Cytokinin levels and signaling respond to wounding and the perception of herbivore elicitors in Nicotiana attenuata

Nearly half a century ago insect herbivores were found to induce the formation of green islands by manipulating cytokinin （CK） levels. However, the response of the CK pathway to attack by chewing insect herbivores remains unclear. Here, we characterize the CK pathway of Nicotiana attenuata （Torr. ex S. Wats.） and its response to wounding and perception of herbivoreassociated molecular patterns （HAMPs）. We identified 44 genes involved in CK biosynthesis, inactivation, degradation, and signaling. Leaf wounding rapidly induced transcriptional changes in multiple genes throughout the pathway, as well as in the levels of CKs, including isopentenyladenosine and cis-zeatin riboside; perception of HAMPs present in the oral secretions （OS） of the specialist herbivore Manduca sexta amplified these responses. The jasmonate pathway, which triggers many herbivore-induced processes, was not required for these HAMP-triggered changes, but rather suppressed the CK responses. Interestingly CK pathway changes were observed also in systemic leaves in response to wounding and OS application indicating a role of CKs in mediating long distance systemic processes in response to herbivory. Since wounding and grasshopper OS elicited similar accumulations of CKs in Arabidopsis thaliana L., we propose that CKs are integral components of wounding and HAMP-triggered responses in many plant species.

Impaired contractility and remodeling of the upper gastrointestinal tract in diabetes mellitus type-1

AIM: To investigate that both the neuronal function of the contractile system and structural apparatus of the gastrointestinal tract are affected in patients with longstanding diabetes and auto mic neuropathy. METHODS: The evoked esophageal and duodenal contractile activity to standardized bag distension was assessed using a specialized ultrasound-based probe. Twelve type-1 diabetic patients with autonomic neuropathy and severe gastrointestinal symptoms and 12 healthy controls were studied. The geometry and biomechanical parameters (strain, tension/stress, and stiffness) were assessed. RESULTS: The diabetic patients had increased frequency of distension-induced contractions (6.0 ± 0.6 vs 3.3 ± 0.5, P 0.14). CONCLUSION: The impaired contractile activity with an imbalance in the distension-induced contractions likely reflects neuronal abnormalities due to autonomic neuropathy. However, structural changes and remodeling of the gastrointestinal tract are also evident and may add to the neuronal changes. This may contribute to the pathophysiology of diabetic gut dysfunction and impact on future management of diabetic patients with gastrointestinal symptoms.

Despite multiple theories on the pathogenesis of pain in chronic pancreatitis,no uniform and consistently successful treatment strategy exists and abdominal pain still remains the dominating symptom for most patients and a major challenge for *** theories focussed on a mechanical cause of pain related to anatomical changes and evidence of increased ductal and interstitial *** observations form the basis for surgical and endoscopic drainage procedures,but the outcome is variable and often *** underscores the fact that other factors must contribute to pathogenesis of pain,and has shifted the focus towards a more complex neurobiological understanding of pain *** other explanations for pain,experimental and human studies have provided evidence that pain perception at the peripheral level and central pain processing of the nociceptive information is altered in patients with chronic pancreatitis,and resembles that seen in neuropathic and chronic pain ***,pain due to e.g.,complications to the disease and adverse effects to treatment must not be overlooked as an additional source of *** review outlines the current theories on pain generation in chronic pancreatitis which is crucial in order to understand the complexity and limitations of current therapeutic ***,it may also serve as an inspiration for further research and development of methods that can evaluate the relative contribution and interplay of different pain mechanisms in the individual patients,before they are subjected to more or less empirical treatment.

Experience after 100 patients treated with cytoreductive surgery and hyperthermic intraperitoneal chemotherapy

AIM:To investigate perioperative patient morbidity/mortality and outcome after cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC).METHODS:Of 150 patients 100 were treated with cytoreductive surgery and HIPEC and retrospectively *** and postoperative follow-up data were *** mass index(BMI),age and peritoneal carcinomatosis index(PCI) were chosen as selection criteria with regard to tumor-free survival and perioperative morbidity for this multimodal ***:CRS with HIPEC was successfully performed in 100 out of 150 *** patients were excluded because of intraoperative *** PCI was 17(1-39).In 89% a radical resection(CC0/CC1) was *** patient died postoperatively due to multiorgan *** PCI,age nor BMI was a risk factor for postoperative complications/outcome according to the DINDO *** 9% Re-CRS with HIPEC was performed during the follow-up ***:Patient selection remains the most important *** PCI,age nor BMI alone should be an exclusion criterion for this multimodal therapy.

AIM To study mucosal addressin cellular adhesion molecule-1(MAd CAM-1) and vascular endothelial growth factor(VEGF)-targeted contrast enhanced ultrasound(CEUS) for the assessment of murine colitis and carcinogenesis. METHODS C57BL/6 mice were challenged with 3% dextran sodium-sulfate(DSS) for three, six or nine days to study the development of acute colitis. Ultrasound was performed with and without the addition of unspecific contrast agents. MAd CAM-1-targeted contrast agent was used to detect and quantify MAd CAM-1 expression. Inflammatory driven colorectal azoxymethane(AOM)/DSS-induced carcinogenesis was examined on day 42 and 84 using VEGF-targeted contrast agent. Highly specific tissue echogenicity was quantified using specialized software. Sonographic findings were correlated to tissue staining, western blot analysis and immunohistochemistry to quantify the degree of inflammation and stage of carcinogenesis. RESULTS Native ultrasound detected increased general bowel wall thickening that correlated with more progressed and more severe DSS-colitis(healthy mice: 0.3 mm ± 0.03 vs six days DSS: 0.5 mm ± 0.2 vs nine days DSS: 0.6 mm ± 0.2, P < 0.05). Moreover, these sonographic findings correlated well with clinical parameters such as weight loss(r2 = 0.74) and histological damage(r2 = 0.86)(P < 0.01). In acute DSS-induced murine colitis, CEUS targeted against MAd CAM-1 detected and differentiated stages of mild, moderate and severe colitis via calculation of mean pixel contrast intensity in decibel(9.6 d B ± 1.6 vs 12.9 d B ± 1.4 vs 18 d B ± 3.33, P < 0.05). Employing the AOM/DSSinduced carcinogenesis model, tumor development was monitored by CEUS targeted against VEGF and detected a significantly increased echogenicity in tumors as compared to adjacent healthy mucosa(healthy mucosa, 1.6 d B ± 1.4 vs 42 d, 18.2 d B ± 3.3 vs 84 d, 18.6 d B ± 4.9, P < 0.01). Feather Vibration as a Stimulus for Sensing Incipient Separation in Falcon Diving Flight

Based on our preceding studies on the aerodynamics of a falcoperegrinus in diving flight along a vertical dam it is known that even when the body shape of the bird is rather streamlined in V-shape some feathers tips may elevate in certain regions of the body. These regions were identified in wind tunnel tests for typical diving flight conditions as regions of locally separated flow. A life-size model in V-shape of a falcoperegrinus with artificial feathers fixed along the body was studied in a wind tunnel to focus on the fluid-structure interaction of feathers located in this sector. The distal ends of the feathers show flow-induced vibrations at typical flight conditions which grow linear in amplitude with increasing angle of incidence until incipient separation. In light of the proven existence of vibration-sensitive mechanoreceptors in the follicles of secondary feathers in birds it is hypothesized that this linear amplitude response offers the bird to sense the angle of incidence during the diving flight using the vibration magnitude as sensory stimulus. Thus the bird in streamlined shape has still a good measure to control its attitude to be in the narrow window of safe angle of incidence. This might have implications also for other birds or technical applications of airfoil sensors regarding incipient separation detection.
