Measuring NO Production by Plant Tissues and Suspension Cultured Cells

We describe an inexpensive and reliable detector for measuring NO emitted in the gas phase from plants. The method relies on the use of a strong oxidizer to convert NO to NO2 and subsequent capture of NO2 by a Griess reagent trap. The set-up approaches the sensitivity for NO comparable to that of instruments based on chemiluminescence and photoacoustic detectors. We demonstrate the utility of our set-up by measuring NO produced by a variety of well established plant sources. NO produced by nitrate reductase （NR） in tobacco leaves and barley aleurone was readily detected, as was the production of NO from nitrite by the incubation medium of barley aleurone. Arabidopsis mutants that overproduce NO or lack NO-synthase （AtNOS1） also displayed the expected NO synthesis phenotype when assayed by our set-up. We could also measure NO production from elicitor-treated suspension cultured cells using this set-up. Further, we have focused on the detection of NO by a widely used fluorescent probe 4-amino-5-methylamino-2＇,7＇-difluorofluorescein （DAF-FM）. Our work points to the pitfalls that must be avoided when using DAF-FM to detect the production of NO by plant tissues. In addition to the dramatic effects that pH can have on fluorescence from DAF-FM, the widely used NO scavengers 2-phenyl- 4,4,5, 5-tetramethylimidazoline- 1-oxyl-3-oxide （PTIO） oxide （cPTIO） can produce anomalous and unexpected to bind to dead cells and remain NO-sensitive. and 2-（4-carboxyphenyl）-4,4,5,5-tetramethylimidazoline- 1-oxyl-3- results. Perhaps the most serious drawback of DAF-FM is its ability to bind to dead cells and remain NO-sensitive.

Predicting the mode of flow in pneumatic conveying systems—A review

An initial prediction of the particulate mode of flow in pneumatic conveying systems is beneficial as this knowledge can provide clearer direction to the pneumatic conveying design process. There are three general categories of modes of flow, two dense flows： fluidised dense phase and plug flow, and dilute phase oniy. Detailed in this paper is a review of the commonly used and available techniques for predicting mode of flow. Two types of predictive charts were defined： basic particle parameter based （e.g. particle size and density） and air-particle parameter based （e.g. permeability and de-aeration）. The basic particle techniques were found to have strong and weak areas of predictive ability, on the basis of a comparison with data from materials with known mode of flow capability. It was found that there was only slight improvement in predictive ability when the particle density was replaced by loose-poured bulk density in the basic parameter techniques. The air-particle-parameter-based techniques also showed well-defined regions for mode of flow prediction though the data set used was smaller than that for the basic techniques. Also, it was found to be difficult to utilise de-aeration values from different researchers and subsequently, an air-particle-based technique was developed which does not require any de-aeration parameter in its assessment.

Recording neural signals from delicate autonomic nerves is a challenging task that requires the development of a lowinvasive neural interface with highly selective,micrometer-sized *** paper reports on the development of a three-dimensional(3D)protruding thin-film microelectrode array(MeA),which is intended to be used for recording low-amplitude neural signals from pelvic nervous structures by penetrating the nerves transversely to reduce the distance to the *** gold pillars(O 20 or 50μm,~60μm height)were fabricated on a micromachined polyimide substrate in an electroplating *** sidewalls were insulated with parylene C,and their tips were optionally modified by wet etching and/or the application of a titanium nitride(TiN)*** microelectrodes modified by these combined techniques exhibited low impedances(~7 kΩ at 1 kHz for ∅ 50μm microelectrode with the exposed surface area of~5000μm^(2))and low intrinsic noise *** functionalities were evaluated in an ex vivo pilot study with mouse retinae,in which spontaneous neuronal spikes were recorded with amplitudes of up to 66μ*** novel process strategy for fabricating flexible,3D neural interfaces with low-impedance microelectrodes has the potential to selectively record neural signals from not only delicate structures such as retinal cells but also autonomic nerves with improved signal quality to study neural circuits and develop stimulation strategies in bioelectronic medicine,e.g.,for the control of vital digestive functions.

Investigations are reported into the use of an electromagnetically convoluted arc,external to a magnetic field(B-field)producing coil,in combination with a parallel R,L,C resonant circuit for interrupting quasi-steady *** order to elucidate the complex interactions between the arc,B-field and R,L,C circuit,the B-field producing coil is energised independently from the current to be interrupted and the R,L,C *** results are presented for the time variation of the currents flowing through the arc gap,the B-field coil and the parallel R,L,C circuit,along with the voltage across the arc *** insight is gained into the role of various effects,which are produced by the complex interactions and which might be used to advantage for direct current interruption.

The Soft X-ray Imager(SXI)is part of the scientific payload of the Solar wind Magnetosphere Ionosphere Link explorer(SMILe)*** is a joint science mission between the european Space Agency(eSA)and the Chinese Academy of Sciences(CAS)and is due for launch in *** is a compact X-ray telescope with a wide field-of-view(FOV)capable of encompassing large portions of earth’s magnetosphere from the vantage point of the SMILe *** is sensitive to the soft X-rays produced by the Solar Wind Charge eXchange(SWCX)process produced when heavy ions of solar wind origin interact with neutral particles in earth’s *** provides a mechanism for boundary detection within the magnetosphere,such as the position of earth’s magnetopause,because the solar wind heavy ions have a very low density in regions of closed magnetic field *** sensitivity of the SXI is such that it can potentially track movements of the magnetopause on timescales of a few minutes and the orbit of SMILe will enable such movements to be tracked for segments lasting many *** is led by the University of Leicester in the United Kingdom(UK)with collaborating organisations on hardware,software and science support within the UK,europe,China and the United States.

Asymptotic and numerical study of a surface breaking crack subject to a transient thermal loading

An asymptotic algorithm is applied to the problem of a finite, thermo-elastic solid containing a surface breaking crack, when the exterior surface is subjected to oscillatory thermal loading. This algorithm involves the study of a model problem. An analytical and numerical study of this model problem of a thermo-elastic half space containing a surface breaking crack and subjected to oscillatory thermal loading is presented. The crack surface is traction free. In particular, the amplitude of the stress intensity factor at the crack vertex is found as a function of the crack depth and the frequency of thermal oscillation.
