Validity and reliability of the Polish version of the Michigan Neuropathy Screening Instrument

BACKGROUND Diabetic sensorimotor polyneuropathy is an important risk factor for foot ulceration and ***,patients with diabetes should be screened for this disorder according to local *** obstacle to the diagnosis of this disease may be the lack of unified diagnostic criteria due to the lack of properly validated scales used for *** To validate both sections(A and B)of the Michigan Neuropathy Screening Instrument(MNSI)in Polish(PL)patients with *** A cross-sectional study using a test(A1,B1)and re-test(A2,B2)formula was performed in 80 patients with *** gold standard used for neuropathy detection was a nerve conduction study(NCS)which was performed in all *** of the MNSI-PL was assessed using the Cronbach’s alpha,Kuder-Richardson formula 20(KR-20),split-half reliability,the Gottman split-half tests,and correlation between first and second half was *** was assessed using an intraclass correlation coefficient(ICC).For external validation,we used simple linear correlation,binomial regression,and agreement between two different tools using a Bland-Altman plot *** The scale was internally consistent(Cronbach’s alpha for the full scale:0.81 for A and 0.87 for B).MNSI-PL scores in test/retest showed high stability(ICC=0.73 for A and ICC=0.97 for B).The statistically important correlations between MNSI-PL and NCS were found for B1,B2,and A1(P<0.005).The cut-off points of≥3 for section A(sensitivity of 90%-100%;specificity of 33%-40%)and≥2 for section B(sensitivity of 81%-84%;specificity of 60%-70%)were obtained during neuropathy *** The MNSI-PL is a reliable and valid instrument in screening for diabetic neuropathy.

A dopamine and noradrenaline reuptake inhibitor （bupropion） does not alter exercise performance of bank voles

Physical performance is determined both by biophysical and physiological limitations and behav- ioral characteristic, specifically motivation. We applied an experimental evolution approach com- bined with pharmacological manipulation to test the hypothesis that evolution of increased aerobic exercise performance can be triggered by evolution of motivation to undertake physical activity. We used a unique model system： bank voles from A lines, selected for high swim-induced aerobic metabolism （VO2swim）, which achieved a 61% higher mass-adjusted VO2swim than those from un- selected C lines. Because the voles could float on the water surface with only a minimum activity, the maximum rate of metabolism achieved in that test depended not only on their aerobic capacity, but also on motivation to undertake intensive activity. Therefore, we hypothesized that signaling of neurotransmitters putatively involved in regulating physical activity （dopamine and noradrenaline） had changed in response to selection. We measured VO2swim after intraperitoneal injections of sa- line or the norepinephrine and dopamine reuptake inhibitor bupropion （20 mg/kg or 30 mg/kg）. Additionally, we measured forced-exercise VO2 （VO2max）. In C lines, VO2swim （mass-adjusted mean ± standard error （SE）： 4.0 ± 0.1 mLO2/min） was lower than VO2max （5.0 ± 0.1 mLO2/min）, but in A lines VO2swim （6.0 ± 0.1 mLO2/min） was as high as VO2max （6.0 ± 0.1 mLO2/min）. Thus, the selection effectively changed both the physiological-physical performance limit and mechanisms responsible for the willingness to undertake vigorous locomotor activity. Surprisingly, the drug had no effect on the achieved level of VO2swim. Thus, the results did not allow firm conclusions concerning involvement of these neurotransmitters in evolution of increased aerobic exercise performance in the experimental evolution model system.

The paper presents a comprehensive, newly developed software – poROSE(poROus materials examination SoftwarE) for the qualitative and quantitative assessment of porous materials and analysis methodologies developed by the authors as a solution for emerging challenges. A low porosity rock sample was analyzed and thanks to the developed and implemented methodologies in poROSE software, the main geometrical properties were calculated. A tool was also used in preprocessing part of the computational analysis to prepare a geometrical representation of the porous material. The basic functions as elimination of blind pores in the geometrical model were completed and the geometrical model was exported for CFD software. As a result, it was possible to carry out calculations of the basic properties of the analyzed porous material sample. The developed tool allows to carry out quantitative and qualitative analysis to determine the most important properties characterized porous materials. In presented tool the input data can be images from X-ray computed tomography(CT), scanning electron microscope(SEM) or focused ion beam with scanning electron microscope(FIB-SEM) in grey level. A geometric model developed in the proper format can be used as an input to modeling mass, momentum and heat transfer, as well as, in strength or thermo-strength analysis of any porous materials. In this example, thermal analysis was carried out on the skeleton of rock sample. Moreover, thermal conductivity was estimated using empirical equations.

The Application of Cellulose Nanocrystals Modified with Succinic Anhydride under the Microwave Irradiation for Preparation of Polylactic Acid Nanocomposites

The aim of this work was to use cellulose nanocrystals that were obtained by hydrolysis in phosphoric acid solution and further modified with succinic anhydride in the microwave field for PLA reinforcement.A series of allbionanocomposites containing unmodified and surface modified cellulose nanocrystals with CNC content in the range of 1–3%_(w.t.) were obtained by melt blending and tested by XRD,SEM,DSC and DMA to investigate the effect of surface esterification of CNCs on the structure,morphology,dynamic mechanical properties of bionanocomposites,as well as phase transitions of PLA in the presence of cellulosic *** investigations showed the highest increase of storage modulus by ca.7%(335 MPa at 25℃)in the glassy state of PLA for 2%_(w.t.)of unmodified ***,addition of 2%_(w.t.)of succinylated CNCs caused the highest increase of the onset of glass transition temperature(by 6.2℃)thus widening the temperature range of biocomposite *** increase of glass transition temperature indicates the strongest interfacial interactions due to improved miscibility of surface modified nanocrystals and thus good dispersion of additive in PLA matrix providing high interface.

The effect of monoamines reuptake inhibitors on aerobic exercise performance in bank voles from a selection experiment

Exercise performance depends on both physiological abilities (e.g., muscle strength) and behavioral characteristics (e.g., motivati on). We tested the hypothesis that evoluti on of in creased aerobic exercise performanee can be facilitated by evolution of neuropsychological mechanisms responsible for motivation to undertake physical activity. We used a unique model system: lines of bank voles Myodes glareolus selected for high swim-induced aerobic metabolism ("aerobic" A lines). In gen eration 21, voles from the 4 A lines achieved a 57% higher /y voluntary maximum" swiminduced aerobic metabolism (V02swim) than voles from 4 unselected,"control" C lines. In C lines, V02swim was 9% lower than the maximum forced-exercise aerobic metabolism (VO2run;P= 0.007), while in A lines it was even higher than VO2run, although not significantly (4%, P=0.15). Thus, we hypothesized that selection changed both the aerobic capacity and the neuronal mechanisms behi nd motivation to un dertake activity. We investigated the influe nee of reuptake in hibitors of dopamine (DARI), serotonin (SSRI), and norepinephrine (NERI) on VOaSwim. The drugs decreased V02swim both in C and A lines (% decrease compared with saline: DARI 8%, P< 0.001;SSRI 6%, P< 0.001;NERI 8%, P< 0.001), but the proportional response differed between selection directions only for NERI (stronger effect in C lines: P= 0.008) and the difference was marginally non-significant for SSRI (P= 0.07) and DARI (P= 0.06). Thus, the results suggest that all the 3 monoamines are involved in signaling pathways controlling the motivation to be active and that norepinephrine could have played a role in the evolution of increased aerobic exercise performance in our animal model.

Genetic Structure of Tetraploid Italian and Westerwolds Ryegrasses （Lolium spp,） as Revealed by Enzyme and ISSR Polymorphism

The genetic variation of Italian ryegrass （Lolium multiflorum Lain.） and westerwolds ryegrass （L. westerwoldicum Breakw.） was investigated in eight tetraploid cultivars originated from Poland. The 337 individual seedlings, cultivated in uniform greenhouse conditions, were tested with horizontal gel electrophoresis according to five enzyme systems： diaforase （DIA）, superoxide dismutase （SOD）, glutamate oxalacetate transaminase （GOT）, peroxidise （PX） and phosphogluco isomerise （PGI）, which were used to array allelic diversity at five polymorphic loci. The estimation of genetic diversity in 64 plants of the same cultivars was studied using inter simple sequence repeats （ISSRs） as molecular marker. The genetic variation described by ISSR suggests that the polymorphism detected appears to be poorly informative at the taxonomic level. For statistical analysis, unweighted pair group method with arithmetic mean （UPGMA） based dendrograms and minimum spanning trees were constructed using Nei＇s distance and Jaccard＇ s similarity coefficient.

Experimental evolution of personality traits: open-field exploration in bank voles from a multidirectional selection experiment

Evolution of complex physiological adaptations could be driven by natural selection acting on behavioral traits. Consequently, animal personality traits and their correlation with physiological traits have become an engaging research area. Here, we applied a unique experimental evolution model lines of bank voles selected for (A) high exercise-induced aerobic metabolism,(H) ability to cope with low-quality herbivorous diet, and (P) intensity of predatory behavior, that is, traits shaping evoluti on ary path and diversity of mammals and asked how the selecti on affected the voles' personality traits, assessed in an open field test. The A- and P-line voles were more active, whereas the H-line voles were less active, compared those from unselected control lines (C). H-line voles moved slower but on more meandering trajectories, which indicated a more thorough explorati on, whereas the A- and P-line voles moved faster and on straighter trajectories. A-line voles showed also an increased escape propensity, whereas P-line voles tended to be bolder. The remarkable correlated responses to the selectio n indicate a comm on gen etic un derlying mecha nism of behavioral and physiological traits, and support the paradigm of evoluti on ary physiology built around the concept of correlated evolution of behavior and physiology.

Small bowel tumors detected and missed during capsule endoscopy: Single center experience

AIM:To characterize small bowel(SB)tumors detected by capsule endoscopy(CE),and identify missed ***:The study included 145 consecutive patients in whom 150 CEs were *** CE,the medical records of the study population were *** of double-or single-balloon enteroscopy performed after CE and the results of surgery in all patients operated on were *** patients were contacted through telephone interviews or postal *** addition,the national cancer registry and the polish clinical gastrointestinal stromal tumor(GIST)Registry were searched to identify missed ***:Indications for CE included overt and occult obscure gastrointestinal bleeding(n=81,53.7%),anemia(n=19,12.7%),malabsorption(n=18,12%),abnormal CB follow through(n=9,6%),abdominal pain(n=7,5%),celiac disease(n=5,3%),neuroendocrine tumor(n=3,2%),Crohn’s disease(n=2,<2%),Peutz-Jeghers syndrome(n=2,<2%),other polyposes(n=2,<2%),and diarrhea(n=2,<2%).The capsule reached the colon in 115(76.6%)*** 150 investigations,CE identified 15SB tumors(10%),14 of which were operated on or treated *** included metastatic melanoma(n=1),adenocarcinoma(n=2),and GIST(n=3).Benign neoplasms included dysplastic Peutz-Jeghers polyps(n=4).Non-neoplastic masses included venous malformation(n=1),inflammatory tumors(n=2),and a mass of unknown histology(n=1).During the follow-up period,three additional SB tumors were found(2 GISTs and one mesenteric tumor of undefined nature).The National Cancer Registry and Polish Clinical GIST Registry revealed no additional SB neoplasms in the post-examination period(follow-up:range 4.2-102.5 mo,median 39 mo).The sensitivity of CE for tumor detection was 83.3%,and the negative predictive value was 97.6%.The specificity and positive predictive value were both 100%.CONCLUSION:Neoplasms may be missed by CE,especially in the proximal *** overt obscure gastrointestinal bleeding,complementary endoscopic and/or rad

Virulence factors of Enterococcus strains isolated from patients with inflammatory bowel disease

AIM: To determine the features of Enterococcus that contribute to the development and maintenance of the inflammatory process in patients with inflammatory bowel disease (IBD). METHODS: Multiplex polymerase chain reaction (PCR) was applied to assess the presence of genes that encode virulence factors [surface aggregating protein (asa1), gelatinase (gelE), cytolysin (cylA), extracellular surface protein (esp) and hyaluronidase (hyl)] in the genomic DNA of 28 strains of Enterococcus isolated from the intestinal tissues of children with IBD (n =16) and of children without IBD (controls; n = 12). Additionally, strains with confirmed presence of the gelE gene were tested by PCR for the presence of quorum sensing genes (fsrA, fsrB, fsrC) that control the gelatinase production. Gelatinase activity was tested on agar plates containing 1.6% gelatin. We also analysed the ability of Enterococcus strains to release and decompose hydrogen peroxide (using Analytical Merckoquant peroxide test strips) and tested their ability to adhere to Caco-2 human gut epithelium cells and form biofilms in vitro. RESULTS: A comparison of the genomes of Enterococcus strains isolated from the inflamed mucosa of patients with IBD with those of the control group showed statistically significant differences in the frequency of theasa1 gene and thegelE gene. Furthermore, the cumulative occurrence of different virulence genes in the genome of a single strain ofEnterococcus isolated from the IBD patient group is greater than in a strain from the control group, although no significant difference was found. Statistically significant differences in the decomposition of hydrogen peroxide and adherence to the Caco-2 epithelial cell line between the strains from the patient group and control group were demonstrated. The results also showed that profuse biofilm production was more frequent amongEnterococcus strains isolated from children with IBD than in control strains. CONCLUSION: Enterococcus strains that a
