Virulence Factor Profile of Staphylococcus aureus Isolated from Bovine Milk from Brazil

This study investigates the biofilm formation, presence and distribution of virulence genes and the capacity to induce an inflammatory response in strains of Staphylococcus aureus isolated from milk samples in Bahia, Brazil. A total of 132 samples of raw milk were collected from four dairy farms (designated A to D) located in southwestern Bahia, in the municipality of Vitória da Conquista, from October/2009 to September/2010. After processing of the samples, 94 (71.2%) S. aureus isolates were obtained. These strains were subjected to the antibiogram method MIC (Minimum Inhibitory Concentration). As for the pathogenicity, tests were performedin vitrobiofilm formation induced by glucose. Moreover, we performed PCR for their virulence genes: sea (enterotoxin A), seb (B), sec (C), pvl (Panton-Valentine Leukocidin), clfA (Clumping Factor A) and spa (protein A) and analysis of cytokine induction in the inflammatory response of J774 macrophages by exocellular lipoteichoic acid. No isolates were resistant to oxacillin and vancomycin. In biofilm production, 5.31% (5/94) isolates did not produce biofilm, 5.31% (5/94) of the samples were poor producers, 15.96% (15/94) strains were moderate producers, 18.09% (17/94) were producers and 55.32% (55/94) of isolates were strong biofilm producers. One (1.06%) isolate expressed the seb gene, one (1.06%) sec, 18 (19.2%) cflA and 44 (46.8%) had spa. There was no expression of sea and pvl between isolates analyzed. The analysis of cytokine induction in the inflammatory response did not show statistical difference in the levels of IL-6, TNF-α and IL-10 induction. However, there was statistical difference in IL-1 induction between isolates from different farms. Thus, it appears that the results obtained in this study show significant effects for the region studied, since it is an important dairy region, hence the need for further studies, with the intent of attracting funding that contributes to improving prevention and control in both dai

IMMEDIATE RELIEF OF KNEE PAIN THROUGH THE COMPARISON OF HAND AND EAR ACUPUNCTURE

Objective: The present study aimed to assess the immediate effect of knee pain relief by comparing two techniques, the Koryo Sooji Chim(hand acupuncture) and ear ***: Forty subjects were included in the present study and randomly divided into 4 groups of 10 subjects in each: real hand acupuncture, sham hand acupuncture, real ear acupuncture and sham ear *** patient was evaluated by Visual Analog Scale(VAS) before and after the ***: In group A, subjects who underwent real hand acupuncture showed 50%(5 individuals) improvement in pain and 50%(5 individuals) unchanged; in group B, subjects who underwent sham hand acupuncture showed 100%(10 individuals) unchanged; in group C, subjects who underwent real ear acupuncture showed 90%(9 individuals) improvement in pain and 10%(1 individual) did not change; and in group D, subjects who underwent sham ear acupuncture showed no changes in 100%(10 individuals).Conclusion: From the results, it is clear that the correct application of the stimulus shows better improvement in pain, with the prevalence rates of ear acupuncture that showed better efficacy compared to Korean hand acupuncture.

The state of Pará, located in the Amazon region of Brazil, has observed in recent years an increase in cocoa (Theobroma cacao) cultivation and has become the largest producer in Brazil. Due to its physiological characteristics, cacao is cultivated in native forests understory or under the shade produced by fast-growing native tree species, serving as an important species for restoration of degraded areas. However, mapping and monitoring cocoa plantation using optical sensor images is a challenge given its botanical and arboreal characteristics that can be confused with other native species at various stages of secondary regrowth. Agroforestry systems are important components of sustainable production in the Amazon and our work sought to better describe the evolution of cocoa plantations in terms of their historical expansion, farming properties practices, land use transitions and fire regimes. Our findings to analyze the relationships between cocoa plantations and hotspots, data from the INPE’s reference satellite between the years 2004 to 2020 were used in this study, polygons classified as cocoa areas, generated by the MapCacau research project, were used, in a total of 69,904 hectares distributed throughout the state of Pará. Finally, we used the protected areas’ official limits in the State of Pará to analyze the plantations’ occurrence in regions in discordance with environmental legislation. The data show that cocoa-producing properties are statistically fewer than non-producing properties, as well as having lower deforestation rates. In our study, we observed that 52,778 hectares (88.87%) of the cocoa area planted had already been deforested by the year 2008—the threshold of deforestation defined by Brazil’s Forest Code. It was also possible to verify that approximately 20,900 hectares continue to be mapped as forest by PRODES, despite our field data identifying cocoa plantations shaded by explored forest in these areas. AIM To assess lactase gene(LCT)-13910C>T polymorphisms in Brazilian non-alcoholic fatty liver disease(NAFLD) and nonalcoholic steatohepatitis(NASH) patients in comparison with healthy *** This was a transverse observational clinical study with NAFLD patients who were followed at the Hepatology Outpatient Unit of the Hospital das Clínicas, S?o Paulo, Brazil. The polymorphism of lactase non-persistence/lactase persistence(LCT-13910C>T) was examined by PCR-restriction fragment length polymorphism technique in 102 liver biopsy-proven NAFLD patients(steatosis in 9 and NASH in 93) and compared to those of 501 unrelated healthy volunteers. Anthropometric, clinical, biochemical and liver histology data were analyzed. Continuous variables were compared using the t or Mann-Whitney tests, and categorical data were compared with the Fisher's exact test. Univariate logistic regression and multivariate logistic regression adjusted for gender and age were *** No differences in the LCT-13910 genotype frequencies were noted between the NAFLD patients(66.67% of the patients with steatosis were CC, 33.33% were CT, and none were TT; 55.91% of the patients with NASH were CC, 39.78% were CT, and 4.3% were TT; P = 0.941) and the healthy controls(59.12% were CC, 35.67% were CT, and 5.21% were TT) or between the steatosis and NASH patients. That is, the distribution of the lactase non-persistence/lactase persistence polymorphism(LCT-13910C>T) in the patients with NAFLD was equal to that in the general population. In the NASH patients, the univariate analysis revealed that the lactase nonpersistence(low lactase activity or hypolactasia) phenotype was associated with higher insulin levels(23.47 ± 15.94 μU/m L vs 15.8 ± 8.33 μU/m L, P = 0.027) and a higher frequency of insulin resistance(91.84% vs 72.22%, P = 0.02) compared with the lactase persistence phenotype. Antiasthmatic Effect of Eugenol （4-Allyl-2-Methoxyophenol） Mediated by Both Bronchodilator and Immunomodulatory Properties

Eugenol, an aromatic product, exhibits anti-inflammatory properties, however, little is known about its effect in allergic inflammation mediated by Th2-type cells and cytokines. We examined the pharmacological potential of this compound in a murine model of respiratory allergy to Bt （Blomia tropicalis）. For this, AJ mice were sensitized （100μg/animal s.c.） and challenged （10 μg/animal i.n.） with Bt mite extract. Sensitized animals were treated or not with eugenol （20, 40 or 80 mg/kg） and the following parameters were analyzed： number of total cells in BAL （bronchoalveolar lavage）; EPO （eosinophil peroxidase activity） and histopathological changes in the lungs; serum level of specific IgE, IgG1 and IgG2a and; concentration of Th2 cytokines on BAL and spleen cultures. In addition, the capability ofeugenol in relaxing tracheal smooth muscle was also evaluated. Our results showed that treatment with eugenol significantly reduced the airway inflammation, decreasing the cellular infiltrate, EPO and mucus in the lungs, as well as the production of Th2 cytokines. It was also demonstrated a dilator effect of eugenol observed by the relaxation of normal and hyper-reactivity isolated trachea upon carbachol stimulation. These results suggest that eugenol has potential as an anti-asthmatic drug by both bronchodilator and immunomodulatory properties.
