the secondary injury cascade after spinal cord injury: an analysis of local cytokine/chemokine regulation

After spinal cord injury, there is an extensive infiltration of immune cells, which exacerbates the injury and leads to further neural degeneration. therefore, a major aim of current research involves targeting the immune response as a treatment for spinal cord injury. Although much research has been performed analyzing the complex inflammatory process following spinal cord injury, there remain major discrepancies within previous literature regarding the timeline of local cytokine regulation. the objectives of this study were to establish an overview of the timeline of cytokine regulation for 2 weeks after spinal cord injury, identify sexual dimorphisms in terms of cytokine levels, and determine local cytokines that significantly change based on the severity of spinal cord injury. Rats were inflicted with either a mild contusion, moderate contusion, severe contusion, or complete transection, 7 mm of spinal cord centered on the injury was harvested at varying times post-injury, and tissue homogenates were analyzed with a Cytokine/Chemokine 27-Plex assay. Results demonstrated pro-inflammatory cytokines including tumor necrosis factor α, interleukin-1β, and interleukin-6 were all upregulated after spinal cord injury, but returned to uninjured levels within approximately 24 hours post-injury, while chemokines including monocyte chemoattractant protein-1 remained upregulated for days post-injury. In contrast, several anti-inflammatory cytokines and growth factors including interleukin-10 and vascular endothelial growth factor were downregulated by 7 days post-injury. After spinal cord injury, tissue inhibitor of metalloproteinase-1, which specifically affects astrocytes involved in glial scar development, increased more than all other cytokines tested, reaching 26.9-fold higher than uninjured rats. EP Aquarii:A New Picture of the Circumstellar Envelope

New analyses of earlier ALmA observations of oxygen-rich AGB star EP Aquarii are presented,which complete a previously published analysis and offer a different interpretation of the morpho-kinematics of the circumstellar *** birth of the equatorial density enhancement(EDE)is shown to occur very close to the star where evidence for rotation has been *** to the star and where outflows have been observed:their interaction with the gas of the nascent EDE is seen to play an important role in the development of the wind and the evolution of its radial velocity from 8 to 10 km s^(-1) on the polar symmetry axis to~2 km s^(-1) at the *** implies complex morpho-kinematics:making reliable interpretations with reasonable confidence is *** particular,it questions an earlier interpretation implying the presence of a white dwarf companion orbiting the star at an angular distance of -0″4 from its *** proposes instead an interpretation in terms of a standard mass ejection associated with a shock wave leaving a void of emission in its *** Doppler velocity wings are seen to consist of two components,the upper velocity end of the global wind,reaching above±12 km s^(-1),and an effective line broadening,confined within 200 mas from the center of the star,reaching above±20 km s^(-1) and interpreted as caused by the pattern of shock waves resulting from the interaction between stellar pulsation and convective cell partition.

Perovskite solar cells(PSCs)have made great advances in terms of power conversion efficiency(PCE),yet their subpar stability continues to hinder their *** interface between the perovskite layer and the charge-carrier transporting layers plays a crucial role in undermining the stability of *** this work,we propose a strategy to stabilize high-performance PSCs with PCE over 23%by introducing a cesium-doped graphene oxide(GO-Cs)as an interlayer between the perovskite and hole-transporting *** GO-Cs treated PSCs exhibit excellent operational stability with a projected t80(the time where the device PCE reduces to 80%of its initial value)of 2143 h of operation at the maximum powering point under one sun illumination.

Frequency-dependent viscoelasticity effects on the wave attenuation performance of multi-layered periodic foundations

In this paper,layered periodic foundations(LPFs)are numerically examined for their responses to longitudinal and transverse modes in the time and frequency *** different unit-cells,i.e.,2-layer,4-layer,and 6-layer unit-cells,comprising concrete/rubber,concrete/rubber/steel/rubber,and concrete/rubber/steel/rubber/lead/rubber materials,respectively,are taken into ***,the viscoelasticity behavior of the rubber is modeled with two factors,i.e.,a frequency-independent(FI)loss factor and a linear frequency-dependent(FD)loss *** the extraction of the complex dispersion curves and the identification of the band gaps(BGs),the simulations of wave transmission in the time and frequency domains are performed using the COmSOL *** parametric studies evaluate the effects of the rubber viscoelasticity models on the dispersion curves and the wave transmission for the longitudinal and transverse *** results show that considering the rubber viscoelasticity enhances the wave attenuation ***,the transverse-mode damping is more sensitive to the viscoelasticity model than its longitudinal *** 6-layer unit-cell LPF exhibits the lowest BG,ranging from 4.8 Hz to 6.5 Hz.

Basis for the diversity and extent in loss of digestible nutrients created by dietary phytin: Emphasis on fowl and swine

Phytin is the Ca2+-mg2+-K+salt of phytic acid that is created and deposited in the aleurone layer and/or germ of grains and legumes. Its high presence in feedstuffs for fowl and swine diets results in it being a universal and significant impediment to optimum performance. Phytin impairs gastrointestinal recovery of a wide array of nutrients, the effect varying with the nutrient concerned. On exposure to low pH during gastric digestion, phytin dissociates into phytic acid and solubilized Ca2+. Even at low gastric pH,phytic acid is negatively charged which forms the basis of its anti-nutritive behavior. Pepsinogen has extensive basic amino acids on its activation peptide that are presented as cations at low pH which are targeted by pepsin for activation. Partially crystalized Ca2+near the enzyme?s active site further stabilizes its newly formed structure. thus, phytic acid appears to interfere with gastric digestion by several mechanisms; interfering with pepsinogen activation by binding to the polypeptide?s basic amino acids;coordinating free Ca2+, destabilizing pepsin; binding some dietary proteins directly, further compromising gastric proteolysis. Upon digesta attaining neutrality in the duodenum, Ca2+and other cations rebind with accessible anions, phytic acid being a significant contender. Phytate not only binds free cations but can also strip them from enzymes(e.g. Ca2+, Zn2+) which reduces their structural resistance to autolysis and ability as co-factors(e.g. Zn2+) to increase enzyme activity. Goblet cells initially employ Ca2+as an electronic shield between mucin layers enabling granule formation and cell storage. After mucin granule release, Ca2+is progressively displaced by Na+to free the viscous mucins enabling its translocation. mucin entangles with the glycocalyx of adjacent enterocytes thereby constructing the unstirred water layer（USWL）. Excessive removal of Ca2+from mucin by phytic acid increases its fluidity facilitating its loss from the USWL with

the bumpy road of purinergic inhibitors to clinical application in immune-mediated diseases

Purinergic signaling plays important roles throughout the body in the regulation of organ functions during and following the disruption of homeostasis. this is also reflected by the widespread expression of two families of purinergic receptors(P1 and P2) with numerous subtypes. In the last few decades, there has been increasing evidence that purinergic signaling plays an important role in the regulation of immune functions. mainly, signals mediated by P2 receptors have been shown to contribute to immune system-mediated pathologies. thus, interference with P2 receptors may be a promising strategy for the modulation of immune responses. Although only a few clinical studies have been conducted in isolated entities with limited success, preclinical work suggests that the use of P2 receptor inhibitors may bear some promise in various autoimmune diseases. Despite the association of P2 receptors with several disorders from this field, the use of P2 receptor antagonists in clinical therapy is still very scarce. In this narrative review, we briefly review the involvement of the purinergic system in immunological responses and clinical studies on the effect of purinergic inhibition on autoimmune processes. We then open the aperture a bit and show some preclinical studies demonstrating a potential effect of purinergic blockade on autoimmune events. Using suramin, a non-specific purinergic inhibitor, as an example, we further show that off-target effects could be responsible for observed effects in immunological settings, which may have interesting implications. Overall, we believe that it is worthwhile to further investigate this hitherto underexplored area.

Effects of layer interactions on instantaneous stability of finite Stokes flows

the stability analysis of a finite Stokes layer is of practical importance in flow control. In the present work, the instantaneous stability of a finite Stokes layer with layer interactions is studied via a linear stability analysis of the frozen phases of the base flow. the oscillations of two plates can have different velocity amplitudes, initial phases, and frequencies. the effects of the Stokes-layer interactions on the stability when two plates oscillate synchronously are analyzed. the growth rates of two most unstable modes when δ most equal, and δ = δ*/h*, where δ*and h*are the Stokes-layer thickness and the half height of the channel, respectively. However, their vorticities are different. the vorticity of the most unstable mode is symmetric, while the other is asymmetric. the Stokes-layer interactions have a destabilizing effect on the most unstable mode when δ tabilizing effect when δ > 0.68. However, the interactions always have a stabilizing effect on the other unstable mode. It is explained that one of the two unstable modes has much higher dissipation than the other one when the Stokes-layer interactions are strong. We also find that the stability of the Stokes layer is closely related to the inflectional points of the base-flow velocity profile. the effects of inconsistent velocity-amplitude, initial phase, and frequency of the oscillations on the stability are analyzed. the energy of the most unstable eigenvector is mainly distributed near the plate of higher velocity amplitude or higher oscillation frequency. the effects of the initial phase difference are complicated because the base-flow velocity is extremely sensitive to the initial phase.
