Neutrophils are the most important circulating.phag.cytes. Circulating.mono-cytes and precursors of tissue macrophag.s also have the ability to phag.cytize. Pidotimod (ADIMODTM) exerts immunostimulatory and immunoreg.latory effects throug. the stimulation and reg.lation of cellular immune responses by lymphocytes Canine herpesvirus (CHV) mainly affect puppies between the first and second weeks of ag., causing.hig. morbidity in the litter. To date, there is only one commercial vaccine in Europe to prevent disease. In this work, inactivated CHV cultures were inoculated in rabbits, adsorbed and not adsorbed to chitosan nanoparticles. Phag.cytosis in the presence or absence of specific antibodies was measured. Response of virus neutralizing.antibodies was also evaluated. AdimodTM enhanced the nonspecific and specific phag.cytotic response. The association of the virus to the nanoparticles increased the phag.cytic ability of blood cells;however, AdimodTM alone had a g.eater effect on phag.cytic activity and g.nerated a strong.r immune response that corresponded to the increased phag.cytosis (p TM was used.

Antinuclear antibodies are found in animals suffering.from Systemic Lupus Erythematosus (SLE) and some other diseases, their presence in the blood is determined by antinuclear antibody (ANA) test using.indirect immunofluorescence (IF) with HEp2 cells as a substrate. In this work, an immunoperoxidase (IP) assay was developed to evaluate the ANAs in canine sera, using.canine kidney cell lines (MDCK) and compared with a commercial immunofluorescence test on Hep2 cells for this system, a fluoresceinated anti-canine Ig.antibody was standardized. The study was performed on 50 sera from dog. submitted to the laboratory with different clinical diag.oses of autoimmune-associated diseases. The procedures on both cells were unified to perform comparisons of the reactions, direct sera or at different dilutions were added to a monolayer of permeabilized MDCK cells, followed by a peroxidized anti-canine Ig. conjug.te, and a substrate for the IP reaction. The same sera were tested on the commercial IF assay on Hep2 cell system. In 22/50 cases, the presence of LE cells in peripheral blood was determined. A hig. correlation was found in the detection of antinuclear antibodies between both cell lines and techniques, however there were differences in the reaction patterns in the nucleus and cytoplasm between cell lines. The diffuse nuclear pattern observed in MDCK cells was more related to the presence of hig. percentag.s of LE cells in peripheral blood. The differences found in the results were possibly associated with the presence of homolog.us antig.ns between the MDCK cells and the dog. In addition, the methodolog. and standardization for the use and interpretation of a reference serum was developed to unify the interpretation criteria in the laboratory.

A characteristic common to herpesviruses is the ability to establish a latent infection in the hosts, a transcriptionally active reg.on has detected during.latency as well as a set of RNA that are known as Latency Associated Transcripts (LATs), their functions have been clarified in recent work. The present work was carried using.different bioinformatics method in order to determine if Herpesvirus Canine 1 (CHV-1) has a reg.on associated with latency. Our result was the selection of nine sequences candidate of micro RNA (miRNA) (MIREval 2.0 software), and 26 miRNA (miRNAFold v.1.0 software), of them, were selected 14 with real precursors of miRNA, two were found between the RL2 and RS1 g.nes, one in the RL2 g.ne and 11 in the RS1 g.ne. The results showed that the similarities of these reg.ons are very low among.the herpesviruses analyzed, so it was not possible to deduce the presence of the LAT g.ne in canine herpesvirus type 1 with bioinformatics. On the other hand, the comparison showed that the miRNA predicted: chv1-mir-mirnafold-8 has similarity with the ebv-mir-BART7-3p of Epstein-Barr Virus (EBV), in this way, the microRNAs predicted by means of bioinformatic prog.ams met the theoretical requirements of these molecules, however at not having.a deg.ee of preservation in other herpesviruses, the expression by CHV-1 in latency cannot be confirmed and it is necessary to identify throug. experimental tests.

An experimental inoculation of canine Herpesvirus (HVC), strain H17, was performed in rabbits previously and simultaneously treated with dexamethasone, the behavior of the leukocyte count was followed prior and during.the experimental phase. Upon death or euthanasia of the animals, a necropsy and evaluation of various org.ns was performed by histopatholog. and by nested PCR ag.inst the Herpesvirus Polymerase g.ne. All animals inoculated with dexamethasone showed leukopenia (p g.ificant histolog.cal lesions, but showed amplification of the Herpesvirus Polymerase g.ne in various org.ns despite not showing.clinical sig.s of the illness. A dolphin Herpesvirus isolate was used as a positive control as rabbits developed fatal systemic disease and lesions typical of active (lytic) infection in various org.ns within 72 hours post-inoculation. The absence of clinical sig.s, sig.ificant histolog.cal lesions, and the presence of viral DNA in some org.ns sug.ested a state of latency due to canine Herpesvirus. Dexamethasone allowed HVC infection, but did not promote viral reactivation in rabbits contrary to that observed in canines experimentally induced to the lytic cycle by HVC.

Canine herpesvirus (CHV-1) causes disease associated with hig. mortality in infect-ed puppies, which represents larg. financial losses for dog.breeders. Since CHV-1 at the time of the study he had not been reported in Mexico, the main objective of this study was to determine the prevalence of antibodies ag.inst CHV-1 in canine kennels in the metropolitan area of Mexico City. A commercial enzyme-linked immuno-sorbent assay (ELISA) was used, and the results were compared to those of a viral neutralization test. The ELISA kit uses the complete viral particle as the antig.n. The plaque reduction neutralization test was combined with the immunoperoxidase technique because of the low cytopathic effect of CHV-1. Neutralizing.antibodies were also detected in 20 randomly selected samples. The prevalence of CHV-1 with ELISA was 87%. The concordance between ELISA and serum neutralization (SN) was 0.1129, the sensitivity of the ELISA ag.inst SN was 1.0 (100%), the positive predic-tive value was 0.39 (39%), and the neg.tive predictive value was 1 (100%). These results show that ELISA is useful for monitoring.the dog.population for CHV-1;a positive test result requires confirmation with an SN test, and a neg.tive ELISA result indicates a hig. probability of being.SN-neg.tive. The only variables that were sta-tistically associated with CHV-1 prevalence were breed and kennel. A statistically sig.ificant relationship between the deg.ee of ELISA and SN titer was obtained, with a confidence level of 95%. None of the clinical presentation factors was statistically sig.ificant. These results sug.est that most of the canine population studied in Mex-ico is in a herpesvirus latency state.

This work presents the patholog. description, isolation and identification of canine herpesvirus (CHV-1) in Mexico, a virus that causes a g.neralized hemorrhag.c infection in puppies from the canidae family. Methods: Isolates were obtained from puppies that died within the first four weeks of life and had lesions consistent with canine herpesvirus. Results: The main g.oss lesions were petechial and ecchymotic hemorrhag.s in kidneys, liver and lung.;proliferative interstitial nephritis;multifocal necrosis in liver and kidneys;and encephalitis with intranuclear inclusion bodies. Herpesvirus was confirmed throug. direct immunofluorescence, electron microscopy and polymerase chain reaction for DNA polymerase and g.ycoprotein B g.nes. Discussion: Eig.t strains were isolated and identified as canine herpesvirus corresponding.to three of the working.cases with g.oss and microscopic lesions very similar to those described in the literature;then, isolates were confirmed by PCR g.ne amplification, positive reactions on immunofluorescence and observations from electron microscopy. This work represents the first report of this disease, including.g.oss and histolog.cal lesions, and confirmation by isolation and identification of the canine herpesvirus in Mexico.
