Bacterial Sample Concentration and Culture Monitoring Using a PEG-Based Osmotic System with Inline Impedance and Voltammetry Measurements

Impedance measurements using graphite electrodes were used to detect the increase in culture medium conductivity due to bacteria growth in real time along with simultaneous voltammetric monitoring of pyocyanin *** methods were compared to conventional continuous monitoring of bacterial cultures using turbidity measurement at an optical density of 600 nm(OD600).A practical osmotic system was further designed for concentrating bacterial cultures during growth to enable earlier detection using the electrochemical *** cultures,starting from an initial culture density of 1×10^(8) cells/mL,were grown inside a sealed cellulose ester dialysis membrane,while polyethylene glycol in LB medium was used as the draw solution outside the membrane to gradually concentrate the growing cultures.0.7-mm-diameter graphite for mechanical pencils was utilized as working and counter electrodes with a platinum wire reference electrode for electrochemical measurements with and without the osmotic *** the absence of forward osmosis,impedance meas-urements detected culture growth~1 h faster than conventional *** osmosis showed a twofold decrease in the time to detect pyocyanin production as an indicator for bacterial *** impedance monitoring,removal of water by osmosis was conflated with the impedance decrease due to cell growth;however,the results show a promising ability to detect bacteria growth via an observed shift in osmotic impedance profile when bacteria are present in a *** monitoring the deviation in the impedance profile,a threefold improvement in detection time was achieved when compared to conventional OD600 measurements.

Introduction: Females with sickle cell disease (SCD), despite having a delayed pubertal development, are subject to many worries relating to their abilities to conceive, their capacity to maintain a maternofetal-risk-free pregnancy till term and give birth to healthy children without sickle cell disease. Knowing that unplanned pregnancies are more likely to increase maternofetal morbidity and mortality, we sought out to explore the pregnancy preferences in female patients with sickle cell disease to promote healthy conception and childbirth in this vulnerable population. Methodology: We conducted a cross-sectional study involving female patients of child-bearing age with sickle cell disease followed at Laquintinie Hospital Douala or who were members of a local sickle cell association. Pregnancy preferences were determined using the Desire to Avoid Pregnancy (DAP) scale. Factors associated with pregnancy preferences were determined using multivariable linear regression model. Threshold for significance was set at p Results: Seventy-seven patients were included with a mean age of 24.71 ± 5.53 years. Forty patients (51.95%) had one sexual partner and thirteen women (32.5%) used contraceptive methods. Most patients (46.0%) responded “Strongly Agree” or “Agree” to the statement “it would be a good thing for me if I became pregnant in the next 3 months”. The mean DAP score was 1.63 ± 0.91 [1.28 ± 0.82 in those who had a sexual partner and 1.99 ± 0.86 in those who had no sexual partner]. Factors that were positively associated with DAP score were students (b = 0.32, 95% CI [0.21, 0.95], p = 0.003), monthly income ≥ 100,000 FCFA (b = 0.24, 95% CI [0.07, 0.81], p = 0.022), and not having a sexual partner (b = 0.26, 95% CI [0.11, 0.85], p = 0.012). Conclusion: Most female patients with SCD have a low desire to avoid pregnancy. This is especially true for patients who are not students, have a partner and have a low monthly income.

This paper analyses the oxidation of ferrous iron via chemical and biological means in a class of Sequential Batch Reactors(SBR-type).For this,a kinetic model for the study of iron oxidation system is proposed,followed by a parametric sensitivity analysis and a bifurcation analysis,which allow selecting the most influential kinetic parameters in order to ensure a suitable prediction capacity of the mathematical *** system consists of two SBR bioreactors,the first being used to produce hydrogen peroxide(H2O2)that is fed to a second reactor where the iron oxidation is carried out by chemical–biological *** predictions were compared with experimental data for the production of H2O2 and for ferrous iron oxidation,finding suitable correlation coefficients(r2 N 0.98)for each state *** bifurcation analysis showed the trajectories of the main variables,such as,biomass,H2O2 and ferrous iron,under the change of the most influential kinetic *** analysis demonstrates the usefulness of the constructed model to predict the kinetic behaviour of the SBR-type process.

B cell development in bone marrow is a precisely regulated complex *** successive stages of differentiation,which are regulated by a multitude of signaling pathways and an array of lineage-specific transcription factors,the common lymphoid progenitors ultimately give rise to mature B *** to early thymocyte development in the thymus,early B cell development in bone marrow is critically dependent on IL-7 *** this IL-7-dependent stage of differentiation,several transcription factors,such as E2A,EBF1,and Pax5,among others,play indispensable roles in B lineage specification and *** recent studies have implicated several other transcription factors in B cell development,the role of NFATc1 in early B cell developmental stages is not ***,using multiple gene-manipulated mouse models and applying various experimental methods,we show that NFATc1 activity is vital for early B cell *** of NFATc1 activity in pro-B cells suppresses EBF1 expression,impairs immunoglobulin gene rearrangement,and thereby preBCR formation,resulting in defective B cell ***,deficiency in NFATc1 activity arrested the pro-B cell transition to the pre-B cell stage,leading to severe B cell *** findings suggest that,along with other transcription factors,NFATc1 is a critical component of the signaling mechanism that facilitates early B cell differentiation.

Changes in tree mortality due to severe drought can alter forest structure,composition,dynamics,ecosystem services,carbon fl uxes,and energy interactions between the atmosphere and land *** utilized long-term(2000‒2017,3 full inventory cycles)Forest Inventory and Analysis(FIA)data to examine tree mortality and biomass loss in drought-aff ected forests for East Texas,*** that experienced six or more years of droughts during those censuses were selected based on 12-month moderate drought severity[Standardized Precipitation Evaporation Index(SPEI)-1.0].Plots that experienced other disturbances and inconsistent records were excluded from the *** total,222 plots were retained from nearly 4000 *** nonlinear mixed models(GNMMs)were used to examine the changes in tree mortality and recruitment rates for selected *** results showed that tree mortality rates and biomass loss to mortality increased overall,and across tree sizes,dominant genera,height classes,and *** average mortality rate of 5.89%year−1 during the study period could be incited by water stress created by the regional prolonged and episodic drought *** overall plot and species-group level recruitment rates decreased during the study *** mortality showed mixed results regarding basal area and forest density using all plots together and when analyzed the plots by stand origin and *** mortality rates of smaller trees were detected and were likely compounded by densitydependent *** analysis of drought-induced tree mortality using hydro-meteorological data along with drought severity and length gradient is suggested to better understand the eff ects of drought on tree mortality and biomass loss around and beyond East Texas in the southeastern United States.

Conversion of laparoscopic colorectal resection for cancer: What is the impact on short-term outcomes and survival?

Laparoscopic resection for colon and rectal cancer is associated with quicker return of bowel function, reduced postoperative morbidity rates and shorter length of hospital stay compared to open surgery, with no differences in long-term survival. Conversion to open surgery is reported in up to 30% of patients enrolled in randomized control trials comparing open and laparoscopic colorectal resection for cancer. In this review, reasons for conversion are anatomical-related factors, disease-related-factors and surgeon-related factors. Body mass index, local tumour extension and co-morbidities are independent predictors of conversion. The current evidence has shown that patients with converted resection for colon cancer have similar outcomes compared to patients undergoing a laparoscopic completed or open resection. The few studies that have assessed the outcomes after conversion of laparoscopic rectal resection reported significantly higher rates of complications and longer length of hospital stay in converted patients compared to laparoscopically treated patients. No definitive conclusions can be drawn when converted and open rectal resections are compared. Early and pre-emptive conversion appears to have more favourable outcomes than reactive conversion; however, further large studies are needed to better define the optimal timing of conversion. With regard to long-term oncologic outcome, overall and disease-free survival in the case of conversion in laparoscopic colorectal cancer surgery seems to be worse than those achieved in patients in whom resection was successfully completed by laparoscopy. Although a worse long-term oncologic outcome has been suggested, it remains difficult to draw a proper conclusion due to the heterogeneity of the long-term outcomes as well as the inclusion of both colon and rectal cancer patients in most of the studies.
