Microsatellites and the Polyploid Guarana Plant:Diversity under a Sea of Alleles

Repeat blocks, microsatellites or simple sequence repeats (SSRs) can produce good co-dominant molecular markers for genetic diversity analysis and the determination of self-pollination rates in progenies originating from open pollination of selected genotypes. The enrichment of guarana genomic libraries was underway when it was confirmed that we are working with a complex polyploid species with 210 chromosomes. The probes (CA)12, (CT)12 and (TC)14 were used to finish the enrichment of four libraries for repeat blocks and the screening of a databank of expressed sequence tags (ESTs) from guarana seeded-fruits was accomplished as well. Fifteen clonal cultivars were genotyped with three replicas at 10 out of 27 identified loci using the 59 alleles that passed the reproducibility criterion. A large number of short repeat blocks were identified and this was considered to be a consequence of the recent polyploidization event. However, blocks with eight or more repeats ideal for genotyping were scarce. Annealing of most probes to short blocks by partial complementarity could explain the scarcity of longer blocks in genomic libraries but cannot explain why they were rare in the ESTs. Due to the complexity of the genotypes, alleles were treated as dominant traits. ESTs harboring repeat blocks had the functional annotation renewed. Locus GRN07 is inserted in a homologue of the MOTHER OF FLOWERING LOCUS T AND TFL1 (MFT), in which 3’-UTR displays clear post-transcriptional regulatory features. MFT and its variants are probably involved in the determination of seed germination and embryo growth characteristics. Other accessed loci can be involved in plant architecture and defense reactions. It was concluded that the alleles described in the present work can be used to distinguish guarana cultivars and possibly to analyze segregation using the progenies of controlled pollinations between divergent genitors. Agronomic Performance of Cultivars of Upland Rice in the Southern of the Region of Rondonia,Brazil

The upland rice is grown in most Brazilian states. However, in many of them the yield obtained is low. The choice of cultivar more suited to a particular environment can contribute to increased productivity of this crop. This research aimed to evaluate the performance of a hybrid cultivar of upland rice (Ecco) and five conventional cultivars (BRS Monarca, BRS Primavera, AN Cambará, BRS Sertaneja and BRS MG Curinga) in two environments (Vilhena and Cerejeiras). The tests were conducted in the agricultural year 2010/2011. We evaluated the following characteristics: tillering, panicle number per unit area, number of filled grains per panicle sterile, mass of grains per panicle, 1000 grain weight, grain yield (kg·ha-1) and whole grain yield. There was no cultivar x environment interaction for all traits. Greater tillering, higher panicle number per area, higher productivity and yield of whole grains in the municipality of Cerejeiras were observed, while the highest number of sterile grains per panicle was obtained in Vilhena. The hybrid Ecco highlighted in relation to all other cultivars is in relation to tillering, panicle number per area and grain yield. The municipality of Cerejeiras is the most suitable for the cultivation of upland rice.

Educational interventions aimed at improving the oral health conditions of workers: A critical review

The aim of this research was to identify studies describing the educational and/or health-promoting interventions and strategies used in various programs of oral healthcare conducted in populations of industrial workers. A review of the Science, MEDLINE, LILACS and SCIELO databases was performed for the period between 1980 and 2009. The criteria for a publication to be in- cluded were: 1) the population had to consist of adult workers;and 2) the educational and/or health-promoting interventions had to have been described. A great variety of educational strategies was found, ranging from individual counseling to small-group discussions and instructions on oral hygiene with supervised tooth brushing. The presence of plaque and gingivitis were the most commonly used indicators of the effect of these interventions. Cost evaluation was also an important topic. In general, the indicators analyzed reflected positive results that included a reduction in the indicators themselves (plaque and gingivitis) and in the cost of the programs, suggesting a certain effect of the educational programs developed. Nevertheless, differences in forms of exposure according to the type of occupation are not discussed and neither are specific social determinants such as income and education level. The quality of the studies is limited, which reduces the robustness of the evidence on the effect of educational programs on the work population. Studies should be carried out using mixed approaches to evaluate the efficacy of these programs and propose improvements.

Effect of Pseudomonas fluorescensand Burkholderia pyrrociniaon the Growth Improvement and Physiological Responses in Brachiaria brizantha

The use of beneficial microorganisms in forage grasses is a potentially advantageous technique for a more sustainable pasture management by decreasing the need for chemical fertilization. Our aims were to determine the best method of microorganism inoculation on Brachiaria (Syn. Urochloa) brizantha cv. BRS Piata, compare the responses of inoculated plants of this forage grass with fertilized and unfertilized controls and examine its effect on some morphological, physiological and biochemical responses. On the first experiment, three inoculation methods were tested: in the seed, seed and soil, and soil, with Pseudomonas fluorescens (BRM-32111) and Burkholderia pyrrocinia (BRM-32113). In the second experiment, fertilized and unfertilized plants were either inoculated with BRM-32111, BRM-32113 and co-inoculated (BRM-32111 + BRM-32113). In a final experiment, B. brizantha was inoculated by soil drenching with BRM-32111, BRM-32113 and co-inoculated (BRM-32111 + BRM-32113), and compared to fertilized- and unfertilized-controls. The inoculation by soil drenching, at seedling stage, was more effective than inoculation only in the seed or both in the seed and by soil drenching. The fertilizer may have suppressed the beneficial bacterial effects on the growth of B. brizantha. P. fluorescens and B. pyrrocinia co-inoculated increased nitrate, protein, nitrogen concentration, Spad index (chlorophyll content), leaf area, number of tillers, net photosynthesis and total biomass production of B. brizantha plants. Our results point out to a potentially valuable source of practical information in the search of an eco-friendlier approach to increase pasture productivity.

Construction and Validation of an Educational Technology for Mammary Health Promotion

Mammary health is issue faced by public health nurses and the science follows new ways into this thematic area of learning course wares. The purpose of this study was to construct and to validate a distance course for mammary health promotion for nurses. Methodological proposal has been composed of five stages: 1) analysis and planning, 2) modeling, 3) implementation, 4) evaluation and maintenance and 5) distribution. A total of nine experts as research population were consulted who were expert judges of nursing, pedagogy and web designer, three of each specific area, which were included in the study. The variables that had concordance index of 50% were: graphic designer pages favors learning, the user has easy navigation, the learning of the student experience, the environment provides interdisciplinary, content is built on process rather than there is a unique sequence, allows access to the available environment and holds a discussion on the subject. The distance learning course is intended to broaden and to enrich the living spaces and construct their own knowledge, favors interactivity, communication, autonomy and cooperation between the participants and provides access to educational technologies.

Evaluation of the Therapeutic Response and Improvement in the Quality of Life of the Patient with darier Disease to Acitretin through an Analogy between Close Relatives

darier disease (DD), also known as follicular keratosis, is a rare autosomal dominant hereditary acantholytic dermatosis caused by a mutation in the ATP2A2 gene. Skin lesions are characterized by persistent confluent papules, preferentially located on the seborrheic areas of the face, chest and back. The hands, feet and nails are also commonly affected. DD has a chronic course with frequent relapses. Their treatment is often difficult and unsatisfactory. We present the case of a 54-year-old male with a history of keratotic papules with a brownish-black color, with a foul-smelling, pruritic and ardent appearance on the back and trunk, since the age of 19, which worsen in the summer. His 27-year-old daughter has a similar picture with her skin lesions started at age of 17 in the region below the breast, back, trunk, feet and hands. After the skin biopsy, the diagnosis was confirmed and acitretin 30 mg/day was instituted. The importance of this case report is to alert health professionals that the diagnosis of DD should be considered in family members of patients, especially those in the first degree, and also in patients with follicular keratolytic dermatoses, being an important differential diagnosis of this group of patients diseases.

Chlorhexidine (CHX) is regarded as one of the most successful antiplaque agents in controlling the formation of dental biofilm. Nevertheless, molecular mechanisms of their effects in Streptococcus mutans are largely unknown. In this work, the effects of sub-lethal and lethal concentrations of chlorhexidine (CHX) on planktonic or biofilm-organized Streptococcus mutans cells were investigated in dose- and time-dependent manner. The Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) for planktonic cells and biofilm conditions were determined by standard methods. Quantitative PCR (qPCR) was used to quantify the relative levels of glucosyltransferase B (gtfB), gtfC and gtfD transcription of S. mutans in the presence of CHX. The CHX activity in the initial biofilm structure and morphological alterations in planktonic cells were examined by Scanning Electron Microscopy (SEM). The results indicate that CHX increased expression of gtfC and gtfD in planktonic S. mutans cells and CHX reduced the expression of gtfB, gtfC, and gtfD in biofilms. High concentrations of CHX resulted in several wilted S. mutans planktonic cells with spilled intracellular material, while decreased cells’ chain length and matrix was found when the initial biofilm was exposed to increasing concentrations of CHX. CHX’s effects against bacteria depend on the type of growth organization and the concentration and time of exposure to the drug. At sub-lethal concentrations, CHX affects the expression of glucosyltransferases, which may have anticariogenic effect.

Background： Beef cattle breeding programs in Brazil have placed greater emphasis on the genomic study of reproductive traits of males and females due to their economic importance. In this study, genome-wide associations were assessed for scrotal circumference at 210 d of age, scrotal circumference at 420 d of age, age at first calving, and age at second calving, in Canchim beef cattle. data quality control was conducted resulting in 672,778 SNPs and 392 ***： Associated SNPs were observed for scrotal circumference at 420 d of age（435 SNPs）, followed by scrotal circumference at 210 d of age（12 SNPs）, age at first calving（six SNPs）, and age at second calving（four SNPs）. We investigated whether significant SNPs were within genic or surrounding regions. Biological processes of genes were associated with immune system, multicellular organismal process, response to stimulus, apoptotic process, cellular component organization or biogenesis, biological adhesion, and ***： Few associations were observed for scrotal circumference at 210 d of age, age at first calving, and age at second calving, reinforcing their polygenic inheritance and the complexity of understanding the genetic architecture of reproductive traits. Finding many associations for scrotal circumference at 420 d of age in various regions of the Canchim genome also reveals the difficulty of targeting specific candidate genes that could act on fertility; nonetheless,the high linkage disequilibrium between loci herein estimated could aid to overcome this issue. Therefore, al relevant information about genomic regions influencing reproductive traits may contribute to target candidate genes for further investigation of causal mutations and aid in future genomic studies in Canchim cattle to improve the breeding program.

Fast Determination of Benzodiazepines in Human Urine via Liquid-Liquid Extraction with Low Temperature Partitioning and LC-HRMS

A simple and high-throughput method to simultaneously determine selected benzodiazepines (i.e., diazepam, lorazepam, clonazepam, and bromazepam) in urine was developed and validated. The entire methodology consisted of the application of an innovative extraction/cleanup procedure, namely liquid-liquid extraction with low-temperature partitioning (LLE-LTP), and analysis by liquid chromatography combined with high-resolution mass spectrometry (LC-HRMS). The LLE-LTP procedure was optimized via factorial design and by evaluating crucial variables, specifically the freezing mode (either slow or fast), the urine/acetonitrile volume ratio, and the sample ionic strength. The benzodiazepines were quantified using matrix-matched calibration curves where the following parameters were assessed by validation protocol: in general, linearity range of 17 - 200 μg?L–1 (r > 0.9957);limits of detection lower than 5 μg?L–1;relative standard deviations (RSD) lower than 12.5%;and accuracy ranging from 72.3 to 117%. To test this procedure’s performance, the method was applied to determine the content of diazepam in actual urine samples. The validation results obtained for the method demonstrated that the present methodology could be potentially applied in proficient laboratories as a routine approach for determining benzodiazepines compounds content in urine.
