A High-Resolution Modeling Strategy to Assess Impacts of Climate Change for Mesoamerica and the Caribbean

Mesoamerica and the Caribbean are low-latitude regions at risk for the effects of climate change. Global climate models provide large-scale assessment of climate drivers, but, at a horizontal resolution of 100 km, cannot resolve the effects of topography and land use as they impact the local temperature and precipitation that are keys to climate impacts. We developed a robust dynamical downscaling strategy that used the WRF regional climate model to downscale at 4 - 12 km resolution GCM results. Model verification demonstrates the need for such resolution of topography in order to properly simulate temperatures. Precipitation is more difficult to evaluate, being highly variable in time and space. Overall, a 36 km resolution is inadequate;12 km appears reasonable, especially in regions of low topography, but the 4 km resolution provides the best match with observations. This represents a tradeoff between model resolution and the computational effort needed to make simulations. A key goal is to provide climate change specialists in each country with the information they need to evaluate possible future climate change impacts.

Understanding the initial use stage of cement mortar lining in drinking water distribution systems:Silicon and aluminum as stability indicators

The cement mortar lining(CML)of commonly used ductile iron pipes can severely deteriorate the drinking water quality at the initial stage of use,but the behavioral characteristics of different elements release from the CML in this stage is still unclear.In this study,dynamic immersion experiments with new cement mortar lined ductile iron pipe reactors were conducted under different feed water hardness and alkalinity conditions.The results showed that the release of alkaline substances from the CML at the initial stage of use could strongly influence the pH of water,which consequently greatly impacted the release/precipitation behaviors of calcium,aluminum and silicon.The pH and aluminum concentration of the effluent water could reach 11.5 and 700μg/L within 24 hr of hydraulic retention time,respectively,under conditions of relatively lower hardness and alkalinity.Due to the pH elevation,calcium carbonate precipitation could occur even at much lower feed water alkalinity.Whereas the aluminum and silicon could keep release from the CML in soluble form at different hardness and alkalinity levels,and their release rate depended on the amount of calcium carbonate precipitation.Thus,aluminum and silicon were more suitable as indicators of the corrosion intensity at the initial stage of CML use rather than the traditional calcium carbonate precipitation potential.Appropriate feed water hardness and alkalinity levels for mitigating the initial intense corrosion of CML were proposed:hardness>40 mg/L(CaCO_(3)),alkalinity>100 mg/L(CaCO_(3)).

Chronic effects of stretching on range of motion with consideration of potential moderating variables:A systematic review with meta-analysis

Background:It is well known that stretch training can induce prolonged increases in joint range of motion(ROM).However,to date more information is needed regarding which training variables might have greater influence on improvements in flexibility.Thus,the purpose of this metaanalysis was to investigate the effects of stretch training on ROM in healthy participants by considering potential moderating variables,such as stretching technique,intensity,duration,frequency,and muscles stretched,as well as sex-specific,age-specific,and/or trained state-specific adaptations to stretch training.Methods:We searched through PubMed,Scopus,Web of Science,and SportDiscus to find eligible studies and,finally,assessed the results from 77 studies and 186 effect sizes by applying a random-effect meta-analysis.Moreover,by applying a mixed-effect model,we performed the respective subgroup analyses.To find potential relationships between stretch duration or age and effect sizes,we performed a meta-regression.Results:We found a significant overall effect,indicating that stretch training can increase ROM with a moderate effect compared to the controls(effect size=-1.002;Z=-12.074;95%confidence interval:-1.165 to-0.840;p<0.001;I^(2)=74.97).Subgroup analysis showed a significant difference between the stretching techniques(p=0.01)indicating that proprioceptive neuromuscular facilitation and static stretching produced greater ROM than did ballistic/dynamic stretching.Moreover,there was a significant effect between the sexes(p=0.04),indicating that females showed higher gains in ROM compared to males.However,further moderating analysis showed no significant relation or difference.Conclusion:When the goal is to maximize ROM in the long term,proprioceptive neuromuscular facilitation or static stretching,rather than ballistic/dynamic stretching,should be applied.Something to consider in future research as well as sports practice is that neither volume,intensity,nor frequency of stretching were found to play a significant role in ROM yields.

Utilization of RNA sequencing to investigate olfactory dysfunction in chronic rhinosinusitis without nasal polyps:A pilot study

Objectives:Prior research on olfactory dysfunction in chronic rhinosinusitis(CRS)has focused on patients with polyps and suggests that direct inflammation of the olfactory cleft mucosa plays a contributory role.The purpose of this study was to evaluate gene expression in superior turbinate mucosal specimens,comparing normosmic and dysosmic CRS patients without polyps(CRSsNP).Methods:Tissue samples were obtained from the superior turbinates of patients with CRSsNP at the time of endoscopic sinus surgery.Samples subsequently underwent RNA sequencing and functional analysis to investigate biological pathways associated with differentially expressed genes between dysosmic(n=7)and normosmic(n=4)patients.Results:Differential gene expression analysis comparing dysosmic and normosmic CRSsNP patients showed upregulation of 563 genes and downregulation of 327 genes.Using stringent criteria for multiple comparisons,one upregulated gene(Immediate Early Response 3[IER3])had an false discovery rate(FDR)correction adjustedP value considered statistically significant(P<0.001,fold change 2.69).Reactome functional analysis revealed eight biological pathways significantly different between dysosmic and normosmic patients(P<0.05,FDR correction)including IL-4 and IL-13 signaling,IL-10 signaling,and rhodopsin-like receptors.Conclusions:RNA sequencing of the superior turbinates in patients with CRSsNP can provide valuable information regarding biological pathways and genes involved in olfactory dysfunction.This study supports literature suggesting that Type 2 inflammation may play a role in olfactory dysfunction in at least some patients with CRSsNP.This study also prompts questions regarding the role of IL-10,rhodopsin-like receptors,andIER3 in the pathogenesis of olfactory dysfunction.

Comparison of trigeminal lateralization with differing stimulants

Trigeminal-specific stimulants have been shown to activate different receptors preferentially and this likely accounts for variation in sensory perception.It is unclear whether trigeminal sensitivity is similar across different transient receptor potential(TRP)receptors or if dysfunction of different receptors results in differing patient symptoms.Therefore,a prospective cohort study was conducted,consisting of trigeminal lateralization testing with three different stimulants(eucalyptol,isothiocyanate,acetic acid),olfaction testing with Sniffin'Sticks,and measurement of various patient‐reported outcome measures(PROMs).A total of 50 participants were enrolled across the olfactory spectrum.Mean TDI score was 27.1±8.3(range 7.0–39.5)with 38%normosmic and 62%dysosmic.Mean trigeminal lateralization scores out of 20 in the overall cohort were 16.18(2.78)for eucalyptol,14.94(3.49)for mustard oil,and 15.28(3.68)for vinegar.Eucalyptol showed a significant correlation with threshold scores of Sniffin'Sticks.A significant correlation was found between acetic acid and various PROMs.None of the lateralization scores of the trigeminal stimulants correlated to each other significantly and there was no correlation to age.The lack of correlation suggests that the measured sensitivity of one type of TRP receptor may not translate to similar sensitivity of the other receptors.Additional investigations with TRPV1 and TRPA1 agonists are needed to corroborate our findings.

Colorectal cancer（CRC）, a widespread malignancy, is closely associated with tumor microenvironmental hydrogen peroxide（H2O2） levels. Some clinical trials targeting H2O2for cancer treatment have revealed its paradoxical role as a promoter of cancer progression. Investigating the dynamics of cancer cell H2O2eustress at the single–cell level is crucial. In this study, non–contact hopping probe mode scanning ion conductance microscopy（HPICM） with high-sensitive Pt–functionalized nanoelectrodes was employed to measure dynamic extracellular to intracellular H2O2gradients in individual colorectal cancer Caco–2cells. We explored the relationship between cellular mechanical properties and H2O2gradients.Exposure to 0.1 or 1 mmol/L H2O2eustress increased the extracellular to intracellular H2O2gradient from 0.3 to 1.91 or 3.04, respectively. Notably, cellular F–actin–dependent stiffness increased at 0.1 mmol/L but decreased at 1 mmol/L H2O2eustress. This H2O2–induced stiffness modulated AKT activation positively and glutathione peroxidase 2（GPX2） expression negatively. Our findings unveil the failure of some H2O2-targeted therapies due to their ineffectiveness in generating H2O2, which instead acts eustress to promote cancer cell survival. This research also reveals the complex interplay between physical properties and biochemical signaling in cancer cells' antioxidant defense, illuminating the exploitation of H2O2eustress for survival at the single–cell level. Inhibiting GPX and/or catalase（CAT） enhances the cytotoxic activity of H2O2eustress against CRC cells, which holds significant promise for developing innovative therapies targeting cancer and other H2O2-related inflammatory diseases.

An examination of predictive markers for successful sperm extraction procedures:a linear model and systematic review

The authors performed a comprehensive review of current literature to create a model comparing commonly evaluated variables in male factor infertility,for example,follicle-stimulating hormone(FSH),testicular volume(TV),and testosterone(T),to better predict sperm retrieval rate(SRR).Twenty-nine studies were included,9 with data on conventional testicular sperm extraction(cTESE)for a total of 1227 patients and 20 studies including data on microdissection testicular sperm extraction(mTESE)for a total of 4760 patients.A weighted-means value of SRR,FSH,T,and TV was created,and a weighted linear regression was then used to describe associations among SRR,type of procedure,FSH,T,and TV.In this study,weighted-means values demonstrated mTESE to be superior to cTESE with an SRR of 51.9%vs 40.1%.Multiple weighted linear regressions were created to describe associations among SRR,procedure type,FSH,T,and TV.The models showed that for every 1.19 mIU ml^(−1)increase in FSH,there would be a significant decrease in SRR by 1.0%.Seeking to create a more clinically relevant model,FSH values were then divided into normal,moderate elevation,and significant elevation categories(FSH20 mIU ml^(−1),respectively).For an index patient undergoing cTESE,the retrieval rates would be 57.1%,44.3%,and 31.2%for values normal,moderately elevated,and significantly elevated,respectively.In conclusion,in a large meta-analysis,mTESE was shown to be more successful than cTESE for sperm retrievals.FSH has an inverse relationship to SRR in retrieval techniques and can alone be predictive of cTESE SRR.

Research on encoding and decoding of non-binary polar codes over GF(2m)

Binary Polar Codes (BPCs) have advantages of high-efficiency and capacity-achieving but suffer from large latency due to the Successive-Cancellation List (SCL) decoding. Non-Binary Polar Codes (NBPCs) have been investigated to obtain the performance gains and reduce latency under the implementation of parallel architectures for multi-bit decoding. However, most of the existing works only focus on the Reed-Solomon matrix-based NBPCs and the probability domain-based non-binary polar decoding, which lack flexible structure and have a large computation amount in the decoding process, while little attention has been paid to general non-binary kernel-based NBPCs and Log-Likelihood Ratio (LLR) based decoding methods. In this paper, we consider a scheme of NBPCs with a general structure over GF(2m). Specifically, we pursue a detailed Monte-Carlo simulation implementation to determine the construction for proposed NBPCs. For non-binary polar decoding, an SCL decoding based on LLRs is proposed for NBPCs, which can be implemented with non-binary kernels of arbitrary size. Moreover, we propose a Perfect Polarization-Based SCL (PPB-SCL) algorithm based on LLRs to reduce decoding complexity by deriving a new update function of path metric for NBPCs and eliminating the path splitting process at perfect polarized (i.e., highly reliable) positions. Simulation results show that the bit error rate of the proposed NBPCs significantly outperforms that of BPCs. In addition, the proposed PPB-SCL decoding obtains about a 40% complexity reduction of SCL decoding for NBPCs.
