BACkGROUND The high prevalence and persistence of Helicobacter pylori(H. pylori) infection, as well as the diversity of pathologies related to it, suggest that the virulence factors used by this microorganism are varied. Moreover, as its proteome contains 340hypothetical proteins, it is important to investigate them to completely understand the mechanisms of its virulence and survival. We have previously reported that the hypothetical protein HP0953 is overexpressed during the first hours of adhesion to inert surfaces, under stress conditions, suggesting its role in the environmental survival of this bacterium and perhaps as a virulence *** To investigate the expression and localization of HP0953 during adhesion to an inert surface and against gastric(AGS) *** expression analysis was performed for HP0953 during H. pylori adhesion. HP0953 expression at 0,3, 12, 24, and 48 h was evaluated and compared using the kruskal-Wallis equality-of-populations rank test. Recombinant protein was produced and used to obtain polyclonal antibodies for immunolocalization. Immunogold technique was performed on bacterial sections during adherence to inert surfaces and AGS cells, which was analyzed by transmission electron microscopy. HP0953 protein sequence was analyzed to predict the presence of a signal peptide and transmembrane helices, both provided by the exPASy platform, and using the GLYCOPP platform for glycosylation sites. Different programs, via, I-TASSeR, RaptorX, and HHalign-kbest, were used to perform three-dimensional *** HP0953 exhibited its maximum expression at 12 h of infection in gastric epithelium *** technique revealed HP0953 localization in the cytoplasm and accumulation in some peripheral areas of the bacterial body, with greater expression when it is close to AGS *** analysis revealed the presence of a signal peptide that interacts with the transmembrane region and then allows the release of the protein

Influence of surface roughness on the color of dental-resin composites

This study deals with the influence of surface roughness on the color of resin *** resin composites (microfilled,hybrid,and microhybrid) were each polished with 500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC *** roughness parameter (Ra) was measured using a Plμ confocal microscope,and field-emission scanning electron microscope (Fe-SeM) images were used to investigate filler *** was measured using a spectroradiometer and a D65 standard illuminant (geometry diffuse/0° specular component excluded (SCe) mode).Surface roughness decreased with grit number and was not influenced by filler size or size distribution.A significant influence of Ra on lightness (L*) was *** increased with decreases in roughness,except for specimens that underwent polishing procedure 4 (PP4;500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC papers consecutively).Generally,it was found that surface roughness influenced the color of resin *** composites that underwent PP1 (500-grit SiC paper) exhibited significant differences in chroma (C*),hue (h°),and lightness (L*) compared to composites that underwent PP3 (500-grit,1 200-grit,and 2 000-grit SiC papers consecutively) and *** difference (e*) between the polishing procedures was within acceptability thresholds in dentistry.

Increasing the detail of european land use/cover data by combining heterogeneous data sets

Data on land use and land cover(LULC)are a vital input for policy-relevant research,such as modelling of the human population,socioeconomic activities,transportation,environment,and their *** europe,CORINe Land Cover has been the only data set covering the entire continent consistently,but with rather limited spatial *** data sets have provided much better detail,but either have covered only a fraction of europe(*** Atlas)or have been thematically restricted(*** High Resolution Layers).In this study,we processed and combined diverse LULC data to create a harmonised,ready-to-use map covering 41 *** doing so,we increased the spatial detail(from 25 to one hectare)and the thematic detail(by seven additional LULC classes)compared to the CORINe Land ***,we decomposed the class‘Industrial and commercial units’into‘Production facilities’,‘Commercial/service facilities’and‘Public facilities’using machine learning to exploit a large database of points of *** overall accuracy of this thematic breakdown was 74%,despite the confusion between the production and commercial land uses,often attributable to noisy training data or mixed land *** learnt from this exercise are discussed,and further research direction is proposed.

efficacy of medium-chain fatty acid salts distilled from coconut oil against two enteric pathogen challenges in weanling piglets

Background:The search for alternatives to antibiotics in pig production has increased the interest in natural resources with antimicrobial properties,such as medium-chain fatty acids(MCFA)as in-feed *** study evaluated the potential of a novel blend of MCFA salts(DIC)from distilled coconut oil with a lauric acid content to reduce enteropathogens and control intestinal diseases around *** experimental disease models were implemented in early-weaned piglets,consisting of two oral challenges:Salmonella Typhimurium(1.2×10~8 CFU)or enterotoxigenic escherichia coli(eTeC)F4(1.5×10~9 CFU).The parameters assessed were:animal performance,clinical signs,pathogen excretion,intestinal fermentation,immune-inflammatory response,and intestinal ***:The Salmonella challenge promoted an acute course of diarrhea,with most of the parameters responding to the challenge,whereas the eTeC F4 challenge promoted a mild clinical course.A consistent antipathogenic effect of DIC was observed in both trials in the hindgut,with reductions in Salmonella *** counts in the cecum(P=0.03)on d 8 post-inoculation(PI)(Salmonella trial),and of enterobacteria and total coliform counts in the ileum and colon(PeTeC F4 trial).When analyzing the entire colonic microbiota(16 S rRNA gene sequencing),this additive tended(P=0.13)to reduce the Firmicutes/Bacteroidetes ratio and enriched Fibrobacteres after the Salmonella *** the eTeC F4 challenge,DIC prompted structural changes in the ecosystem with increases in Dialister,and a trend(P=0.14)to increase the Veillonellaceae *** parameters such as the intestinal fermentation products or serum pro-inflammatory mediators were not modified by DIC supplementation,nor were the histological *** the intraepithelial lymphocyte(IeL)counts were lowered by DIC in animals challenged with Salmonella(P=0.07).With eTeC F4,the IeL counts were higher with DIC on d 8 PI(P=0.08).Conclusions:This study c

This study was conducted to evaluate the effects of dietary free fatty acid(FFA)content and degree of fat saturation on production performance,lipid and calcium digestibility,and intestinal function of laying *** a 15-week period,a total of 144 laying hens(19 weeks old)were randomly assigned to 8 dietary treatments,which were obtained by gradually replacing crude soybean oil with soybean acid oil(AO),or crude palm oil with palm fatty acid distillate(FAD).Thus,there were 4 soybean and 4 palm diets with 6%added fat varying in their FFA percentage(10%,20%,30%,and 45%),following a 2×4 factorial *** treatment included 6 replicates with 3 birds per *** daily feed intake and final body weight were significantly higher in palm diets(Pe no differences were found in egg mass and feed conversion *** levels of FFA in soybean diets resulted in lower egg production and higher egg weight(linear,Pegarding the degree of fat saturation,hens fed soybean diets presented higher digestibility of ether extract(ee),fatty acids,and calcium than palm diets(Pe dietary FFA percentage negatively affected the digestibility of ee and calcium(Pe having little effect on FA *** was a significant interaction in the AMe;lower values were reported in soybean diets as the dietary FFA percentage increased(linear,Pereas palm diets remained *** experimental diets had little effect on gastrointestinal weight and ***,the jejunum of soybean diets showed higher villus height and higher villus height-to-crypt depth ratio than palm diets(Pe dietary FFA percentage increased the crypt depth and decreased the villus height-to-crypt depth ratio(linear,Ped that varying dietary FFA content did not affect fat utilization as much as the degree of saturation did,supporting the use of AO and FAD as alternative fat ingredients.

Inflammatory status in human hepatic cirrhosis

This review focuses on new findings about the inflammatory status involved in the development of human liver cirrhosis induced by the two main causes, hepatitis C virus(HCV) infection and chronic alcohol abuse, avoiding results obtained from animal models. When liver is faced to a persistent and/or intense local damage the maintained inflammatory response gives rise to a progressive replacement of normal hepatic tissue by non-functional fibrotic scar. The imbalance between tissue regeneration and fibrosis will determine the outcome toward health recovery or hepatic cirrhosis. In all cases progression toward liver cirrhosis is caused by a dysregulation of mechanisms that govern the balance between activation/homeostasis of the immune system. Detecting differences between the inflammatory status in HCV-induced vs alcoholinduced cirrhosis could be useful to identify specific targets for preventive and therapeutic intervention in each case. Thus, although survival of patients with alcoholic cirrhosis seems to be similar to that of patients with HCV-related cirrhosis(HCV-C), there are important differences in the altered cellular and molecular mechanisms implicated in the progression toward human liver cirrhosis. The predominant features of HCV-C are more related with those that allow viral evasion of the immune defenses, especially although not exclusively, inhibition of interferons secretion, natural killer cells activation and T cell-mediated cytotoxicity. On the contrary, the inflammatory status of alcohol-induced cirrhosis is determined by the combined effect of direct hepatotoxicity of ethanol metabolites and increases of the intestinal permeability, allowing bacteria and bacterial products translocation, into the portal circulation, mesenteric lymph nodes and peritoneal cavity. This phenomenon generates a stronger pro-inflammatory response compared withHCV-related cirrhosis. Hence, therapeutic intervention in HCV-related cirrhosis must be mainly focused to counter
