BACKgROUND Alpha-1 antitrypsin deficiency is a rare genetic disease and a leading cause of inherited alterations in plasma protein metabolism(APPM).AIM To understand the prevalence,burden and progression of liver disease in patients with APPM including alpha-1 antitrypsin *** We conducted a retrospective analysis of anonymized patient-level claims data from a german health insurance provider(AOK PLUS).The APPM cohort comprised patients with APPM(identified using the german Modification of the International Classification of Diseases-10th Revision[ICD-10-gM]code E88.0 between 01/01/2010-30/09/2020)and incident liver disease(ICD-10-gM codes K74,K70.2-3 and K71.7 between 01/01/2012-30/09/2020).The control cohort comprised patients without APPM but with incident liver *** were incidence/prevalence of liver disease in patients with APPM,demographics/baseline characteristics,diagnostic procedures,progression-free survival(PFS),disease progression and *** Overall,2680 and 26299 patients were included in the APPM(fibrosis,96;cirrhosis,2584)and control(fibrosis,1444;cirrhosis,24855)cohorts,*** 100000 individuals,annual incidence and prevalence of APPM and liver disease was 10-15 and 36-51,*** the APPM cohort,median survival was 4.7 years[95%confidence interval(CI):3.5-7.0]and 2.5 years(95%CI:2.3-2.8)in patients with fibrosis and cirrhosis,respectively.A higher proportion of patients in the APPM cohort experienced disease progression(92.0%)compared with the control cohort(67.2%).Median PFS was shorter in the APPM cohort(0.9 years,95%CI:0.7-1.1)compared with the control cohort(3.7 years,95%CI:3.6-3.8;Pger event-free survival for ascites,hepatic encephalopathy,hepatic failure and esophageal/gastric varices than patients with cirrhosis in the APPM cohort(Pger event-free survival for ascites,cir

Assessment of compliance to sulfadoxine-pyrimethamine 18 months after phasing out chloroquine in Mkuranga District,Coast region-Tanzania

Objective:To observe and assess the compliance to sulfadoxine-pyrimethamine(SP) one and a half years after phasing out chloroquine(CQ) in Mkuranga District,Coast region,Tanzania. Methods:A randomly controlled baseline community study was conducted in rural areas of Mkuranga district,***-structured questionnaire consisted of open-and closed -ended questions including home stocking,home use,last fever episodes and treatment of underfives with malaria using CQ or ***:The prevalence of fever or reported fever rate during the last 48 hours by their mothers or guardians was high(70%).Of all 117 blood samples,only 8 children after drug analysis were found to have CQ and 13 had SP concentrations within their blood *** of these blood drug levels were above therapeutic ranges. Conclusions:Community interventions are urgently needed in rural communities and should specifically target households nucleus on early malaria fever recognition and provision of recommended antimalarials for the sick underfive ***,sadly,there was an increase in underweight and undernourishment in the study areas,probably because of malaria in the area and poverty which are associated with poor nutrition in these youngsters.

Huanglongbing(HLB)is the most devastating disease for citrus *** Liberibacter asiaticus(C Las),vectored by Asian citrus psyllid(ACP,Diaphorina citri Kuwayama),is the most common pathogen causing the *** citrus varieties are highly susceptible to HLB,whereas trifoliate orange(Poncirus trifoliata)is considered highly tolerant to *** F1 segregating population and their parent trifoliate orange and sweet orange,which had been exposed to intense HLB pressure for three years,was evaluated for disease symptoms,ACP colonization,C Las titer and tree vigor repeatedly for two to three *** orange and sweet orange showed significant differences for most of the phenotypic traits,and the F1 population exhibited a large variation.A high-density SNP-based genetic map with 1402 markers was constructed for trifoliate orange,which exhibited high synteny and high coverage of its reference genome.A total of 26 quantitative trait locus(QTLs)were identified in four linkage groups Lg-t6,Lg-t7,Lg-t8 and Lg-t9,of which four QTL clusters exhibit a clear co-localization of QTLs associated with different *** genome-wide analysis of gene expression in response to C Las infection in‘Flying Dragon’and‘Larger-Flower DPI-50-7’trifoliate orange,85 differentially expressed genes were found located within the QTL *** them,seven genes were classified as defense or immunity protein which exhibited the highest transcriptional change after C Las *** results indicate a quantitative genetic nature of HLB tolerance and identified candidate genes that should be valuable for searching for genetic solutions to HLB through breeding or genetic engineering.

Determination of selected semi-volatile organic compounds in water using automated online solid-phase extraction with large-volume injection/gas chromatography/mass spectrometry

A rapid,sensitive,and cost-effective analyticalmethod was developed for the analysis of selected semivolatileorganic compounds in *** method used anautomated online solid-phase extraction technique coupledwith programmed-temperature vaporization large-volumeinjection gas chromatography/mass *** samples were extracted by using a fully automatedmobile rack system based on x-y-z robotic techniquesusing syringes and disposable 96-well extraction *** method was validated for the analysis of 30 semivolatileanalytes in drinking water,groundwater,andsurface *** a sample volume of 10 mL,the linearcalibrations ranged from 0.01 or 0.05 to 2.5μg·L^(-1),and themethod detection limits were less than 0.1μg·L^(-1).For thereagent water samples fortified at 1.0μg·L^(-1)and2.0μg·L^(-1),the obtained mean absolute recoveries were70%-130%with relative standard deviations of less than20%for most *** the drinking water,groundwater,and surface water samples fortified at 1.0μg·L^(-1),theobtained mean absolute recoveries were 50%-130%withrelative standard deviations of less than 20%for *** new method demonstrated three advantages:1)no manipulation except the fortification of surrogatestandards prior to extraction;2)significant cost reductionassociated with sample collection,shipping,storage,andpreparation;and 3)reduced exposure to hazardous solventsand other *** a result,this new automatedmethod can be used as an effective approach for screeningand/or compliance monitoring of selected semi-volatileorganic compounds in water.

Comparative transcriptome analysis during early fruit development between three seedy citrus genotypes and their seedless mutants

Identification of genes with differential transcript abundance(gDTA)in seedless mutants may enhance understanding of seedless citrus *** analysis was conducted at three time points during early fruit development(Phase 1)of three seedy citrus genotypes:Fallglo(Bower citrus hybrid(Citrus reticulata×***×***)×Temple(***×***)),grapefruit(***),Pineapple sweet orange(***),and their seedless *** abortion in seedless mutants was observed at 26 days post anthesis(Time point 2).Affymetrix transcriptomic analysis revealed 359 to 1077 probe sets with differential transcript abundance in the comparison of seedless versus seedy fruits for each citrus genotypes and time *** gDTA identified by 18 microarray probe sets were validated by *** clustering analysis revealed a range of gDTA associated with development,hormone and protein metabolism,all of which may reflect genes associated with seedless fruit *** were 14,9 and 12 genes found exhibiting similar abundance ratios in all three seedless versus seedy genotype comparisons at time point 1,2 and 3,*** those genes were genes coding for an aspartic protease and a cysteine protease,which may play important roles in seedless fruit *** insights into seedless citrus fruit development may contribute to biotech approaches to create seedless cultivars.

Huanglongbing(HLB)is a disease that has devastated the Florida citrus industry,threatens the entire *** industry,and globally is rapidly ***’s citrus production is 90%sweet orange,which is quite sensitive to *** heavy reliance on sweet orange for Florida citrus production makes the industry especially vulnerable to diseases that are damaging to this type of ***,90%of Florida oranges are used in producing orange juice that is defined by a federal regulation known as the“orange juice standard”,specifying that at least 90%of“orange juice”must be derived from Citrus *** analyses definitively reveal that sweet orange is not a true species,but just one of many introgression hybrids of *** and ***,with phenotypic diversity resulting from accumulated mutations in this single hybrid,the“sweet orange”.No other fruit industry is limited by law to such a narrow genetic ***,there are new citrus hybrids displaying reduced sensitivity to HLB,and in some cases they produce juice,alone or in blends,that consumers would recognize as“orange juice”.Reconsidering current regulations on orange juice standards may permit use of such hybrids in“orange juice”,providing greater latitude for commercialization of these hybrids,leading to higher-quality orange juice and a more sustainable Florida orange juice industry.

A Reflection of Minimum Adequate education in South Carolina More Than Fifty Years Later

The issues and challenges of minimum adequate public school education in the State of South of South Carolina has been a struggle between lawmakers and taxpayers for more than fifty years. The enactment of the Civil Rights Act of 1964 led to forced school integration, busing, and led to whites＇ physical moving out of selected school districts. Selected public schools filed a lawsuit against the State of South Carolina to the Supreme Court with the claim of failing to provide adequate and equal education to all children in the state. This reflection focuses on the Abbeville case in context, funding issues, and inadequate facilities, which concludes with a reflective view of the Brown v. The Board of edueation 1954 decision.

<i>Agrobacterium</i>-Mediated Transformation of Mexican Lime (<i>Citrus aurantifolia</i>Swingle) Using Optimized Systems for Epicotyls and Cotyledons

Transgenic Mexican lime (Citrus aurantifolia Swingle) was produced through two explant sources, each using systems previously optimized for each source. One used epicotyls segments, which was the predominant explant for transgenic Citrus production following co-cultivation with Agrobacterium, and has a well-established protocol. The other procedure used embryo cotyledons from mature seeds, which was developed in our lab as an alternative for stable Citrus transformation. Cotyledon transformation and regeneration protocols were optimized by comparing variables in culture medium composition on shoot regeneration and four parameters in transient transformation. The optimized protocols were compared, and frequency of regeneration, frequency of transgenic plant-recovery and stable transformation efficiency indicated the superiority of the cotyledon protocol for Agrobacterium-mediated genetic transformation in Mexican lime. The tissue choice resulted in marked improvement in shoot regeneration (14.1% of explants producing shoots in epicotyls;55.8% in cotyledons), stable transformation frequency (11.4% of epicotyls explants;40.2% in cotyledons), and frequency of transgenic plant-recovery (37.9% in epicotyl explants;92.6% in cotyledons). Thus, easy availability of explants using embryo cotyledons from mature seeds, technical simplicity, shortening of transformation time-course, and higher transformation and regeneration frequencies makes this new system an attractive alternative over the previously published Citrus transformation protocols. In the course of this project, we generated Mexican lime with a Recombinase Mediated Exchange Cassette landing pad, which was designed for stacking transgenes.

Bioadsorption of 4-Chlorophenol to the Activated Sludge

The adsorption behaviour of 4 chlorophenol from aqueous solution to activated sludge was quantitatively characterized in this paper. The effects of the initial pH values, initial chlorophenol concentration and adsorbent dosage on bioadsorption were investigated. The maximum adsorption capacity was found to be 110.5 mg/g at 100 mg/L initial concentration. The Freundlich and Langmuir adsorption isotherms were applied to describe the biosorption processes and the isotherm constants were evaluated. 
