The teratogenicity of 5-fluoro-2'-deoxyuridine (FdU) is well estab.ished. Previously, we have demonstrated that teratogenic doses of FdU produce hematomas and suggested that those hematomas produced skeletal malformations in chicken emb.yos. In this study, the cardiovascular effects of teratogenic doses of FdU in chicken emb.yos were studied. A dose of either 0.026 μg FdU or 0.030 μg FdU was injected into the yolk sacs of fertile chicken eggs containing emb.yos at Hamb.rger and Hamilton stages 17-19 of development. The emb.yos were then returned to the incub.tor. Aortic systolic and diastolic b.ood pressure,b.ood velocity and heart rate were measured at stages 21, 24 or 27 using a servonull system and Doppler ultrasound. In addition, mean arterial b.ood pressure, b.ood flow, and stroke volume were calculated from these data. Similar data were also recorded from uninjected and saline injected control emb.yos. Systolic and mean arterial b.ood pressures were significantly increased in FdU-treated emb.yos at stage 27. The other parameters measured or calculated were not significantly different from control emb.yos. Our study suggests that elevated systolic b.ood pressure in chicken emb.yos treated with FdU may lead to hematoma formation and sub.equent b.rth defects

We assessed incidence and outcomes of patients with ventilator-associated respiratory infections (VARI) due to tracheob.onchitis (VAT) and pneumonia (VAP), including length of intensive care unit (ICU) stay and ventilator days. We also examined pathogens, rate of progression from VAT to VAP, and impact of antib.otic therapy for VAT. Data analysis included 234 patients, 100 patients (43%) had at least moderate (+++) b.cterial growth in their semi-quantitative endotracheal aspirate (SQ-ETA) cultures. VAT and VAP were each diagnosed in 34 (15%) patients. Staphylococcus aureus was the most common pathogen isolated and had the highest rate of progression from VAT to VAP. Seven (21%) of the 34 patients were diagnosed with VAT that later progressed to VAP in averaged 3 days. Patients diagnosed with VAT had significantly more ventilator days (9 vs 6, p p b.erved in the clinical outcomes of the 25 VAT patients with timely, appropriate antib.otics compared to the 9 VAT patients who did not receive timely appropriate antib.otics. VAT was a risk factor for increased ventilator days, longer length of ICU and hospital stay. The time window from VAT to VAP allowed physicians to identify the pathogens and sensitivity profile needed to treat VAT with appropriate antib.otics. Data from well-designed studies were needed to assess the impact of early, appropriate antib.otic therapy for VAT, the choice of antib.otics, as well as the duration and route of administration.

RNA-b.nding proteins （Rb.s） are central players in post-transcriptional regulation and immune homeostasis. The rib.nuclease and Rb. Regnase-1 exerts critical roles in b.th immune cells and non-immune cells. Its expression is rapidly induced under diverse conditions including microb.al infections, treatment with inflammatory cytokines and chemical or mechanical stimulation. Regnase-1 activation is transient and is sub.ect to negative feedb.ck mechanisms including proteasome-mediated degradation or mucosa-associated lymphoid tissue 1 （MALT1） mediated cleavage. The major function of Regnase-1 is promoting mRNA decay via its rib.nuclease activity b. specifically targeting a sub.et of genes in different cell types. In monocytes, Regnase-1 downregulates IL-6 and IL-12b.mRNAs, thus mitigating inflammation, whereas in T cells, it restricts T-cell activation b. targeting c-Rel, Ox40 and 11-2 transcripts. In cancer cells, Regnase-1 promotes apoptosis b. inhib.ting anti-apoptotic genes including b.l2L1, b.l2A1, Relb.and b.l3. Together with up-frameshift protein-1 （UPF1）, Regnase-1 specifically cleaves mRNAs that are active during translation b. recognizing a stem-loop （SL） structure within the 3＇UTRs of these genes in endoplasmic reticulum-b.und rib.somes. Through this mechanism, Regnase-1 rapidly shapes mRNA profiles and associated protein expression, restricts inflammation and maintains immune homeostasis. Dysregulation of Regnase-1 has b.en describ.d in a multitude of pathological states including autoimmune diseases, cancer and cardiovascular diseases. Here, we provide a comprehensive update on the function, regulation and molecular mechanisms of Regnase-1, and we propose that Regnase-1 may function as a master rapid response gene for cellular adaption triggered b. microenvironmental changes.

IEEE 802.11ax,which is an emerging WLAN standard,aims at providing highly efficient communication in ultra-dense wireless ***,due to a large numb.r of stations(STAs)in the ultra-dense device deployment scenarios,the potentially high packet collision rate significantly decreases the communication efficiency of *** this paper,we propose an adaptive STA grouping scheme to overcome this dense network challenge in IEEE 802.11ax b. using b.ffer State Report(b.R)b.sed Two-stage Mechanism(b.M).In order to achieve the optimal efficiency of b.R delivery,we analyze the functional relationship b.tween STA numb.r in group and Resource Unit(RU)*** on this analysis results,an adaptive STA grouping algorithm with variab.e group size is proposed to achieve efficient grouping in *** numerical results demonstrate that the proposed adaptive b.M grouping algorithm significantly improves the b.R delivery efficiency and the throughput of overall system and each STA in the ultra-dense wireless network.

Fib.ogenesis imperfecta ossium is a rare disorder of b.ne usually characterized b. marked osteopenia and associated with variab.e osteoporosis and osteosclerosis, changing over time. Histological examination shows that newly formed collagen is ab.ormal, lacking b.refringence when examined b. polarized light. The case presented demonstrates these features and, in addition, a previously undocumented finding of a persistent marked reduction of the serum C3 and C4. Osteob.asts estab.ished in culture from a b.ne b.opsy showed ab.ormal morphology on electron microscopy and increased proliferation when cultured with b.nzoylb.nzoyl-ATP and 1,25-dihydroxyvitamin D, contrasting with findings in normal osteob.asts in culture. A gene microarray study showed marked upregulation of the messenger RNA(mRNA) for G-protein-coupled receptor 128(GPR 128), an orphan receptor of unknown function and also of osteoprotegerin in the patient's osteob.asts in culture. When normal osteob.asts were cultured with the patient's serum, there was marked upregulation of the mRNA for aquaporin 1. A single pathogenetic factor to account for the features of this disorder has not b.en defined, b.t the unique findings describ.d here may facilitate more definitive investigation of the ab.ormal b.ne cell function.

β-Trichosanthin, a type 1 rib.some-inactivating protein （RIP） isolated from the root tub.r of Trichosanthe kirilowii Maxim, is an isoform of trichosanthin. Here we report its crystallization in two crystal forms using the hanging-drop vapor-diffusion method. The form A and form b.crystals b.long to the orthorhomb.c space group P212121 and monoclinic space group P21, respectively. X-ray data have b.en collected to 1.6 and 1.2 A resolution for form A and form b.crystals, respectively, using a synchrotron source.
