Mechanisms of action of leptin in preventing gastric ulcer

To investigate the effects of leptin （1-20μg/kg） on acidified ethanol （AE）- and indomethacin （Indo）-induced gastric lesions in rats and compare it with ranitidine, lansoprazole, and omeprazole and to determine its mechanisms of ***： Gastric ulcers, which were approximately 1 mm in width, formed in the glandular portion of the gastric mucosa produced by oral administration of either AE or Indo were taken as ulcer index. The inhibitory effect of subcutaneous administration of leptin, two proton pump inhibitors （PPIs） lansoprazole and omeprazole, or H2-receptor antagonist ranitidine 30 min before AE or Indo was evaluated.A radioimmunoassay was used to determine the PGE2 concentration in the homogenate of the glandular portion of the stomach. We performed histological study of the glandular stomach for the evaluation of total, acidic, and sulfated mucus ***： Subcutaneous administration of leptin, two PPI slansoprazole and omeprazole or H2-receptor antagonist ranitidine 30 min before AE or Indo produced a dosedependent and reproducible inhibition of gastric ulcers （GUs）. This inhibition was found to be more potent than other antagonists used. In N^G-nitro L-arginine methyl ester （L-NAME）-pretreated animals, the ulcer prevention ability of leptin in AE-induced ulcer was significantly reduced,compared to rats without L-NAME pretreatment. However,the ulcer prevention ability of leptin was not altered by L-NAME treatment in Indo-induced ulcers. Leptin produced a dose-dependent increase in PGE2 level in the gastric glandular tissues. Leptin also increased mucus ***： The results of the present study show that leptin inhibits GU formation by AE or Indo in a dosedependent and reproducible manner in rats. The results also suggest that leptin prevents ulcer formation by increasing the activities of the cyclo-oxygenase and/or nitric oxide pathways and by increasing mucus secretion.

Multi-length scale modeling of carburization,martensitic microstructure evolution and fatigue properties of steel gears

Multi-length scale modeling is performed to(i)predict the carburized case depth of SAE8620 steel gears by solving the Fick’s second law of diffusion,(ii)model the martensitic microstructure evolution in a grain inside the carburized case as well as to study the effect of stress cycling on retained austenite(RA)and martensite using a 3D phase-field model,(iii)simulate the effect of carburization and different RA contents on macroscale fatigue behavior of SAE8620 steel spur gear using the finite element *** diffusion model predicts that the case depth increases with increasing heat treatment time and *** phase-field simulations show that RA can transform to martensite during fatigue loading,where the extent of the transformation will depend on the type of stresses applied,*** in a high stress regime or low stress regime of fatigue *** transformation of martensite to austenite is also observed in low RA sample under high stress *** macroscale simulations show that the carburized case with high RA gives rise to better fatigue life compared to that with low RA.

The ever-increasing capability of computational methods has resulted in their general acceptance as a key part of the materials design *** this has been achieved using a so-called computational funnel,where increasingly accurate-and expensive–methodologies are used to winnow down a large initial library to a size which can be tackled by *** this paper we present an alternative approach,using a multi-output Gaussian process to fuse the information gained from both experimental and computational methods into a single,dynamically evolving *** challenges with computational funnels,such as mis-ordering methods,and the inclusion of non-informative steps are avoided by learning the relationships between methods on the *** show this approach reduces overall optimisation cost on average by around a factor of three compared to other commonly used approaches,through evaluation on three challenging materials design problems.

The significance of clinical practice guidelines on adult varicocele detection and management

Varicoceles are the most common correctable etiology of male factor infertility. However, the detection and management of varicoceles have not been standardized. This has led to decades of debate regarding the effect of varicocele on male infertility and subsequently whether repair leads to an improved fertility status. The current body of evidence investigating the role of varicocele and varicocelectomy is weak and conflicting. The stance taken by the AUA and ASRM suggests that there is insufficient outcomes data to support evidenced-based guidelines, citing evidence used to provide current recommendations are generally of a low quality level. on the other hand, the EAU Guidelines give a level la of evidence for management of varicoceles that are clinically palpable, associated with subnormal semen analyses and having otherwise unexplained fertility. Besides aiding with clinical varicocele detection and management, clinical practice opinion statements and guidelines aim to direct and strengthen the infrastructure of future studies. We review the current status of opinion statements and guidelines in varicocele and management detection with focus on their application in practice.

Assimilation of GoES-R Geostationary Lightning Mapper Flash Extent Density Data in GSI 3DVar, EnKF, and Hybrid En3DVar for the Analysis and Short-Term Forecast of a Supercell Storm Case

Capabilities to assimilate Geostationary operational Environmental Satellite “R-series ”(GoES-R) Geostationary Lightning Mapper(GLM) flash extent density(FED) data within the operational Gridpoint Statistical Interpolation ensemble Kalman filter(GSI-EnKF) framework were previously developed and tested with a mesoscale convective system(MCS) case. In this study, such capabilities are further developed to assimilate GoES GLM FED data within the GSI ensemble-variational(EnVar) hybrid data assimilation(DA) framework. The results of assimilating the GLM FED data using 3DVar, and pure En3DVar(PEn3DVar, using 100% ensemble covariance and no static covariance) are compared with those of EnKF/DfEnKF for a supercell storm case. The focus of this study is to validate the correctness and evaluate the performance of the new implementation rather than comparing the performance of FED DA among different DA schemes. only the results of 3DVar and pEn3DVar are examined and compared with EnKF/DfEnKF. Assimilation of a single FED observation shows that the magnitude and horizontal extent of the analysis increments from PEn3DVar are generally larger than from EnKF, which is mainly caused by using different localization strategies in EnFK/DfEnKF and PEn3DVar as well as the integration limits of the graupel mass in the observation operator. overall, the forecast performance of PEn3DVar is comparable to EnKF/DfEnKF, suggesting correct implementation.

Bayesian optimization for Field-Scale Geological Carbon Storage

Endovascular treatment of vasculogenic erectile dysfunction

The treatment of erectile dysfunction （ED） has been a fascination involving multiple medical specialities over the past century with urologic, cardiac and surgical experts all contributing knowledge toward this multifactorial disease. With the well-described association between ED and cardiovascular disease, angiography has been utilized to identify vasculogenic impotence. Given the success of endovascular drug-eluting stent （DES） placement for the treatment of coronary artery disease, there has been interest in using this same technology for the treatment of vasculogenic ED. For men with inflow stenosis, DES placement to bypass arterial lesions has recently been reported with a high technical success rate. Comparatively, endovascular embolization as an approach to correct veno-occlusive dysfunction has produced astonishing procedural success rates as well. However, after a thorough literature review, arterial intervention is only recommended for younger patients with isolated vascular injuries, typically from previous traumatic experiences. Short-term functional outcomes are less than optimal with long-term results yet to be determined. In conclusion, the hope for a minimally invasive approach to ED persists but additional investigation is required prior to universal endorsement.
