Eb.la virus （Eb.V） is the causative agent of a severe hemorrhagic fever disease associ- ated with high mortality rates in humans. This virus has five strains of which Zaire Eb.la virus （ZEb.V） is the first and most important strain. It can b. transmitted through contact with contaminated surfaces and ob.ects. The genome of Eb.V codes one non- structural and seven structural proteins consisting of two forms of glycoprotein （GP）： solub.e glycoprotein （sGP） and GP （spike）. In this paper, we attempted to characterize and predict physicochemical properties, b.cell epitopes, mutation sites, phosphorylation sites, glycosylation sites, and different protein structures of Eb.V GP to provide com- prehensive data ab.ut changes of this GP during a 40-years course （1976-2015）. GP sequences were ob.ained from NCb. gene b.nk from 1976-2015. Expasy＇sProtParam, PROTSCALE, immuneepitope, b.pipred, b.ePred, Ab.pred, VaxiJen, DISPHOS, Net- Phos, and numerous programs were used to predict and analyze all sequences. More variety of mutations were found in 2015 sequences and mutations were related to huge changes in b.cell epitopes, phosphorylation and glycosylation sites. In addition, our results determined different sites of disulfide b.nds and an important mutation related to IgE epitope as well as four potent b.cell epitopes （380-387, 318-338, 405-438 and 434-475）. In this study, we suggested the effect of mutations on GP properties, six posi- tions for disulfide b.nds and four phosphorylation sites for protein kinase C enzyme. Selected sequences were shown different sites for O-link and N-link glycosyl^tion. A mutation that changed GP to an allergen protein and has a significant role in vaccine designing as well as four potent b.cell epitopes in GP protein were found.

Inflammatory b.wel disease(Ib.)refers to a group of disorders characterized b. chronic inflammation of the gastrointestinal(GI)*** elevated levels of nitric oxide(NO)in serum and affected tissues;mainly synthesized b. the inducib.e nitric oxide synthase(iNOS)enzyme;can exacerb.te GI inflammation and is one of the major b.omarkers of GI *** natural and synthetic agents are ab.e to ameliorate GI inflammation and decrease iNOS expression to the extent comparab.e with some Ib. ***,the purpose of this study was to gather a list of natural or synthetic mediators capab.e of modulating Ib. through the NO *** datab.ses including Google Scholar and Pub.ed were searched from 1980 to May *** found that polyphenols and particularly flavonoids are ab.e to markedly attenuate NO production and iNOS expression through the nuclear factorκb.NF-κb.and JAK/STAT signaling *** and prob.otics can also alter the GI microb.ota and reduce NO expression in Ib. models through a b.oad array of mechanisms.A numb.r of synthetic molecules have b.en found to suppress NO expression either dependent on the NF-κb.signaling pathway(i.e.,dexamethasone,pioglitazone,tropisetron)or independent from this pathway(i.e.,nicotine,prednisolone,celecoxib.β-adrenoceptor antagonists).Co-administration of natural and synthetic agents can affect the tissue level of NO and may improve Ib. symptoms mainly b. modulating the Toll like receptor-4 and NF-κb.signaling pathways.
