Spermatozoa protein alterations in infertile men with bilateral varicocele

Among infertile men, a diagnosis of unilateral varicocele is made in 90% of varicocele cases and bilateral in the remaining varicocele cases. However, there are reports of under-diagnosis of bilateral varicocele among infertile men and that its prevalence is greater than 10%. In this prospective study, we aimed to examine the differentially expressed proteins （DEP） extracted from spermatozoa cells of patients with bilateral varicocele and fertile donors. Subjects consisted of 17 men diagnosed with bilateral varicocele and 10 proven fertile men as healthy controls. Using the LTQ-orbitrap elite hybrid mass spectrometry system, proteomic analysis was done on pooled samples from 3 patients with bilateral varicocele and 5 fertile men. From these samples, 73 DEP were identified of which 58 proteins were differentially expressed, with 7 proteins unique to the bilateral varicocele group and 8 proteins to the fertile control group. Majority of the DEPs were observed to be associated with metabolic processes, stress responses, oxidoreductase activity, enzyme regulation, and immune system processes. Seven DEP were involved in sperm function such as capacitation, motility, and sperm-zona binding. Proteins TEKT3 and TCP11 were validated by Western blot analysis and may serve as potential biomarkers for bilateral varicocele. In this study, we have demonstrated for the first time the presence of DEP and identified proteins with distinct reproductive functions which are altered in infertile men with bilateral varicocele. Functional proteomic profiling provides insight into the mechanistic implications of bilateral varicocele-associated male infertility.

A pilot clinical study of developing an External Assist Targeting Device for rapid and precise renal calyx access during percutaneous nephrolithotomy

Objective:To design a device to increase the accuracy of the targeting process and reduce the radiation exposure to both the patients and the medical ***:We analyzed the inherent problem and designed the External Assist Targeting Device(EATD)to assist in the alignment of needle targeting on the desired renal calyx under fluoroscopic *** EATD was designed to allow rapid and precise access to calyces at all angles,with a simple two-step puncture protocol developed for puncturing a target renal *** then tested the device in a pilot human trial with four ***:In experiments with phantom models,the time for successful targeting was reduced by 31%using the *** mean fluoroscopic time was reduced by 40%.In initial human trial,the puncture time was shortened by 66%and the radiation dose was decreased by 65%compared to free-hand *** complication was observed during the ***:The EATD was found to be cost effective,portable,simple to set up,and safe to operate for assisting in the percutaneous nephrolithotomy *** preliminary tests showed high degree of accuracy in gaining precise access to a targeted renal calyx with much shorter time and lesser radiation *** EATD also has the potential to be used to access other organs with precision under fluoroscopic guidance.

China’s Mars exploration mission has stimulated tremendous interest in planetary science exploration *** propose potential scientific research projects,this study presents a concept simulation for the measurement of Martian atmospheric winds using the Doppler Michelson interferometry *** simulation is based on the satellite instrument initially designed for the Dynamic Atmosphere Mars Observer(DYNAMO)project to measure vertical profiles of winds from the 1.27μm airglow observations in the Martian atmosphere.A comprehensive DYNAMO measurement simulation forward model based on an orbit submodel,an atmospheric background field submodel,and an instrument submodel is developed using the Michelson *** simulated interferogram signal over the field of view(FOV)calculated by the forward model is associated with the filter transmittance function,column emission rate of airglow,wind velocity,temperature,and the Michelson *** agreement between the derived atmospheric signals from the simulated interferogram without altitude inversion and the input parameters used to initiate the forward model confirms the validity of the forward model.

Use of Malaria Rapid Diagnostic Tests among Medical Doctors in a Tertiary Hospital, South East Nigeria

Malaria is a major health problem in Nigeria that has as high as 25% of all global cases, and 30% of deaths attributable to malaria. In 2010, World Health Organization (WHO) recommended universal confirmation of diagnosis of malaria through blood parasitological test, prior to initiation of treatment. In addition to Microscopy, Malaria Rapid diagnostic Tests (mRDTs) are blood tests for malaria. Early diagnosis of malaria ensures that the correct treatment is commenced in good time, and this subsequently improves the prognosis. The objective of this study is to assess the use of mRDTs among medical doctors working in a tertiary hospital, Southeast Nigeria. This study was conducted in Enugu State Teaching hospital, Southeast Nigeria. The study was of cross-sectional design, and conducted in October 2016 among medical doctors working in the General Outpatient, Internal Medicine, and Paediatrics departments of the Teaching hospital. Statistical Package for Social Sciences (SPSS) version 20.0 was used for data analysis. A total of 86 medical doctors, out of the eligible 91 in the three departments participated in the study, and were distributed as follows;24 (27.9%) in the General Outpatient, 30 (34.9%) in the Internal Medicine, and 32 (37.2%) in the Paediatrics departments. More medical doctors in the Paediatrics department (31.3%) used mRDTs in making diagnosis of malaria, followed by Internal medicine (13.3%), then General Outpatient Department (8.3%). Malaria rapid diagnostic tests are cost-effective, and useful tools in malaria control and elimination programmes. If tangible progress on the implementation of the WHO guidelines on confirming diagnosis of malaria before treatment;and the T3: Test, Treat, Track initiative is to be made;then the government and the Management of hospitals ought to take more determined efforts aimed at educating and informing health workers, especially medical doctors on the benefits of mRDTs.

Analysis Unking directly genomics, neuroimaging phenotypes and clinical measurements is crucial for understanding psychiatric disorders, but remains ***, we describe a multi-scale analysis using genome-wide SNPs, gene expression, grey matter volume (GMV), and the positive and negative syndrome scale scores (PANSS) to explore the etiology of schizophrenia. With 72 drug-naive schizophrenic first episode patients (FEPs) and 73 matched heathy controls, we identified 108 genes, from schizophrenia risk genes, that correlated significantly with GMV, which are highly co-expressed in the brain during development. Among these 108 candidates, 19 distinct genes were found associated with 16 brain regions referred to as hot clusters (HCs), primarily in the frontal cortex, sensory-motor regions and temporal and parietal *** patients were subtyped into three groups with distinguishable PANSS scores by the GMV of the identified HCs. Furthermore, we found that HCs with common GMV among patient groups are related to genes that mostly mapped to pathways relevant to neural signaling, which are associated with the risk for *** results provide an integrated view of how genetic variants may affect brain structures that lead to distinct disease *** method of multi-scale analysis that was described in this research, may help to advance the understanding of the etiology of schizophrenia.

Formal Education Related Pattern of Awareness and Basic Knowledge on Zika Virus Disease, among Women Visiting Children Immunization Unit in a Tertiary Hospital, Southeast Nigeria

Zika virus is transmitted through the bite of infected Aedesa egypti, or Aedes albopictus mosquito, and re-emerged as a disease of global public health importance in March/April 2015 in Brazil. The objective of this study is to assess the formal education related pattern of awareness, and basic knowledge on Zika virus disease, among women visiting children immunization unit in a tertiary hospital, in Nigeria. Study was conducted in 2016/2017 with 256 randomly selected respondents. Semi-structured questionnaire was used in the cross-sectional study. Findings including 77.8% of respondents with postgraduate educational qualification were aware that there is a disease known as Zika Virus Disease (ZVD), 62.0% for tertiary education qualification, 53.8% for secondary education level, 20.0% for primary education level, while none of the three respondents that had no formal education were aware of ZVD. The level of awareness increased with increasing educational qualification. These findings were analyzed using the Likelihood Ratio which was calculated to be 28.329, with P-value of <0.001. Television was the commonest source of first ZVD information. Overall mean percentage knowledge level of the different educational groups revealed as followed;no formal education 22.2%, primary level 12.0%, secondary level 46.1%, tertiary level 54.4%, and postgraduate level was 51.5%. The mean for the stated scores was 37.2%. Students’ T-Test at 95.0% confidence interval was 0.012, showing that the difference in the mean percentage scores between the groups was significant. Respondents with tertiary and postgraduate education qualifications recorded overall mean score above 50.0% each, while secondary, primary, and no formal education groups recorded overall mean percentage scores below 50.0%. The largest known odonate in China: Hsiufua chaoi Zhang et Wang, gen. et sp. nov. from the Middle Jurassic of Inner Mongolia

A new genus and species, Hsiufua chaoi Zhang et Wang, is established based on a forewing from the Middle Jurassic Haifanggou Formation in Inner Mongolia, China and attributed to Campterophlebiidae, Isophlebioidea, Odonata. It is the largest odonate known in China and the fourth in the world in terms of forewing length. The maximum size (based on forewing length) of Odonata is smaller in the Permian than in the Mesozoic which is probably due to competition for prey between Odonata and Protodonata in the Permian. The reason that the maximum size of Jurassic odonates is larger than that of their extant relatives is most probably less competition and lower predation pressure from contemporary aerial vertebrates.
