Oral treatment with plecanatide or dolcanatide attenuates visceral hypersensitivity via activation of guanylate cyclase-C in rat models

AIM To investigate the effects of plecanatide and dolcanatide on maintenance of paracellular permeability, integrity of tight junctions and on suppression of visceral hypersensitivity. MeTHODS Transport of fluorescein isothiocyanate(FITC)-dextran was measured to assess permeability across cell monolayers and rat colon tissues. effects of plecanatide and dolcanatide on the integrity of tight junctions in Caco-2 and T84 monolayers and on the expression and localization of occludin and zonula occludens-1(ZO-1) were examined by immunofluorescence microscopy. Anti-nociceptive activity of these agonists was evaluated in trinitrobenzene sulfonic acid(TNBS)-induced inflammatory as well as in non-inflammatory partial restraint stress(PRS) rat models. Statistical significance between the treatment groups in the permeability studies were evaluated using unpaired *** Treatment of T84 and Caco-2 monolayers with lipopolysaccharide(LPS) rapidly increased permeability, which was effectively suppressed when monolayers were also treated with plecanatide or dolcanatide. Similarly, when T84 and Caco-2 monolayers were treated with LPS, cell surface localization of tight junction proteins occludin and ZO-1 was severely disrupted. When cell monolayers were treated with LPS in the presence of plecanatide or dolcanatide, occludin and ZO-1 were localized at the cell surface of adjoining cells, similar to that observed for vehicle treated cells. Treatment of cell monolayers with plecanatide or dolcanatide without LPS did not alter permeability, integrity of tight junctions and cell surface localization of either of the tight junction proteins. In rat visceral hypersensitivity models, both agonists suppressed the TNBS-induced increase in abdominal contractions in response to colorectal distension without affecting the colonic wall elasticity, and both agonists also reduced colonic hypersensitivity in the PRS model. Seasonal Dynamics of Turbidity Maximum in the Muthupet estuary, India

Results are presented of the longitudinal and vertical profiling of salinity and suspended particulate matter(SPM) at the Muthupet estuary, India, during a one year period under widely varying freshwater flow conditions. Freshwater flow was available during post-monsoon and monsoon. An up-estuary shift in the location of estuarine turbidity maxima(eTM) was observed during the transition from post-monsoon to pre-monsoon and further it shifted downstream during the transition from pre-monsoon to monsoon, thereby exhibiting a pronounced seasonal cycle. The salinity intrusion was dependent on the freshwater discharge and was expressed as a power function of freshwater flow, explaining 97% of the variance. The formation of a salt plug in Muthupet estuary and its seasonal dynamics were observed, which is not an identified feature of any of the Indian estuaries studied so far. The geographical positions of salt plug and eTM core were more or less the same during their formation. The occurrence of two eTM during the LW of post-monsoon and the absence of eTM during monsoon explains the strong seasonal variation in the formation of eTM. The primary factor affecting the formation of eTM was identified as the freshwater flow over an annual cycle; the resuspension of sediments by tidal current affecting the formation on a flood/ebb cycle was secondary. The extent of shift of eTM was found to be an inverse logarithmic function of the freshwater discharge. The separation between eTM intrusion and salinity intrusion increased two fold with the increase in eTM intrusion.

Validation of reference genes for gene expression studies in the emerald ash borer （Agrilus planipennis）

The emerald ash borer （eAB, Agrilus planipennis Fairmaire） an exotic invasive insect pest has killed millions of ash trees （Fraxinus spp.） across North America and threatens billions more. We validated six A. planipennis reference genes （actin, ACT; beta tubulin, e-TUB; glyceraldehyde-3-phosphate dehydrogenase, GAPDH ; ribosomal protein, RPL7; translation elongation factor 1~, TeF-la; and ubiquitin, UBQ） using geNorm, Normfinder and BestKeeper for accurate determination of target messenger RNA levels in gene expression studies. The stability of the six reference genes was evaluated in different larval tissues, developmental stages and two treatments ofA. planipennis using quantitative real-time polymerase chain reaction. Although there was no consistent ranking observed among the reference genes across the samples, the overall analysis revealed TeF-la as the most stable reference gene. GAPDH and ACT showed least stability for all the samples studied. We conclude that TeF-I~ is the most appropriate reference gene for gene expression studies in A. planipennis. Results obtained can be applicable for transcript profiling in other invasive insect pests. Further, these validated reference genes could also serve as the basis for selection of candidate reference genes in any given insect system post-validation.

This paper presents an efficient prediction model for a good learning environment using Random Forest(RF)*** consists of a series of modules;data preprocessing,data normalization,data split andfinally classification or prediction by the RF *** preprocessed data is normalized using minmax normalization often used before *** the input data or variables are measured at different scales,it is necessary to normalize them to contribute equally to the ***,the RF classifier is employed for course selection which is an ensemble learning method and k-fold cross-validation(k=10)is used to validate the *** proposed Prediction Model for Course Selection(PMCS)system is considered a multi-class problem that predicts the course for a particular learner with three complexity levels,namely low,medium and *** is operated under two modes;locally and *** former considers the gender of the learner and the later does not consider the gender of the *** database comprises the learner opinions from 75 males and 75 females per category(low,medium and high).Thus the system uses a total of 450 samples to evaluate the performance of the PMCS *** show that the system’s performance,while using locally i.e.,gender-wise has slightly higher performance than the global *** RF classifier with 75 decision trees in the global system provides an average accuracy of 97.6%,whereas in the local system it is 97%(male)and 97.6%(female).The overall performance of the RF classifier with 75 trees is better than 25,50 and 100 decision trees in both local and global systems.

The First Photometric and Spectroscopic Study of Contact Binary V2840 Cygni

The first photometric,spectroscopic and period variation studies of neglected short-period eclipsing binary V2840Cygni are *** mass ratio contact binaries(HMRCBs),especially those in the weak-contact configuration,are vital when probing the evolutionary models of contact binaries(CBs) using stellar *** photometric solutions reveal the weak-contact nature of V2840 Cygni with a high mass ratio(~1.36),motivating us to investigate the nature of such *** period variation study of V2840 Cygni spanning 15 yr shows a secular period decrease at a rate of ~5.5 × 10^(-7) day yr^(-1),indicating mass transfer between the *** superimposed cyclic variation provides a basic understanding of the possible third body(P_(3)- 8 yr,m_(3)- 0.51 M_(⊙)).Following the derived parameters,the evolution of the system is discussed based on the thermal relaxation oscillation(TRO) *** is found that V2840 Cygni falls in a special category of HMRCBs,which validates *** characterize the nature of HMRCBs,a catalog of 59 CBs with high mass ratios has been compiled along with their derived parameters from the *** all the HMRCBs in the study,a possible correlation between their contact configuration and observed period variations for relative log J_(rel) is *** spectroscopic study of V2840 Cygni provides evidence of the presence of magnetic activity in the system and the existence of ongoing mass transfer which is additionally deduced from the period variation *** LAMOST spectra of 17 HMRCBs are collected to interpret the stellar magnetic activity in such systems.

The maintenance of genetic variation in mutualism-related traits is key for understanding mutualism evolution,yet the mechanisms maintaining variation remain *** asked whether genotype-byenvironment(G3e)interaction is a potential mechanism maintaining variation in the model legume–rhizobia system,Medicago truncatula–ensifer *** planted 50 legume genotypes in a greenhouse under ambient light and shade to reflect reduced carbon availability for *** found an expected reduction under shaded conditions for plant performance traits,such as leaf number,aboveground and belowground biomass,and a mutualism-related trait,nodule *** also found G3e for nodule number,with83%of this interaction due to shifts in genotype fitness rank order across light environments,coupled with strong positive directional selection on nodule number regardless of light *** results suggest that G3e can maintain genetic variation in a mutualism-related trait that is under consistent positive directional selection across light environments.

Impact of patch size on woody tree species richness and abundance in a tropical montane evergreen forest patches of south India

We examined the effects of forest patch size on woody tree species richness and abundance in tropical montane evergreen forest patches of the Nilgiri region,south India. We sampled woody trees(>1 cm dbh) from 21 forest patches in the upper Nilgiri hills(> 2000 m elevation) and recorded a total of 35,146 individuals of 61 species, 45 genera and 30 families. Species richness and abundance of sapling/shrubs(≥ 1 to eased significantly with increasing patch size, but the species richness and abundance of small, medium and larger trees(≥ 10 to espectively) did not. Overall, forest interior species richness and abundance increased significantly with increasing patch size but edge species richness did not. Species richness and abundance of shade-tolerant and shade-demanding tree species also increased with increasing patch size. The abundance of zoochory dispersed tree species was significantly related to increasing patch size, but those dispersed by autochory did not display any clear relationship between patch size and species richness or *** findings suggest that with increasing forest patch area,tree compositional patterns may be driven by species specific shade-tolerance adaptations and dispersal *** responses in these traits by the plant community within the individual habitat zones of forest edge and interiors likely plays a major role in determining the inherent plant community and thus the subsequent ecological processes of forest patches, including their responses to increasing patch area.

Optical and near-infrared photometric study of NGC 6724

BVRI CCD photometry of the poorly studied open cluster NGC 6724 has been carried out down to a limiting magnitude of V-20 mag. The stars of the cluster have been observed using the Newtonian focus （f/4.84） of the 74-inch telescope at Kottamia Astronomical Observatory in egypt. Also, the 2MASS - JHK system is used to confirm the results we obtained. The main photometric parameters have been estimated for the present object; the diameter is found to be 6 arcmin, the distance is 15304-60pc from the Sun and the age is 900＋50Myr. The optical reddening e（B - V） = 0.65 mag, while the infrared reddening is e（J - H） = 0.20 mag. The slope of the mass function distribution and the relaxation time estimations indicate that cluster NGC 6724 is dynamically relaxed.
