Synthesis, Characterization of Ruthenium Compounds and Studies of Biological Effects in MCF-7 Tumors Cell Lines

This work presents the synthesis and characterization of compounds derived from the ruthenium transition metal with the nitrogenous ligand 4-aminopy- ridine (4-ampy). The synthesized compounds were characterized by FTIRmed spectroscopy and TG-DTA thermal analysis. For the cytotoxic evaluation of ruthenium compounds, a 66.0 μM aqueous solution containing the complex and the study of data observed in the biological assessment was performed using variance (ANOVA) analysis, followed by Tukey’s multiple comparisons test. Differences between treatments were considered significant when the p-value was less than 0.05 (p 3(4-ampy)(H2O)2]·1/2H2O, which, due to the low solubility in an aqueous medium, was modified to increase its solubility for biological tests. The analysis of the spectra in the medium infrared region (FTIR) for the complex [Ru(Cl)3(4-ampy)(H2O)2]·1/2H2O, shows displacements of the bands observed at 1625 - 1566 cm﹣1 ν(C=C) e (C=N), indicating that coordination to the metallic center occurred by this group. Band displacements were observed in the modified Ru (III) complex, which suggests the presence of the 4-ampy ligand and the coordination by the groups ν(C=C) and (C=N) after the modification. In recent years, researchers worldwide have concentrated on obtaining, developing, and modifying drugs used as chemotherapeutic agents. The evaluation of the cell viability of the modified Ru (III) compound demonstrated cytotoxic effects in the MCF-7 cell line (15.33% ± DP 2.7) but did not affect normal cells (PBMC), which reflects the potential for possible applications.

Superoxide Anion Release by Human Blood Phagocytes Can Increase the Microbicidal Activity Induced by a New Microemulsioned System Containing Cotton Oil

The study aimed to develop and characterize a microemulsified system based on cotton oil and verify its effect on superoxide release anion and microbicidal activity by human peripheral blood cells. Microemulsions were formulated using distilled water, degummed cotton oil, Span 80 (SP), Tween 80 (TW), and 1-butanol (BT). The pseudo-ternary diagram delimited ME regions, and the points were pre-selected. The physical-chemical and rheological characterization of the microemulsions was carried out. The ME activity on the interactions between leukocytes and bacteria was analyzed by superoxide release, phagocytosis, and microbicidal activity. The developed formulation was classified as Oil/Water, with an average pH of 5.76, and the viscosity showed resistance to temperature changes. The rheological model of the Power Law classified the microemulsion as a non-Newtonian fluid with pseudoplastic characteristics. The cell viability of cotton oil microemulsion was greater than 90%. There was an increase in the superoxide release by MN phagocytes when treated with cotton oil microemulsion. The cotton oil microemulsion increased phagocytosis and microbicidal activity. The present study suggests that cotton oil is an alternative biomaterial for therapeutic applications, especially in treating infections.

Caffeine is an alkaloid present in a wide variety of plants. Currently the most consumed psychostimulant worldwide, its consumption is associated with several health benefits, including modulation of the innate and adaptive immune response, reduction of oxidative cellular stress, and decreased incidence of some cancers, including breast cancer. Breast cancer is the most common cause of cancer among women and the second leading cause of cancer death in women worldwide. The interaction between biomaterials and drugs has enabled a great advance in science for developing controlled drug delivery systems and has been used to treat numerous pathologies. This work aimed to evaluate the immunomodulatory effects of caffeine associated or not with polyethylene glycol adsorbed in microemulsion (MLP) on MCF-7 cells, phagocytic cells (MN), and coculture. For biological assays, ATCC (American Type Culture Collection, USA) cell lines of breast adenocarcinoma (MCF-7) and phagocytes (MN) obtained from voluntary donors were used. The cells (MN and MCF-7) and coculture were treated with caffeine and MLP and incubated for rheological characterization analyses: flow curve and viscosity, oxidative stress: superoxide anion assay (), and activity of the enzyme superoxide dismutase (Cu-Zn-SOD). Caffeine and MLP increased viscosity and blood and MCF-7 cells and affected the immunomodulation of oxidative stress metabolism of MN and MCF-7 cells treated with caffeine and associated caffeine to the MLP. These data suggest that caffeine is associated or not with MLP-induced immunomodulatory effects on MN phagocytes and MCF-7 cells, demonstrating the antitumor activity via oxidative stress and can be a complementary alternative for treating breast cancer.

Synthesis, Characterization, and Evaluation of Antitumor Potential in MCF-7 Cells of Ruthenium-Derived Compounds

To synthesize, characterize and evaluate the antitumor potential derived from ruthenium compounds was generated in this study, from the precursor K[RuCl4(bipy)] a route in a simple and reproducible synthesis for a novel compound of coordinating Ru+3 with bipy and L-trip. The spectroscopic characterization in the middle infrared region (FTIR) shows the interactions between Ru-(L-trip), evidenced by the displacement of the carboxylate ion band for higher energies, and also by the displacements of aliphatic amine bands, suggesting that bidentate coordination of the L-trip ligand occurred. Analysis of the results obtained with thermoanalytical techniques showed that the minimum formula of the compound, [RuCl2(bipy)(L-trip)]1/2H2O. Evaluation of the antitumor potential of precursor K[RuCl4(bipy)] showed the toxic effects on MCF-7 cell line, but did not show selectivity and not reached PBMC cells to the same extent. The evaluation of the antitumor potential of the newly synthesized compound, [RuCl2(bipy)(L-trip)], demonstrated that the insertion of an L-tryptophan molecule into the precursor coordination sphere made it selective when compared to PBMC cells, for MCF-7 type tumor cells.

Background: Liver transplantation is the treatment for end-stage liver diseases and well-selected malignancies. The allograft shortage may be alleviated with living donation. The initial UCLouvain experience of adult living-donor liver transplantation(LDLT) is presented. Methods: A retrospective analysis of 64 adult-to-adult LDLTs performed at our institution between 1998 and 2016 was conducted. The median age of 29(45.3%) females and 35(54.7%) males was 50.2 years(interquartile range, IQR 32.9–57.5). Twenty-two(34.4%) recipients had no portal hypertension. Three(4.7%) patients had a benign and 33(51.6%) a malignant tumor [19(29.7%) hepatocellular cancer, 11(17.2%) secondary cancer and one(1.6%) each hemangioendothelioma, hepatoblastoma and embryonal liver sarcoma]. Median donor and recipient follow-ups were 93 months(IQR 41–159) and 39 months(22–91), respectively. Results: Right and left hemi-livers were implanted in 39(60.9%) and 25(39.1%) cases, respectively. Median weights of right-and left-liver were 810 g(IQR 730–940) and 454 g(IQR 394–534), respectively. Graft-to-recipient weight ratios(GRWRs) were 1.17%(right, IQR 0.98%-1.4%) and 0.77%(left, 0.59%-0.95%). One-and five-year patient survivals were 85% and 71%(right) vs. 84% and 58%(left), respectively. Oneand five-year graft survivals were 74% and 61%(right) vs. 76% and 53%(left), respectively. The patient and graft survival of right and left grafts and of very small( < 0.6%), small(0.6%–0.79%) and large( ≥0.8%) GRWR were similar. Survival of very small grafts was 86% and 86% at 3-and 12-month. No donor died while five(7.8%) developed a Clavien–Dindo complication IIIa, IIIb or IV. Recipient morbidity consisted mainly of biliary and vascular complications; three(4.7%) recipients developed a small-for-size syndrome according to the Kyushu criteria. Conclusions: Adult-to-adult LDLT is a demanding procedure that widens therapeutic possibilities of many hepatobiliary diseases. The donor procedure can be done safely

The focus of this study was to investigate the wide use of Eucalyptus spp.,an exotic plant with high allelopathic ability,in the reforestation programs of urban parks in São Paulo City,Brazil,over the last *** understand the implications of using Eucalyptus *** the city’s parks,this study aimed to compare nutrient cycling and litter decomposition between a mixed composition of litter(i.e.,native and exotic species)and the litter of a single species(i.e.,Eucalyptus).To accomplish this,newly deciduous leaves were collected from two native and two exotic species that are commonly used in the afforestation of São Paulo as well as from Eucalyptus *** mixed composition of litter yielded a higher dry mass loss and return of macro-and micronutrients to the forest *** decomposition constant(k)values were 0.00322 and 0.00207 g g^(−1) day^(−1) for the mixed composition and Eucalyptus spp.,*** time required for decomposition of 50 and 95%of deciduous material was 215 and 931 days,respectively,and for the mixed litterfall 334 and 1449 days,respectively,for Eucalyptus ***,the mixed litter exhibited greater dry mass loss and nutrient cycling in an urban forest of São Paulo City,since dry mass losses as well as speed and amount of nutrients returned to the forest floor were relatively higher compared to Eucalyptus *** cycling via Eucalyptus *** was less efficient than mixed composition of litter,demonstrating that reforestation programs carried out in the twentieth century using only one species may have had little *** results of this work emphasize the fact that in urban reforestation programs the City of São Paulo must consider the environmental and biogeographic characteristics of the species employed and use high levels of biodiversity,since the city lies in a megadiverse biome.

Physiological Effect of Kinetin on the Photosynthetic Apparatus and Antioxidant Enzymes Activities During Production of Anthurium

The results observed in the literature raise the hypothesis according to which cytokinin plays important roles in photosynthetic metabolisms and antioxidant enzymes. Thus, the study aimed to evaluate the effect of foliar application of the isolated cytokinin kinetin at the production cycle, seeking to analyze its effects on enzyme activity and photosynthetic parameters. The plants treated with CK presented reduction of leaf CO_2 assimilation rate(Pn) and stomatal conductance(Gs), while that transpiration rate(Tr) was unaffected. The internal CO_2 concentrations decreased with the increase in cytokinin levels, but were maintained under CK 50 mg·L^(-1). The plants treated with CK 75 mg·L^(-1) was verified higher carboxylation efficiency(Pn/Ci), which was associated to values of CO_2 assimilation and transpiration unaltered. Apparent electron transport rate showed variations in the concentration of 25 mg·L^(-1). Considering the study of enzyme activity, on the other hand, it cannot be stated that kinetin has an effective action in delaying oxidative damage. It presents mixed results, since an efficiency in the application of cytokinin was not observed, presenting induction levels of ascorbate peroxidase activity. Thus, further research is needed to determine more precisely the effects of kinetin on gas exchange and antioxidant enzymes in anthurium plants.

Background: Nowadays we observe growing rates of obesity, mainly among women. It has a great impact on maternal and fetal morbimortality and requires a specific obstetric approach. Aim: To discuss different approaches to abdominal incision in cesarean section in obese patients. Case presentation: Patient with morbid obesity submitted to an elective cesarean section due to fetal macrosomia and who underwent a cesarean section with a supraumbilical median incision evolving without any postpartum complications. Conclusion: It’s important to evaluate and chose the best incision in morbidly obese pregnant women aiming to reduce difficulties in fetal extraction and postoperative complications. Midline supraumbilical incision is a good choice in these cases, reducing surgical time and postpartum hemorrhage.

Synthesis and Characterization of a New Compound of Cobalt Ⅱ with Isonicotinamide and Evaluation of the Bactericidal Potential

In this work, the isonicotinamide was coordinated to the Cobalt ion in oxidation state +2. The relevance of this work is the investigation of the in vitro bactericidal potential of the synthesized complex when tested in Gram-positive and negative bacteria strains. This study is motivated by the need to obtain new materials that have antibiotic properties and that, in the future, may become an effective drug against resistant bacteria. A new coordination compound of Cobalt and isonicotinamide, [Co(H2O)(isn)3](BF4)2, was synthesized and described. The compound was characterized by thermoanalytical techniques TG-DTG and TG-DSC, where it was possible to propose the mechanism of thermal decomposition. Through the spectroscopy in the region of the medium infrared (FTIR), it is possible to infer the site of connection between isonicotinamide and metal. The bactericidal activity of [Co(H2O)(isn)3](BF4)2, CoCl2 and free Isonicotinamide were tested for the bacteria Streptococcus mutans (Gram+) and Escherichia coli (Gram−) and the synthesized compound showed to be sensitive for both bacteria.
