Biodegradable polydioxanone stents were developed for the treatment of refractory benign esophageal strictures but have been suggested as a new therapeutic option for intestinal strictures.The primary advantage of biodegradable stents over self-expandable metallic stents is that removal is not required.There are,however,few data available on their use in the small or large bowel.We herein describe the case of a 33-yearold patient with long-standing Crohn's disease(CD) who developed a fibrotic stricture of the sigmoid too long to be amenable to balloon dilation.The use of a biodegradable polydioxanone stent was chosen to avoid surgery.Combined endoscopic and fluoroscopic placement of the stent was technically simple,safe and clinically successful,and no recurrence of obstructive symptoms occurred during a 16-mo follow-up.Further studies are needed to evaluate the long-term efficacy and safety of biodegradable stents in the treatment of intestinal strictures,particularly in the context of CD.

"Synthetic"allopolyploids recreated by interspecific hybridization play an important role in providing novel genomic variation for crop improvement.Such synthetic allopolyploids often undergo rapid genomic structural variation(SV).However,how such SV arises,is inherited and fixed,and how it affects important traits,has rarely been comprehensively and quantitively studied in advanced generation synthetic lines.A better understanding of these processes will aid breeders in knowing how to best utilize synthetic allopolyploids in breeding programs.Here,we analyzed three genetic mapping populations(735 DH lines)derived from crosses between advanced synthetic and conventional Brassica napus(rapeseed)lines,using whole-genome sequencing to determine genome composition.We observed high tolerance of large structural variants,particularly toward the telomeres,and preferential selection for balanced homoeologous exchanges(duplication/deletion events between the A and C genomes resulting in retention of gene/chromosome dosage between homoeologous chromosome pairs),including stable events involving whole chromosomes("pseudoeuploidy").Given the experimental design(all three populations shared a common parent),we were able to observe that parental SV was regularly inherited,showed genetic hitchhiking effects on segregation,and was one of the major factors inducing adjacent novel and larger SV.Surprisingly,novel SV occurred at low frequencies with no significant impacts on observed fertility and yield-related traits in the advanced generation synthetic lines.However,incorporating genome-wide SV in linkage mapping explained significantly more genetic variance for traits.Our results provide a framework for detecting and understanding the occurrence and inheritance of genomic SV in breeding programs,and support the use of synthetic parents as an important source of novel trait variation.

目的探究山药多糖(Chinese yam polysaccharide,CYPS)在地塞米松(dexamethasone,DEX)诱导的3T3-L1胰岛素抵抗(IR-3T3-L1)细胞模型中的降血糖作用。方法将IR-3T3-L1脂肪细胞随机分为对照组(Con)、DEX组(DEX)、Met组(Met,阳性药组)、CYPS组(0.05、0.15、0.45 mg/mL),通过DEX(1μmol/L)诱导建立IR-3T3-L1细胞模型,进行给药后细胞对葡萄糖的消耗量和细胞中血脂(总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C))与丙二醛(malondialdehyde,MDA)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、己糖激酶(hexokinase,HK)、丙酮酸激酶(pyruvate kinase,PK)含量的检测,并采用qRT-PCR对AMPK-PI3K/Akt信号通路中相关基因表达进行检测。结果DEX组与Con组相比,1μmol/L DEX处理48 h显著降低了IR-3T3-L1细胞的葡萄糖消耗量(P<0.01),且在60 h内维持稳定。0.05、0.15、0.45 mg/mL CYPS处理IR-3T3-L1细胞显著增加了细胞中葡萄糖的消耗(P<0.01)、HK、PK、GSH-Px酶活性(P<0.01)、HDL-C含量(P<0.01);降低了MDA酶活性(P<0.01)、T-CHO、TG、LDL-C含量(P<0.01)。同时,CYPS可以显著回调PI3K、Akt、GLUT-4、AMPK、IRS-2、PPARa、Adiponectin的mRNA水平(P<0.05),降低IRS-1、GSK-3β、ACC、FAS的mRNA表达水平(P<0.01)。结论CYPS能够改善DEX诱导的IR-3T3-L1细胞对葡萄糖的消耗,调节脂代谢紊乱,缓解氧化应激,其作用机制可能通过调控IR-3T3-L1脂肪细胞的AMPK-PI3K/Akt信号通路来实现糖脂代谢紊乱和胰岛素抵抗调节来实现。

目的探究黄花菜醇提物(Hemerocallis citrina Baroni,HCE)在利血平(reserpine)诱导的斑马鱼幼鱼(Danio rerio)模型中对抑郁行为的改善作用。方法将斑马鱼幼鱼分为对照(Con)组、利血平组、氟西汀组、黄花菜低剂量组(1.5 mg/L)、黄花菜中剂量组(3 mg/L)、黄花菜高剂量组(4.5 mg/L)。采用斑马鱼声音刺激和光刺激进行抑郁样行为学分析。利用RT-qPCR研究黄花菜醇提物对抑郁症有关的星状胶质细胞标志物及其衍生物基因(GFAP、C3、C4B、EMP-1和S100α-10)以及神经营养因子(BDNF)及其自身受体转录基因(P75、TrkB)的调节功能。结果与对照组相比,模型组在440 Hz声音刺激与8000 Lux光照刺激下运动距离缩短、运动时间减少(P<0.05,P<0.001,P<0.0001),呈现运动的不敏感性。而经过黄花菜醇提物给药后的斑马鱼幼鱼对于光与声音的刺激敏感度相较于模型组有明显升高(P<0.05,P<0.0001)。与对照组相比,利血平组斑马鱼脑中的星形胶质细胞标志物基因表达上调(P<0.0001),而各给药组相较于利血平组,星形胶质细胞标志物表达显著下调(P<0.0001),神经营养因子及其受体基因表达与对照组相比,模型组斑马鱼脑中的BDNF、P75、TrkB下调(P<0.0001),而黄花菜醇提物给药组相较于模型组BDNF、P75、TrkB表达显著上调(P<0.01,P<0.0001)。表明黄花菜醇提物抑制了星形胶质细胞激活诱发的炎症表现,同时促进了神经营养因子及其受体基因的生成,对抑郁起到了改善作用。结论黄花菜醇提物可以改善利血平诱导的斑马鱼幼鱼的抑郁样行为变化,斑马鱼脑中星形胶质细胞标志物的表达量减少,同时促进神经营养因子及其受体基因的生成,起到抗抑郁作用。

目的通过网络药理学技术预测酸枣仁复方（Ziziphi spinosae semen formula，ZSSF）对于抑郁症的关键作用靶点，并通过利血平诱导的抑郁斑马鱼模型验证酸枣仁复方治疗抑郁症的作用机制。方法通过TCMSP数据库检索酸枣仁复方的药物作用靶点，利用UniProt数据库对靶点名称进行校正。采用GeneCards、OMIM、NCBI数据库检索抑郁症相关靶点，并对3个数据库的靶点结果进行了汇总去比重。利用String数据库预测了交集靶点的蛋白-蛋白相互作信息，利用Metascape数据库构建进行富集研究，并利用微生信对KEGG和GO的富集结果实现了可视化。通过利血平诱导的斑马鱼抑郁模型进行行为学实验和RT-qPCR实验，验证酸枣仁复方对抑郁症的治疗作用。结果筛选出抑郁症与酸枣仁复方的交集靶点188个，蛋白质-蛋白质相互作用（proteinprotein interaction，PPI）结果显示，酸枣仁复方抗抑郁主要作用于肿瘤坏死因子-α（TNF-α）、血清白细胞介素2（IL-2）、血清白细胞介素6（IL-6）、血清白细胞介素1β（IL-1β）、血清白细胞介素10（IL-10）等靶点，KEGG通路富集分析表明，酸枣仁复方通过TNF信号通路、PI3K-Akt信号通路、cGMP-PKG信号通路等多种信号通路发挥其治疗抑郁症的作用。动物实验结果显示，与模型组相比，酸枣仁复方高、中、低剂量组斑马鱼在声光刺激下的运动距离显著延长（P < 0.05），运动速度显著增快（P < 0.01）。RT-qPCR结果显示，与模型组相比，酸枣仁复方高、中、低剂量给药组斑马鱼脑组织中TNF-α、IL-2、IL-6、IL-1β、IL-10 mRNA表达水平上调（P < 0.001）。结论酸枣仁复方通过多成分、多靶点发挥抗抑郁作用，且其抗抑郁作用可能与抑制炎症因子的表达有关。

Propagation of light beams in turbid media such as underwater environments,fog,clouds,or biological tissues finds increasingly important applications in science and technology,including bio-imaging,underwater communication,and free-space communication technologies.While many of these applications traditionally relied on conventional,linearly polarized Gaussian beams,light possesses many degrees of freedom that are still largely unexplored,such as spin angular momentum(SAM)and orbital angular momentum(OAM).Here,we present nonlinear light-matter interactions of such complex light beams with"rotational"degrees of freedom in engineered nonlinear colloidal media.By making use of both variational and perturbative approach,we consider non-cylindrical optical vortices,elliptical optical vortices,and higher-order Bessel beams integrated in time(HOBBIT)to predict the dynamics and stability of the evolution of these beams.These results may find applications in many scenarios involving light transmission in strongly scattering environments.

Low-voltage(LV) distribution networks are unbalanced and present loads with nonlinear behavior, which introduce harmonics in the networks. The predictable increase in photovoltaic microgeneration(PV μG) accentuates this unbalanced characteristic, as well as poses new technical problems,namely voltage rise and reverse power flow. To accurately account for the distributed PV and loads in the LV network, unbalanced three-phase power flow algorithms should be utilized,where different approaches may be used to represent lines with various degrees of accuracy. The more accurate algorithm considers the electromagnetic coupling between the line conductors,whereas the simpler algorithm represents each conductor of the line as a single-phase line with pure resistive behavior. This paper aims to analyze the influence of the line model on the load flow in a highly unbalanced LV network with a high penetration of PV production, and considers the impact of the harmonics produced by nonlinear loads. Based on the results obtained,it is possible to identify the most suitable model to be used, depending on the study to be performed. Different scenarios of PV generation and loads are addressed in this paper.

Depression and dementia can lead to generalised cognitive and memory dysfunction.Thus,differentiating these disorders is important and challenging.Pseudodementia is a term used clinically to describe symptoms that resemble dementia but are caused by other conditions(most frequently depression),rather than being recognised as an official diagnosis.Pseudodementia is characterised by a cognitive impairment that mimics dementia but which does not have its origin in neurological degeneration,deriving instead from functional psychiatric conditions.This condition is more commonly observed in older adults(particularly those over the age of 50 or 60 years),and its risk factors overlap with those for depression.Pseudodementia is essentially characterised by deficits in memory,executive function and speech and,therefore,can easily be confused with dementia,although there are aspects that allow its differentiation.Diagnosing pseudodementia can be difficult,especially as there is significant overlap between its symptoms and those of other conditions.However,it is important to recognise characteristic aspects of this disorder,as its correct identification is essential for proper treatment.

Purpose:Non-prosthetic peri-implant fractures are challenging injuries.Multiple factors must be care-fully evaluated for an adequate therapeutic strategy,such as the state of bone healing,the type of implant,the time and performed personnel of previous surgery,and the stability of fixation.The aim of this study is to propose a rationale for the treatment.Methods:The peri-implant femoral fractures(PIFFs)system,a therapeutic algorithm was developed for the management of all patients presenting a subtype A PIFF,based on the type of the original implant(extra-vs.intra-medulllary),implant length and fracture location.The adequacy and reliability of the proposed algorithm and the fracture healing process were assessed at the last clinical follow-up using the Parker mobility score and radiological assessment,respectively.In addition,all complications were noticed.Continuous variables were expressed as mean and standard deviation,or median and range according to their distribution.Categorical variables were expressed as frequency and percentages.Results:This is a retrospective case series of 33 PIFFs,and the mean post-operative Parker mobility score was(5.60±2.54)points.Five patients(15.1%)achieved complete mobility without aids(9 points)and 1(3.0%)patient was not able to walk.Two other patients(6.1%)were non-ambulatory prior to PPIF.The mean follow-up was(21.51±9.12)months(range 6-48 months).There were 7(21.2%)complications equally distributed between patients managed either with nailing or plating.There were no cases of nonunion or mechanical failure of the original implant.Conclusion:The proposed treatment algorithm shows adequate,reliable and straightforward to assist the orthopaedic trauma surgeon on the difficult decision-making process regarding the management of PIFF occurring in previously healed fractures.In addition,it may become a useful tool to optimize the use of the classification,thus potentially improving the outcomes and minimizing complications.
