S-adenosyl-L-methionine modifies antioxidant-enzymes,glutathione-biosynthesis and methionine adenosyltransfera.es-1/2 in hepatitis C virus-expressing cells

AIM: To elucidate the mechanism(s) by which S-adenosyl-L-methionine(SAM) decreases hepatitis C virus(HCV) ***: We examined the effects of SAM on viral expression using an HCV subgenomic replicon cell culture system. Huh7 HCV-replicon cells were treated with 1 mmol/L SAM for different times(24-72 h), then total RNA and proteins were isolated. c DNA was synthesized and real time-PCR was achieved to quantify HCV-RNA, superoxide dismutase 1 and 2(SOD-1, SOD-2) catalase, thioredoxin 1, methionine adenosyltransfera.e 1A and 2A(MAT1A, MAT2A) expression, and GAPDH and RPS18 as endogenous genes. Expression of cellular and viral protein was evaluated by western-blot analysis using antibodies vs HCV-NS5 A, SOD-1, SOD-2, catalase, thioredoxin-1, MAT1 A, MAT2 A, GAPDH and actin. Total glutathione levels were measured at different times by Ellman's recycling method(0-24 h). Reactive oxidative species(ROS) levels were quantified by the dichlorofluorescein assay(0-48 h); Pyrrolidin dithiocarbamate(PDTC) was tested as an antioxidant control and H2O2 as a positive oxidant ***: SAM exposition decreased HCV-RNA levels 50%-70% compared to non-treated controls(24-72 h). SAM induced a synergic antiviral effect with standard IFN treatment but it was independent of IFN signaling. In addition, 1 mmol/L SAM exposition did not modify viral RNA stability, but it needs cellular translation machinery in order to decrease HCV expression. Total glutathione levels increased upon SAM treatment in HCV-replicon cells. Transcriptional antioxidant enzyme expression(SOD-1, SOD-2 and thioredoxin-1) was increased at different times but interestingly, there was no significant change in ROS levels upon SAM treatment, contrary to what was detected with PDTC treatment, where an average 40% reduction was observed in exposed cells. Hydrocarbon distribution along the Soapaga thrust(Eastern Cordillera, Colombia) based on new strategic geochemistry samples

The Colombian Eastern Cordillera constitutes a region with potential for finding hydrocarbon reserves that are still under-explored, despite the existence of oil and tar sand production. The intense tectonic deformation affects the petroleum systems and increases exploration challenges due to the different generation, maturation, and entrapment conditions that they produce. Diverse geochemical analysis conducted on strategic samples determines that these are likely generated by the same anoxic marine source rock type. Two source rock samples we investigated are characterized by very different biomarker signals as compared to the tar sands and oil samples. Both samples are dominated by hopanes as compared to tri-and tetracyclic terpanes, with very low(Tibasosa Formation) to no(Chipaque Formation) extended tricyclic terpanes. The Soapaga Thrust, along which the analyzed samples are located, has exercised first-rate regional structural control of the distribution of hydrocarbons in the studied area. The uplift of their Mesozoic hanging wall produces intense erosion with sediment transport towards the eastern footwall forming the present infill of the Arcabuco basin. The kitchen areas were connected with the shallower entrapment sectors where excellent reservoir levels such as the Picacho Formation before the Soapaga Thrust movement. The high degree of biodegradation observed in the Picacho Fm. tar sands support early charge before the Soapaga thrust movement, and severe biodegradation following the thrust movement when hydrocarbon supply from the kitchen was interrupted. This work aims to contribute to a better understanding of the geochemical characteristics of the petroleum systems due to the action of the Soapaga Thrust.

Mitochondrial disorders are phenotypically varied, with serious clinical repercussions. Among them, there is the deficiency of combined oxidative phosphorylation of type 20, which occurs due to a defect in the VARS2 gene. This article presents a case of a 2-year-old female with progressive myoclonic epilepsy and psychomotor regression, with refractoriness to multiple anticonvulsants. The diagnosis was only made after the examination was carried out. Therefore, this article highlights the aspects of this rare disease and the importance of the exome for the diagnosis of rare conditions.

H∞position transfer and regulation for floating offshore wind turbines

This paper proposes H∞controller design for platform position transfer and regulation of floating offshore wind *** platform movability of floating wind turbines can be utilized in mitigating the wake effect in the wind farm,thereby maximizing the wind farm's total power capture and *** controller is designed so that aerodynamic force is adjusted to meet the three objectives simultaneously,that is,1)to generate the desired electrical power level,2)to achieve the desired platform position,and 3)to suppress the platform *** acquire sufficient aerodynamic force to move the heavy platform,the pitch-to-stall blade pitching strategy is taken instead of the commonly-used pitch-to-feather *** desired power level is attained by the standard constant-power strategy for the generator torque,while Hstate-feedback control of blade pitch and nacelle yaw angles is adopted for the position regulation and platform oscillation *** constants for the H∞controller design are adjusted to take the trade-off between the position regulation accuracy and the platform motion *** demonstrate the efficiency of the proposed controler,a virtual 5-MW semi-submersible wind turbine is *** results show that the designed H∞controller successfully accomplishes the platform position transfer and regulation as well as the platform oscillation reduction against wind and wave disturbances,and that it outperforms a previously-proposed linear quadratic controller with an integrator.

Sperm quality in mice acutely treated with parathion

Aim: To investigate the toxic effect of a single injection of the organophosphorous agropesticide, parathion, on sper-matogenesis in immature male mice. Methods: Seven-day old mice received a single injection of parathion intraperi-toneally at a dose of 1/3 LD_50. The epididymal sperm count, sperm morphology and chromatin thermal stability wereanalyzed 28 and 50 days after injection. Results: Sperm counts were decreased and teratozoospermia and thermal de-naturation of DNA increased after parathion injection. Sperm parameters were changed to a greater extent in younger an-imals, denoting a higher lability of spermatogenic process at its beginning. The damages could recover a long time afterparathion administration. Conclusion: Organophosphorous agropesticides are testicular toxicants, eliciting reversiblecytotoxic and cytogenetic alterations in germ cells.

Development of a nanostructured filter for pyrolysis wax purification:Effects of particulate filter aids

Wax obtained from the pyrolysis of polyethylene waste can contain inorganic contaminants and must be purified before *** evaluated different filter aids(perlite,sand,and rice husk ash)for the filtration of a crude wax and a pyrolysis wax through a filter medium that was coated with nylon 6 nanofibers to improve filtration *** first examined the performance of the filter aids in improving crude wax filtration,with the filtration time,temperature,and pressure held constant at 1 h,170℃,and 2.5 bar,*** wax was then produced by pyrolysis of high-density polyethylene(HDPE)waste in the temperature range 425-475℃.The best filter aid was perlite,which had a filtrate production rate of 5.47×10^(-5)m^(3)/*** highest oily wax yield(87.8%)was achieved at a pyrolysis temperature of 425℃.The assembly comprising the filter medium coated with nylon 6 nanofibers plus perlite as a filter aid was most suitable for removing impurities from the oily wax generated by the HDPE pyrolysis as it achieved a purification efficiency of 99.9%.

Provenance and drainage system of the Early Cretaceous volcanic detritus in the Himalaya as constrained by detrital zircon geochronology

The age range of the major intra-plate volcanic event that affected the northern Indian margin in the Early Cretaceous is here defined precisely by detrital zircon geochronology. U-Pb ages of Early Cretaceous detrital zircons found in the Cretaceous to the Paleocene sandstones cluster mainly between 142 Ma and 123 Ma in the northern Tethys Himalayan unit, and between 140 Ma and 116 Ma in the southern Tethys Himalayan unit. The youngest and oldest detrital zircons within this group indicate that volcanism in the source areas started in the latest Jurassic and ended by the early Albian. Stratigraphic data indicate that volcaniclastic sedimentation began significantly earlier in southern Tibet （Tithonian） than in Nepal （Valangin- ian）, and considerably later in Spiti and Zanskar （Aptian/AIbian） to the west. This apparent westward migration of magmatism was explained with progressive westward propagation of extensional/transtensional tectonic activity and development of fractures cutting deeply across the Indian continental margin crust. However, detrital zircon geochronology provides no indication of heterochroneity in magmatic activity in the source areas from east to west, and thus lends little support to such a scenario. Westward migration of volcaniclastic sedimentation may thus reflect instead the westward progradation of major drainage systems supplying volcanic detritus sourced from the same volcanic centers in the east. Development of multiple radial drainage away from the domal surface uplift associated with magmatic upwelling, as observed for most large igneous provinces around the world, may also explain why U-Pb ages of detrital zircons tend to cluster around 133-132 Ma （the age of the Cornel igneous province） in Tethys Himalayan units, but around 118-117 Ma （the age of the Rajmahal igneous province） in Lesser Himalayan units.
