Compounds with dual action on cyclic adenosine monophosphate （cAMP） and cyclic guanosine monophosphate （cGMP） may be a treatment option for erectile dysfunction, as they not only promote penile erection but also prevent the upregulation of phosphodiesterase-5. In this study, we examined the possible relaxant effect and mechanism of 17-nor-subincanadine E （SEC, 0.2-200 pmol I^-1）, a plant-derived alkaloid, in rabbit corpus cavernosum （rbCC） strips that had been precontracted by exposure to phenylephrine （10 pmol I^- 1） or a high concentration of K＋（60 mmol I^- 1） in vitro. In addition to SEC＇s effect on cAMP and cGMP levels, electrical field stimulation （EFS） in phenylephrine-precontracted rbCC and calcium chloride （1-100 mmol I^-1） evoked responses in depolarized rbCC were analysed. SEC relaxed the phenylephrine-precontracted rbCCs in a concentration-dependent manner. Atropine, guanethidine and N-co-nitro-L-arginine methyl ester （L-NAME） did not have any effect on the relaxation of rBCCs. When 1H-1,2,4oxadiazole[4,3-a] quinoxalin-1-one （ODQ） was added, it effectively blocked the relaxant response of SEC. Although SEC enhanced the maximal relaxation produced by sodium nitroprusside （SNP） and forskolin in phenylephrine-precontracted cavernosal smooth muscle, it caused a decrease in the maximal contractile response induced by calcium chloride in depolarized rbCCs. The relaxant effect of SEC was paralleled by an increase in the tissue levels of the Cyclic nucleotides cAMP and cGMP. We conclude that SEC promotes the relaxation of rbCC, possibly favouring cAMP and cGMP accumulation and calcium blockade. This novel mechanism could be useful for patients who do not benefit from phosphodiesterase inhibitors and for those with endothelial and nitrergic dysfunction, such as patients with diabetes, hypertension and dyslipidaemias.

The use of Vis-NIr-SWIr spectroscopy in the prediction of soil available ions after application of rock powder

Some of the problems attributed to traditional laboratory analyses that limit the correct assessment of the nutrient contents in the soil are time requirements and high cost of the soil nutrient *** solve these problems,a study was carried out to evaluate the use of visible,near-infrared,and short-wave infrared(Vis-NIr-SWIr)spectroscopy in the prediction of soil available ions submitted to the application of rock *** study was carried out on an Arenosol in ParanavaíCity/***(rock powders)were arranged within a split-plot system designed in randomized blocks with four *** was cultivated for 14 months after the application of rock ***,96 soil samples were collected for measuring the pH and available ions P,K^(+),Ca^(2+),Mg^(2+),S-SO_(4)^(2-),Si,Cu^(2+),Fe^(2+),Mn^(2+),and Zn^(2+)as well as spectral reading through a Vis-NIr-SWIr spectroradiometer to predict the soil chemical attributes through the partial least square regression(PLS)*** results showed that the elements K^(+),Ca^(2+),Mg^(2+),Cu^(2+),and Fe^(2+)could be predicted with a reasonable rightness degree(r^(2)_(p)>0.50,rPDp>1.40)from spectral ***,for the attributes pH,P,S-SO_(4)^(2-),Si,Mn^(2+),and Zn^(2+),there were no satisfactory models(r^(2)_(p)rPDprock powder changed the spectral curves and,because of that,allows the building of PLS models to predict the elements K^(+),Ca^(2+),Mg^(2+),Cu^(2+),and Fe^(2+).Therefore,Vis-NIr-SWIr spectroscopy is a promising alternative to the routine analyses of soil fertility since it has advantages such as fast analytical speed,low cost,easy to operate,non-destructive,and environmentally friendly,because it does not use harmful chemicals.

There is a recent and growing interest in assessing differential responders to resistance training(rT)for diverse ***,the individual ability to respond to an intervention for a specific measurement,called responsiveness,remains to be better ***,the current study aimed to summarize the available information about the effects of rT on functional performance and muscle strength,power,and size in healthy adults,through the prevalence rate in different responsiveness classifications models.A systematic review was conducted according to the Preferred reporting Items for Systematic reviews and Meta-Analyses(PrISMA)and was registered at the International Prospective register of Systematic reviews(PrOSPErO,CrD42021265378).PubMed/MEDLINE,Scopus,and Embase databases were systematically searched in October 2023.A total of 13 studies were included,totaling 921 *** two studies presented a low risk of *** the effectiveness of rT,the prevalence rate for non-responders ranged from 0%to 44%for muscle strength,from 0%to 84%for muscle size,and from 0%to 42%for functional performance,while for muscle power,the only study found showed a responsiveness rate of 37%.In conclusion,a wide range of differential responders is described for all variables ***,the evidence summarized in this systematic review suggested some caution while interpreting the findings,since the body of evidence found seems to be incipient,and widely heterogeneous in methodological and statistical aspects.

Extracellular vesicles-coupled mirNAs from oviduct and uterus modulate signaling pathways related to lipid metabolism and bovine early embryo development

Background Extracellular vesicles(EVs) present in oviductal(OF) and uterine fluid(UF) have been shown to enhance bovine embryo quality during in vitro culture by reducing lipid contents and modulating lipid metabolism-related genes(LMGs), while also influencing cell proliferation, suggesting their involvement on the regulation of different biological pathways. The regulation of signaling pathways related to cell differentiation, proliferation, and metabo-lism is crucial for early embryo development and can determine the success or failure of the pregnancy. Bioactive molecules within EVs in maternal reproductive fluids, such as micro rNAs(mirNAs), may contribute to this regulatory process as they modulate gene expression through post-transcriptional *** This study evaluated mirNA cargo in OF-EVs from the early luteal phase and UF-EVs from the mid-luteal phase, coinciding with embryo transit within oviduct and uterus in vivo, and its possible influence on LMGs and sign-aling pathways crucial for early embryo development. A total of 333 mirNAs were detected, with 11 exclusive to OF, 59 to UF, and 263 were common between both groups. From the 20 differentially expressed mirNAs, 19 up-regulated in UF-EVs(bta-mir-134, bta-mir-151-3p, bta-mir-155, bta-mir-188, bta-mir-181b, bta-mir-181d, bta-mir-224, bta-mir-23b-3p, bta-mir-24-3p, bta-mir-27a-3p, bta-mir-29a, bta-mir-324, bta-mir-326, bta-mir-345-3p, bta-mir-410, bta-mir-652, bta-mir-677, bta-mir-873 and bta-mir-708) and one(bta-mir-148b) in OF-EVs. Numerical Solutions of the Classical and Modified Buckley-Leverett Equations Applied to Two-Phase Fluid Flow

Climate change is a reality. The burning of fossil fuels from oil, natural gas and coal is responsible for much of the pollution and the increase in the planet’s average temperature, which has raised discussions on the subject, given the emergencies related to climate. An energy transition to clean and renewable sources is necessary and urgent, but it will not be quick. In this sense, increasing the efficiency of oil extraction from existing sources is crucial, to avoid waste and the drilling of new wells. The purpose of this work was to add diffusive and dispersive terms to the Buckley-Leverett equation in order to incorporate extra phenomena in the temporal evolution between the water-oil and oil-water transitions in the pipeline. For this, the modified Buckley-Leverett equation was discretized via essentially weighted non-oscillatory schemes, coupled with a three-stage runge-Kutta and a fourth-order centered finite difference methods. Then, computational simulations were performed and the results showed that new features emerge in the transitions, when compared to classical simulations. For instance, the dispersive term inhibits the diffusive term, adding oscillations, which indicates that the absorption of the fluid by the porous medium occurs in a non-homogeneous manner. Therefore, based on research such as this, decisions can be made regarding the replacement of the porous medium or the insertion of new components to delay the replacement.

Evidence-Based Complementary and Alternative Medicine Effects of Capybara Oil on Cardiac remodeling of C57bl/6 Mice

Cardiovascular diseases are the main cause of morbidity and mortality in the world, and obesity and the metabolic syndrome are risk factors for its development. One of the therapies to reduce cardiovascular risk is the use of polyunsaturated fatty acids. In Brazil, a source of such acid is the oil extracted from the fat of the capybara. The objective of this work is to study the effects of the capybara oil on lipid and glucose metabolism, as well as its effects on the adipose tissue and cardiac remodeling. We assessed the effects of capybara oil treatment on body mass, lipid and carbohydrate metabolism, systolic blood pressure, adipose tissue and cardiac remodeling, and performed an ultrastructural evaluation of the myocardium in C57Bl/6 mice treated with high-fat diet. Treatment with capybara oil reduced total cholesterol and triglyceride levels, systolic blood pressure, visceral and subcutaneous adipose tissue, and adipocyte diameter. In addition, cardiac remodeling was attenuated, preserving cardiomyocytes, increasing vascularization, reducing cardiomyocyte hypertrophy and the extracellular matrix, and preserving the morphological integrity of mitochondria. Capybara oil has several beneficial effects on the cardiovascular and metabolic system, and further studies are needed to better understand its role in the prevention or treatment of cardiovascular diseases.

Green Hydrogen: Perspectives and Challenges in Using the Natural Gas Network in Ceará/Brazil

Climate change, mainly caused by the use of non-renewable fuels, has raised global concerns and led to the search for less polluting energy sources, making hydrogen a promising energy alternative with the potential to contribute to changes in the energy mix of various countries through the use of technologies that enable its production and use with low or zero carbon emissions. In this context, Brazil has aroused great interest from other countries in exploring its renewable resources for the production of hydrogen (green hydrogen). In this sense, the use of natural gas pipelines and the use of hydrogen in mixtures with natural gas have become the subject of studies due to their economically viable alternative for the immediate use of this energy vector. However, there are still technical and regulatory challenges regarding the integration of hydrogen into the existing natural gas pipeline network. In this context, the present study aims to address the effects of hydrogen interaction with the structure of natural gas pipeline steel and the regulatory barriers to the use of this network for the transportation of green hydrogen, particularly in the state of Ceará/Brazil. After extensive analysis of literature and regulatory documents, it was concluded that: 1) Ceará/Brazil has strong potential to meet the demand for green hydrogen through the use of solar and wind energy sources;2) there is feasibility for the adaptation or conversion of natural gas infrastructure for the transportation of green hydrogen;3) discussions regarding the regulatory competence of green hydrogen transportation and distribution through the natural gas network in Brazil are still incipient;4) the current regulation of the natural gas industry can serve as a subsidy for the regulation of green hydrogen and natural gas transportation.
