Elimination of viral hepatitis in sub-Saharan Africa by 2030 is an ambitious ***,as stated by the World Health Organization,there are unprecedented opportunities to act and make significant contributions to the elimination *** 60 million people chronically infected with hepatitis B virus(HBV)of whom 38800 are at risk of developing highly fatal hepatocellular carcinoma(HCC)every year,sub-Saharan Africa faces one of the greatest battles towards elimination of viral *** is a need to examine progress in controlling the disproportionate burden of HBV-associated HCC in sub-Saharan Africa within the context of this elimination *** scaling-up coverage of hepatitis B birth dose and early childhood vaccination,we can significantly reduce new cases of HCC by as much as 50%within the next three to five *** the substantial reservoir of chronic HBV carriers however,projections show that HCC incidence and mortality rates in sub-Saharan Africa will double by *** warrants urgent public health *** trends in the burden of HCC over the next two decades,will be determined to a large extent by progress in achieving early diagnosis and appropriate linkage to care for high-risk chronic HBV infected persons.

Background:It remains unclear if the vascular and connective tissue structures of primary and metastatic tumors are intrinsically determined or whether these characteristics are defined by the host *** we examined the microanatomical steps of vasculature and connective tissue development of C38 colon carcinoma in different ***:Tumors produced in mice at five different locations(the cecal wall,skin,liver,lung,and brain)were ana-lyzed using fluorescent immunohistochemistry,electron microscopy and quantitative real-time polymerase chain ***:We found that in the cecal wall,skin,liver,and lung,resident fibroblasts differentiate into collagenous matrix-producing myofibroblasts at the tumor *** activated fibroblasts together with the produced matrix were incorporated by the *** connective tissue development culminated in the appearance of intratumoral tissue columns(centrally located single microvessels embedded in connective tissue and smooth muscle actin-expressing myofibroblasts surrounded by basement membrane).Conversely,in the brain(which lacks fibroblasts),C38 metasta-ses only induced the development of vascularized desmoplastic tissue columns when the growing tumor reached the fibroblast-containing ***:Our data suggest that the desmoplastic host tissue response is induced by tumor-derived fibrogenic molecules acting on host tissue *** concluded that not only the host tissue characteristics but also the tumor-derived fibrogenic signals determine the vascular and connective tissue structure of tumors.

Short-term effects of cleaning-respacing in a silver fir-dominated stand in Romania

This paper discusses the early silviculture of silver fir(Abies alba Mill.), a major softwood in Romania covering more than 294,000 ha. Three research plots(DRP1, DRP2 and DRP3) were established in a 25-year-old silver fir-dominated stand in May 2012. In DRP1 and DRP2, heavy cleaning-respacing were carried out while DRP3 was kept as a control. After the treatments in 2012,the plots had stocking between 1470 trees ha-1(DRP2) and10,030 trees ha-1(DRP3), basal areas between11.8 m2ha-1(DRP2) and 31.6 m2ha-1(DRP3). The average diameters were between 5.7 cm(DRP3) and 9.6 cm(DRP2). Measurements in September 2016 confirmed the rapid response of silver fir to *** best results in average diameter increments and basal area between 2012 and 2016 were in DRP2: 3.7 cm(36.4%) and 7.7 m2ha-1(85.0%) respectively. The largest diameter trees in 2012 were the best growers, and a strong linear correlation between the initial diameter of the remaining trees and their increment was established. Natural dieback of trees occurred only in DRP3(8.6%). The selection of crop trees based on the vigour, quality, and spacing criteria during cleaning-respacing was possible as individuals stood out in terms of vigour and early diameter growth. An issue encountered in DRP1 and DRP2 was the occurrence of epicormic shoots, especially on less vigorous individuals with small crowns. However, four years after cleaning-respacing, these branches are small and do not significantly affect the wood quality.

14-3-3 proteins play an important role in the regulation of many cellular processes. The Arabidopsis vacuolar two-pore K＋ channel 1 （TPK1） interacts with the 14-3-3 protein GRF6 （GF14-λ）. Upon phosphorylation of the putative binding motif in the N-terminus of TPK1, GRF6 binds to TPK1 and activates the potassium channel. In order to gain a deeper understanding of this 14-3-3-mediated signal transduction, we set out to identify the respective kinases, which regulate the phosphorylation status of the 14-3-3 binding motif in TPK1. Here, we report that the calcium-dependent protein kinases （CDPKs） can phosphorylate and thereby activate the 14-3-3 binding motif in TPK1. Focusing on the stress-activated kinase CPK3, we visualized direct and specific interaction of TPK1 with the kinase at the tonoplast in vivo. In line with its proposed role in K＋ homeostasis, TPK1 phosphorylation was found to be induced by salt stress in planta, and both cpk3 and tpkl mutants displayed salt-sensitive phenotypes. Molecular modeling of the TPK1-CPK3 interaction domain provided mechanistic insights into TPK1 stress-regulated phosphorylation responses and pinpointed two arginine residues in the N-terminal 14-3-3 binding motif in TPK1 critical for kinase interaction. Taken together, our studies provide evidence for an essential role of the vacuolar potassium channel TPK1 in salt-stress adaptation as a target of calcium-regulated stress signaling pathways involving Ca2＋, Ca2＋-dependent kinases, and 14-3-3 proteins.

Globally,a shift in the epidemiology of chronic liver disease has been *** has been mainly driven by a marked decline in the prevalence of chronic hepatitis B virus infection(CHB),with the greatest burden restricted to the Western Pacific and sub-Saharan African *** this is a growing burden of metabolic syndrome(MetS)worldwide.A disproportionate co-burden of human immunodeficiency virus(HIV)infection is also reported in sub-Saharan Africa,which poses a further risk of liver-related morbidity and mortality in the *** reviewed the existing evidence base to improve current understanding of the effect of underlying MetS on the development and progression of chronic liver disease during CHB and HIV *** the mechanistic association between CHB and MetS remains poorly resolved,the evidence suggests that MetS may have an additive effect on the liver damage caused by *** HIV infected individuals,MetS-associated liver disease is emerging as an important cause of non-AIDS related morbidity and mortality despite antiretroviral therapy(ART).It is plausible that underlying MetS may lead to adverse outcomes among those with concomitant CHB and HIV ***,this remains to be explored through rigorous longitudinal studies,especially in sub-Saharan ***,there is a need for a comprehensive package of care that integrates ART programs with routine screening for MetS and promotion of lifestyle modification to ensure an improved quality of life among CHB and HIV coinfected individuals.

Energy Absorption by the Membrane Rafts in the Modulated Electro-Hyperthermia (mEHT)

Aim: Heating by nanoparticles, which are located in the tissue to be treated, is a well-recognized method in hyperthermic oncology. Our objective is to investigate selective, nanoscopic heating without concentrating extra artificial nanoparticles. We have in silico calculation to study the heating of the transmembrane protein clusters (rafts) on cell-membrane. The transmembrane protein domains have significantly higher dielectric constant than their lipid neighborhood in the membrane. This difference causes a local gradient in the Specific Absorption Rate (SAR), which could be a factor of heating of the membranes locally, as well as exciting the receptors for various signal transduction in the cells. We suppose that this process determines the observed cellular effects of modulated electro-hyperthermia (mEHT, trade-name: oncothermia). Materials and Methods: In silico models with highly specialized software (Computer Simulation Technology (CST), Darmstadt, Germany) were performed visualizing the selectivity for the membrane domains. Local raft models were created to simulate the electromagnetic (EM) effect of a 13.56 MHz excitation between two perfect electrical conductor plates, simulating the equipotential conditions of the sides of the membrane in the vicinity of the raft. The simulations were performed with near-field (EQS) solver of CST. The electric field, current density, and electric loss density were monitored by the simulations. The applied material properties and parameters refer to the recent literature. Results: In silico models show ten times higher energy-absorption of the transmembrane domains than that of its lipid-membrane surrounding, and intra- and extracellular neighborhood. Depending on the size, orientation, and location of the membrane rafts, the value of SAR varies, but we use only two simplified models to see the absorption properties. Solubilization of 2-Acetoxy-Benzencarboxylic Acid Using Beta Cyclodextrins

2-acetoxy-benzencarboxylic acid is one of the most famous salicylate drugs today, a pharmaceutically active compound known as aspirin. It is poorly soluble in water which results in decreased bioavailability of the drug in the organism. The increase in water solubility of insoluble or poorly soluble drugs is therefore of great importance, which is the aim of this study. Inclusion of the drug in the molecule with a higher water solubility significantly increases its solubility and biological availability. Natural and hydrophilic derivatives of natural cyclodextrins are in the spotlight for their role as solubilizing excipients. Studies indicate that the use of β-cyclodextrin inclusion complexes with acetylsalicylic acid formed, increases the solubility of the drug in water. Many advantages of drug-complexation with cyclodextrins have been reported in scientific literature which includes increased solubility, enhanced bioavailability, improved stability, masking of bad test or odour, reduced side effect. Orally administered aspirin requires high and frequent dosing because it undergoes extensive pre systematic metabolism. Also chronic oral aspirin use is associated with serious gastrointestinal side-effects. Complexation with CD alleviates the side effects to some extent. The bioavailability and solubility of aspirin has to be increased to overcome the side-effects of aspirin related to stomach and gastro intestinal tract. The phase solubility study was performed according to the method of Higuchi and Connors by adding the 2-acetoxi-benzencarboxylic acid in excess to different concentrations of different beta cyclodextrins solutions. Phase solubility study records shown that the stability constant and complex stoichiometry of 2-acetoxi-benzencarboxylic acid-CD complexes gives linearly improve with the concentration of CD. Complexes were analyzed by UV-VIS spectroscopy and were characterized by infrared spectroscopy.
