Phytochemical bioprospecting, antioxidant, antimicrobial and cytotoxicity activities of saline extract from Tithonia diversifolia(Hemsl) A.Gray leaves

Objective:To evaluate antimicrobial and antioxidant properties of saline extract from Tithonia diversifolia leaves by phytochemical bioprospecting,and investigate its safety against animal cells.Methods:The saline extract was prepared,with NaCl(0.15 M),by constant stirring of the dried and pulverized leaves,followed by volume reduction by lyophilization.The extract was phytochemical characterized using ultra-performance liquid chromatography,and total phenol and flavonoid analysis also was performed.The antioxidant capacity was determined through DPPH*radical,the antimicrobial property was evaluated against standard bacteria and fungi,and the viability assays were performed against mice splenocytes.Results:Fifteen compounds were identified belonging to two main classes terpenoids and phenolics.The extract showed 22.185 mg GAE/g of total phenolic compounds and 3.220 mg QE/g of flavonoid.Moreover,extract showed higher antioxidant ability similar to butylated hydroxytoluene a standard molecule[(3.042±0.019)mg AAE/g and(4.12±0.10)mg AAE/g to saline extract and butylated hydroxytoluene,respectively].The antimicrobial assays demonstrated that the extract had a significant antifungal potential against Candida species and could be used with safety against mice splenocytes,in concentrations lower than 50μg/mL,promoting higher proliferation in these cells.Conclusions:Saline extract from Tithonia diversifolia leaves presents potential antioxidant,antifungal properties and induces immunostimulation in mice splenocytes.

BACKGROUND Challenging lesions,difficult to diagnose through non-invasive methods,constitute an important emotional burden for each patient regarding a still uncertain diagnosis(malignant x benign).In addition,from a therapeutic and prognostic point of view,delay in a definitive diagnosis can lead to worse outcomes.One of the main innovative trends currently is the use of molecular and functional methods to diagnosis.Numerous liver-specific contrast agents havebeen developed and studied in recent years to improve the performance of liver magnetic resonance imaging(MRI).More recently,one of the contrast agents introduced in clinical practice is gadoxetic acid(gadoxetate disodium).AIM To demonstrate the value of the hepatobiliary phases using gadoxetic acid in MRI for the characterization of focal liver lesions(FLL)in clinical practice.METHODS Overall,302 Lesions were studied in 136 patients who underwent MRI exams using gadoxetic acid for the assessment of FLL.Two radiologists independently reviewed the MRI exams using four stages,and categorized them on a 6-point scale,from 0(lesion not detected)to 5(definitely malignant).The stages were:stage 1-images without contrast,stage 2-addition of dynamic phases after contrast(analogous to usual extracellular contrasts),stage 3-addition of hepatobiliary phase after 10 min(HBP 10’),stage 4-hepatobiliary phase after 20 min(HBP 20’)in addition to stage 2.RESULTS The interobserver agreement was high(weighted Kappa coefficient:0.81-1)at all stages in the characterization of benign and malignant FLL.The diagnostic weighted accuracy(Az)was 0.80 in stage 1 and was increased to 0.90 in stage 2.Addition of the hepatobiliary phase increased Az to 0.98 in stage 3,which was also 0.98 in stage 4.CONCLUSION The hepatobiliary sequences improve diagnostic accuracy.With growing potential in the era of precision medicine,the improvement and dissemination of the method among medical specialties can bring benefits in the management of patients with FLL that are difficult to diagnose.

Analysis of alternative parameters of dynamic resistance measurement in high voltage circuit breakers

Throughout its lifetime,high-voltage circuit breaker(HVCB)contacts suffer from inevitable degradation processes,especially during the formation of electric arcs.Highly degraded contacts can reduce the current interruption capacity and increase the chances of operational faults,which might lead to considerable financial losses.Therefore,the use of efficient evaluating techniques is a key factor to achieve systems reliability.The dynamic resistance measurement(DRM)is a technique that can be used to evaluate the degradation level of HVCB'contacts.It stands out from the usual technique,static resistance measurement,by allowing the assessment of both the main contacts and the arcing contacts(ACs).Usually,the mean resistance or the area under the DRM curve is used to estimate the AC degradation.However,DRM is a non-standardised technique and there are no specified parameters to evaluate the real wear conditions.Therefore,an analysis of alternative parameters extractible from the DRM technique is presented in this study,such as amplitude and variance of the DRM curve.The results attest the possible use of more than one parameter for the continuous evaluation of HVCB ACs,characterising the DRM as a more complete and precise diagnosis tool.

Petit-Suisse Cheese Production with Addition of Probiotic <i>Lactobacillus</i><i>casei</i>

Growing concern to increase life expectancy has promoted several studies in the field of nutrition, especially those on food and their effects on the human body. Assays have been performed to improve their nutritional quality. In fact, there is considerable interest in encouraging development of new ingredients, with the innovation in food products and the establishment of new market niches for these ingredients. The global market for functional foods is growing and always envisages new products with functional technological and physiological characteristics. The petitsuisse cheese is a product of high nutritional value, rich in calcium, phosphorus and fat-soluble vitamins, with a high percentage of milk proteins, coupled to excellent digestion and assimilation by the human body. Current assay describes the production of a petit-suisse cheese to which was added the probiotic Lactobacillus casei BGP 93, and determines the physical, chemical and microbiological viability of lactobacilli in cheese during storage period. Levels of protein, fat in the dry extract, moisture and pH were analyzed for physical and chemical parameters;likewise, detection of coliforms, Staphylococcus, Salmonella, Listeria monocytogenes, and yeasts and molds was performed for microbiological parameters;monitoring of the viability of Lactobacillus casei BGP 93 during its life time was conducted. The petit-suisse cheese proved to be a good vehicle for the addition of probiotic microorganism. Lactobacillus casei remained viable during shelf life even with decreasing pH, and may be considered a functional product during 30 days of storage.

Contribution of inrush current to mechanical failure of power transformers windings

This work presents a study regarding the contribution of inrush current to the occurrence of mechanical fatigue in windings of a power transformer and,consequently,the impact of that phenomenon on the equipment's lifetime.In order to perform the study,electromagnetic transient simulations were used to determine the inrush current in a three-phase transformer model.Then,three-dimensional simulations,based on the finite element method,were performed to determine the electromechanical stresses in the equipment windings,under the effect of the inrush current.Next,the methods of critical plans and Palmgren–Miner were applied to perform the fatigue study.From the results,the impact of inrush current on the integrity of the transformer's windings was determined,as well as,to estimate the reduction of lifespan of the equipment due to inrush occurrence.The number of inrushes needed for the total reduction of lifespan of the equipment due to mechanical fatigue was also determined.The results can be used in the transformers design stage,with the purpose of assisting the evaluation of mechanical withstand of equipment,which can minimise the possibility of failure and increase its lifespan.

Food Safety and Fish Farming: Serious Issues for Brazil

Fish farming in Brazil has been growing, and a new branch that has been taking up space and evolving, every year, as a source of leisure, is fish-pay. despite being a leisure practice, this type of fish farming has contributed to the development of Brazilian aquaculture. With the growth of aquaculture in the country, concerns about the quality of the product arise, both financially and microbiologically. Aquaculture has caused some environmental problems, such as the increase in the population growth of bacteria specific to the aquatic environment and the possible proliferation of microorganisms resistant to antibiotics due to the indiscriminate use of these drugs in nurseries. Inadequate water quality conditions can contaminate fish and favor the development of diarrheal infections due to the microbiological load present in the fish. This article aims to review the literature covering the general characteristics of fish, including aspects of nutritional importance, and mechanisms of deterioration and analysis of freshness.

Scrutiny of Microorganisms in Fermented Milk Beverage and Water from Dairy under State Inspection

The aim of the study was to examine microorganisms in samples of fermented milk beverage and water from dairy under state inspection. In a dairy located in the Recôncavo of Bahia, fifteen samples of fermented milk beverage were collected, corresponding to three different batches, and six water samples, two from each point of use in the industry: Point 1—Sanitary barrier;Point 2—Internal faucet for cleaning surfaces and utensils;Point 3—Drinking fountain for employees. All samples were submitted to microbiological research of total coliforms, Escherichia coli and Aeromonas spp. In the water samples, there was also quantification of heterotrophic bacteria and analysis of free residual chlorine. The fermented milk beverage samples did not show growth of total coliforms, E. coli and Aeromonas spp. With regard to water, no sample showed measurable levels of free residual chlorine, while in the microbiological analysis the count of heterotrophic bacteria in the drinking water exceeded the limit established by legislation (5.6 × 102 CFU.mL-1). In addition, the water from the sanitary barrier revealed total coliforms above the allowed value (1.3 × 101 CFU됔 mL-1) and showed contamination by Aeromonas spp. (7.0 × 101 CFU

Microbiological Quality, Transmission of Resistant Bacteria, and Genetic Characterization in Fish Sold in Town Fairs in the Recôncavo da Bahia Region, Brazil

Informal fish marketing is an important source of food and income for many families. However, it is also a public health issue, since the quality of fish can be compromised during marketing. The microbiological quality of fish sold in town fairs in the municipalities of the Bahia Reconcavo region, Brazil, was assessed to characterize isolates based on antimicrobial susceptibility and to investigate gene virulence. Samples of mullets (Mugil sp.), mussels (Mytella sp.), and mollusks (Anomalocardia brasiliana) were collected in town fairs in the municipalities of Cachoeira, Cruz das Almas, and Santo Ant ?nio de Jesus. The presence of mesophylls, molds and yeasts, total coliforms, Escherichia coli, and Staphylococcus aureus was evaluated. E. coli and S. aureus isolates were subjected to antimicrobial susceptibility tests and the presence of the gene stx in E. coli isolates was investigated. Total coliforms and S. aureus ranged between 0 and 9.50 log CFU/g, while high counts of mesophylls, molds, and yeasts were detected. E. coli had greater resistance (76.8%) to amoxicillin and S. aureus had greater sensitivity to gentamicin. The stx gene was not identified in any of the samples studied. Results showed that some fish sold in the town fairs of the three municipalities possessed unsatisfactory microbiological quality, which may pose risks to consumer health, and may be vectors for the dissemination of bacteria resistant to different antimicrobial agents.

Background： Brazil is among the nations with the greatest rates of annual cocaine usage. Pharmacological treatment of cocaine addiction is still limited, opening space for nonconventional interventions.Homeopathic Q-potencies of opium and Erythroxylum coca have been tested in the integrative treatment of cocaine craving among homeless addicts, but this setting had not proven feasible, due to insufficient recruitment.Objective： This study investigates the effectiveness and tolerability of homeopathic Q-potencies of opium and E. coca in the integrative treatment of cocaine craving in a community-based psychosocial rehabilitation setting.design, setting, participants, and interventions： A randomized, double-blind, placebo-controlled, parallelgroup, eight-week pilot trial was performed at the Psychosocial Attention Center for Alcohol and Other Drugs（CAPS-AD）, Sao Carlos/SP, Brazil. Eligible subjects included CAPS-AD patients between 18 and 65 years of age, with an International Classification of Diseases-10 diagnosis of cocaine dependence（F14.2）. The patients were randomly assigned to two treatment groups： psychosocial rehabilitation plus homeopathic Q-potencies of opium and E. coca（homeopathy group）, and psychosocial rehabilitation plus indistinguishable placebo（placebo group）.Main outcome measures： The main outcome measure was the percentage of cocaine-using days. Secondary measures were the Minnesota Cocaine Craving Scale and 12-Item Short-Form Health Survey scores.Adverse events were reported in both groups.Results： The study population comprised 54 patients who attended at least one post-baseline assessment,out of the 104 subjects initially enrolled. The mean percentage of cocaine-using days in the homeopathy group was 18.1%（standard deviation（SD）： 22.3%）, compared to 29.8%（SD： 30.6%） in the placebo group（P 〈0.01）. AIM: To investigate whether Helicobacter pylori (H pykori)infection is associated with hepatitis A virus (HAV) infection, presence of enteroparasites, and other surrogates of fecal exposure.METHODS: We conducted a cross-sectional study in 121 children consecutively admitted at a pediatric hospital inSalvador, Brazil.H pylori and HAV infection were identified by the presence of serum antibodies. Stool specimens were examined for the presence of ova and parasites. A structured questionnaire inquiring about sanitary conditions and life style was applied to each subject. RESULTS: Fifty-one of the 121 children (42.1%) werefound to be seropositive for H pylori, and 45 (37.2%) for HAV. The seroprevalence ofHpyloriand HAV both increased significantly with age. Cross-tabulation of data showed that 26 (21.5%) were seropositive and 51 (42.1%) were negative for bothHpyloriand HAV antibodies (χ2 = 7.18,OR = 2.8, CI 1.30-5.97). The age adjusted OR for an HAV-infected child being H pylori positive was 2.3 (CI1.02-5.03). The agreement between H pylori and HAVseropositivity was fair (κ = 0.24). After controlling for possible confounding, the variables remaining independently associated with seropositivity to H pyloriwere age,presence of Giardia lamblia in feces (OR = 3.2, 95%CI, 1.1-9.5) and poor garbage disposal quality (OR = 2.4,95%CI, 1.1-5.1).CONCLUSION: Our data suggest that H pylori infection is associated with surrogate markers of fecal exposure. Thus, we conclude that the fecal-oral route is relevant in the transmission of HP among children in an urban setting of a developing country. The association observed between G. lamblia and H pylori infection may have several explanations. Further studies to investigate this relationship are warranted.
