normal blood or leukemic cells), the cell’s functional state (e.g., live, apoptotic, or necrotic), and the functional status of the organism. possible future applications, including in vivo early diagnosis and prevention of disease, monitoring immune response and apoptosis, chemo- and radio-sensitivity tests, and drug screening, are also discussed.

The continuous series of perovskite-related oxides with the general formula GdzSr（Al1-xFex）207 0 〈 x 〈 1 were prepared by conventional ceramic technology in air. The pro- cessing stages of Gd2Sr（Al1-xFex）207 0 〈 x 〈 1 phases were investigated by means of X-ray powder diffraction （XRD） and scanning electron microscopy （SEM） with the element phase analysis. The three main stages of GdeSr（All_~Fex）aO7 for- marion are pointed out. The analysis of the mutual influence of intermediate compounds on the synthesis of the target solid solutions of complex composition, in particular for Gd2Sr（All-xFex）207 was carried out. It is determined that the closeness of the reactive mixture composition to the composition of individual compounds Gd2SrA1207 or GdaSrFeaO7 is of importance for the realization of a partic- ular way of GdESr（Al1-xFex）207 solid solutions formation. It is shown that at x ,= 0.2 the reversing in scheme of Gd2Sr（Al1-Fex）207 series formation is observed. The formation of the Gd2Sr（Al1-xFex）207 continuous series is indicated by the monotonic dependence of molar unit cell volume on the iron content x.

NdCoO_(3)nanocrystals formed via glycine-nitrate combustion method followed by heat treatment has been systematically *** of NdCoO_(3)nanocrystals with minimal size of 7-10 nm from X-ray amorphous combustion products has been elucidated to be a very rapid process,occurring at the temperature of 550-600℃for5-30 *** comparison of the minimum sizes of NdCoO_(3)crystallites obtained from the offered empirical relation dmin=lunit cell·N(where N is 7-12 and‘unit cellis elementary cell parameter)and the data determined on the basis of X-ray diffraction(XRD)and transmission electron microscopy(TEM)showed good *** existence of special nanoporous microstructure and spatial limitations prevent NdCoO_(3)particle *** kinetic equation based on Avrami-Erofeev nucleation model was offered to be correlated well with experimental data of fractional conversion(a)versus isothermal time(s).The apparent activation energy(Ea=(338±32)kJ)of formation of NdCoO_(3)nanocrystals from X-ray amorphous combustion products obtained in excess of oxidant followed by heat treatment at 550-600℃was determined.

Selection of shaped charges parameters for producing aluminum particles with velocities in the range of 2.5-16 km/s

Testing rocket and space technology objects in ground conditions for resistance to the impact of meteoroids and fragments of space debris can be carried out using shaped charges. To substantiate the design parameters of shaped charges that ensure the formation of aluminum particles in a wide velocity range(from 2.5 to 16 km/s), numerical modeling of the formation process was carried out within the framework of a two-dimensional axisymmetric problem of continuum mechanics using three different computing codes to increase the reliability of the results. The calculations consider shaped charges with a diameter of 20-100 mm with aluminum liners of various shapes. It is shown that the formation of particles with velocities close to the lower limit of the considered range is ensured by gently sloping segmental liners of degressive thickness. To form higher-velocity particles with velocities over 5 km/s, it is proposed to use combined liners, the jet-forming part of which has the shape of a hemisphere of constant thickness or the shape of a semi-ellipsoid or semi-superellipsoid of rotation of degressive thickness.

Jasmonic Acid Oxidase 2 Hydroxylates Jasmonic Acid and Represses Basal Defense and Resistance Responses against Botrytis cinerea Infection

Jasmonates （JAs） orchestrate immune responses upon wound/herbivore injury or infection by necrotro- phic pathogens. Elucidation of catabolic routes has revealed new complexity in jasmonate metabolism. Two integrated pathways attenuate signaling by turning over the active hormone jasmonoyl-isoleucine （JA-Ile） through w-oxidation or deconjugation, and define an indirect route forming the derivative 12OH-JA. Here, we provide evidence for a second 12OH-JA formation pathway by direct jasmonic acid （JA） oxidation. Three jasmonic acid oxidases （JAOs） of the 2-oxoglutarate dioxygenase family catalyze spe- cific oxidation of JA to 12OH-JA, and their genes are induced by wounding or infection by the fungus Botrytis cinerea. JA02 exhibits the highest basal expression, and its deficiency in jao2 mutants strongly enhanced antifungal resistance. The resistance phenotype resulted from constitutive expression of antimi- crobial markers rather than from their higher induction in infected jao2 plants and could be reversed by ectopic expression of any of the three JAOs injao2. Elevated defense injao2 was dependent on the activity of JASMONATE RESpONSE 1 （JAR1） and CORONATINE-INSENSITIVE 1 （COI1） but was not correlated with erihanced JA-Ile accumulation. Instead, jao2 mutant lines displayed altered accumulation of several JA species in healthy and challenged plants, suggesting elevated metabolic flux through JA-Ile. Collectively, these data identify the missing enzymes hydroxylating JA and uncover an important metabolic diversion mechanism for repressing basal JA defense responses.

Removal of perfluorinated surfactants from wastewater by adsorption and ion exchange--Influence of material properties,sorption mechanism and modeling

perfluorooctane sulfonate（pFOS） has attracted increasing concern in recent years due to its world-wide distribution, persistence, bioaccumulation and potential toxicity. The influence of sorbent properties on the adsorptive elimination of pFOS from wastewater by activated carbons, polymer adsorbents and anion exchange resins was investigated with regard to their isotherms and kinetics. The batch and column tests were combined with physicochemical characterization methods, e.g., N2 physisorption, mercury porosimetry, infrared spectroscopy, differential scanning calorimetry, titrations, as well as modeling. Sorption kinetics was successfully modelled applying the linear driving force（LDF） approach for surface diffusion after introducing a load dependency of the mass transfer coefficient β*** big difference in the initial mass transfer coefficient βs,0, when non-functionalized adsorbents and ion-exchange resins are compared, suggests that the presence of functional groups impedes the intraparticle mass transport. The more functional groups a resin possesses and the longer the alkyl moieties are the bigger is the decrease in sorption *** the selectivity for pFOS sorption is increasing when the character of the functional groups becomes more hydrophobic. Accordingly, ion exchange and hydrophobic interaction were found to be involved in the sorption processes on resins, while pFOS is only physisorptively bound to activated carbons and polymer adsorbents. In agreement with the different adsorption mechanisms, resins possess higher total sorption capacities than adsorbents. Hence, the latter ones are rendered more effective in pFOS elimination at concentrations in the low μg/L range, due to a less pronounced convex curvature of the sorption isotherm in this concentration range.

Morphology of crater and caldera lakes in the Far Eastern region of Russia and the features of their development
