FPR1 Antagonist (BOC-MLF) Inhibits Amniotic epithelial-mesenchymal Transition

Objective:Premature rupture of membranes(PROM)is a common pregnancy disorder that is closely associated with structural weakening of fetal membranes.Studies have found that formyl peptide receptor 1(FPR1)activates inflammatory pathways and amniotic epithelial-mesenchymal transition(eMT),stimulates collagen degradation,and leads to membrane weakening and membrane rupture.The purpose of this study was to investigate the anti-inflammatory and eMT inhibitory effects of FPR1 antagonist(BOC-MLF)to provide a basis for clinical prevention of PROM.Methods:The relationship between PROM,FPR1,and eMT was analyzed in human fetal membrane tissue and plasma samples using Western blotting,PCR,Masson staining,and eLISA assays.Lipopolysaccharide(LPS)was used to establish a fetal membrane inflammation model in pregnant rats,and BOC-MLF was used to treat the LPS rat model.We detected interleukin(IL)-6 in blood from the rat hearts to determine whether the inflammatory model was successful and whether the anti-inflammatory treatment was effective.We used electron microscopy to analyze the structure and collagen expression of rat fetal membrane.Results:Western blotting,PCR and Masson staining indicated that the expression of FPR1 was significantly increased,the expression of collagen was decreased,and eMT appeared in PROM.The rat model indicated that LPS caused the collapse of fetal membrane epithelial cells,increased intercellular gaps,and decreased collagen.BOC-MLF promoted an increase in fetal membrane collagen,inhibited eMT,and reduced the weakening of fetal membranes.Conclusion:The expression of FPR1 in the fetal membrane of PROM was significantly increased,and eMT of the amniotic membrane was obvious.BOC-MLF can treat inflammation and inhibit amniotic eMT.

本研究将外裹糊鱼块置于鼓风干燥箱中,40℃干燥3~9 h(3、4.5、6、7.5、9 h)后180℃油炸60 s,分析油炸前后外裹糊鱼块的水分状态、油炸外裹糊鱼块的油脂含量与分布、微观结构及质构特性,模拟油炸过程中油脂的吸收,探讨预干燥对油炸外裹糊鱼块油脂渗透和质构特性的影响。结果显示,油炸后外壳的深层结合水(T21)、弱结合水(T22)和自由水(T23)以及鱼块的结合水(T21)、肌纤维中(T22)和外部(T23)的水分含量均减小。随着干燥时间的延长,油炸后外壳T22峰的起始时间逐渐减小,水分的自由度降低,且T21和T22峰的信号强度减小,结合水向自由水转化;鱼块的T22峰只有干燥9h的发生明显左移;油炸外裹糊鱼块的表面油脂含量和总油脂含量呈减小趋势,表面渗透油脂含量逐渐增加;外壳结构先紧密后粗糙,鱼块表面先光滑后出现裂缝和孔洞;外壳中荧光强度逐渐减弱,苏丹红染色幅度逐渐减小,外壳与鱼块交界处的染色幅度逐渐增大;外壳的硬度和酥性先减小后增大,而鱼块的弹性和咀嚼性呈现相反趋势。这些结果表明,预干燥降低了外裹糊鱼块水分的自由度,改变了油炸外裹糊鱼块的微观结构,从而影响了油脂的渗透和质构特性。本研究可为低脂油炸外裹糊鱼制品的规模化生产提供科学指导和技术支撑。

本研究以黄鳝加工副产物鱼骨为碳源,通过一步水热法绿色合成黄鳝骨基碳量子点(Mab-CQDs),分析骨粉添加量、反应温度和时间对荧光强度的影响,优化Mab-CQDs的制备工艺。采用透射电子显微镜(TeM)、傅里叶红外光谱(FT-IR)和X射线衍射(XRD)等表征Mab-CQDs的形貌结构和光学性质,并选取金黄葡萄球菌(Staphylococcus aureus)和大肠杆菌(escherichia coli)作为受试菌株,探究Mab-CQDs的抗菌性能。优化的Mab-CQDs制备工艺为:黄鳝骨粉与超纯水比例为1∶30、反应温度为180℃、反应时间为8 h,该条件下制备的Mab-CQDs具有良好的荧光性能;TeM成像显示Mab-CQDs为分散良好的类球形结构,粒径范围为2.05 nm~5.94 nm;FT-IR结果表明Mab-CQDs表面含有大量氨基、羧基等极性官能团,具有良好的亲水性;抑菌实验表明Mab-CQDs对S.aureus和e.coli具有良好的抑制效果,最小抑菌质量浓度分别为5 mg/mL和10 mg/mL。本研究制备的Mab-CQDs粒径均匀,水溶性好,荧光性能优良,且具备明显的抑菌性能,可为淡水鱼加工副产物基CQDs的制备及其应用提供理论指导。

Towards sustainable adversarial training with successive perturbation generation

Adversarial training with online-generated adversarial examples has achieved promising performance in defending adversarial attacks and improving robustness of convolutional neural network models.However,most existing adversarial training methods are dedicated to finding strong adversarial examples for forcing the model to learn the adversarial data distribution,which inevitably imposes a large computational overhead and results in a decrease in the generalization performance on clean data.In this paper,we show that progressively enhancing the adversarial strength of adversarial examples across training epochs can effectively improve the model robustness,and appropriate model shifting can preserve the generalization performance of models in conjunction with negligible computational cost.To this end,we propose a successive perturbation generation scheme for adversarial training(SPGAT),which progressively strengthens the adversarial examples by adding the perturbations on adversarial examples transferred from the previous epoch and shifts models across the epochs to improve the efficiency of adversarial training.The proposed SPGAT is both efficient and effective;e.g.,the computation time of our method is 900 min as against the 4100 min duration observed in the case of standard adversarial training,and the performance boost is more than 7%and 3%in terms of adversarial accuracy and clean accuracy,respectively.We extensively evaluate the SPGAT on various datasets,including small-scale MNIST,middle-scale CIFAR-10,and large-scale CIFAR-100.The experimental results show that our method is more efficient while performing favorably against state-of-the-art methods.

An indicator model for assessing community resilience to the COVID-19 pandemic and its validation: A case study in Hong Kong

The COVID-19 outbreak had a significant negative impact on the world,and the fifth wave of COVID-19 in Hong Kong brought a considerable shock to Chinese society.There is a growing call for more resilient cities.However,empirical evidence and validation of modeling studies of resilience indicators for urban community responses to the COVID-19 pandemic still need to be provided.In this study,a resilience assessment indicator model comprising 4 subsystems,7 indicators,and 12 variables was developed to assess the resilience of Hong Kong communities in response to COVID-19(i.e.,Resilience Index).Furthermore,this study utilized regression models such as geographically weighted regression(GWR)and multiscale GWR(MGWR)to validate the resilience model proposed in this study at the model and variable levels.In the regression model,the Resilience Index and the individual variables in the resilience model are explanatory variables,and the outcomes of the COVID-19 pandemic(confirmed cases,confirmation rate,discharged cases,discharge rate)are dependent variables.The results showed that:(i)the resilience of Hong Kong communities to the COvID-19 pandemic was not strong in general and showed some clustered spatial distribution characteristics;(i)the validation results at the model level showed that the Resilience Index did not explain the consequences of the COvID-19 pandemic to a high degree;(ii)the validation results at the variable level showed that the MGWR model was the best at identifying the relationships between explanatory variables and the dependent variable;and(iv)compared with the model-level assessment results,the variable-level assessment explained the consequences of the COvID-19 pandemic better than the model level assessment results.The above analysis and the spatial distribution maps of the resilience variables can provide empirically based and targeted insights for policymakers.

Synergistic extraction and separation of thorium from rare earths in chloride media using mixture of Cextrant 230 and Cyanex 923

In order to lower the usage of expensive Cyanex 923 and increase the extraction capacity of the system of Cextrant 230,the synergistic extraction of thorium from chloride media by a mixture of Cextrant 230 and Cyanex 923 was investigated.The maximum synergistic enhancement coefficient(R)of 1.53 is obtained at 1:1 molar ratio of Cextrant 230/Cyanex 923.The syne rgistic extracted species of Th^(4+)is determined as ThCl_(4)·2Cextrant 230·Cyanex 923.The synergistic extraction of Th^(4+)is an entropy-driven exothermic process.The loading capacity of 0.60 mol/L mixed extractant for thorium is about 17.10 g/L(calculated as ThO_(2)),and the loaded thorium in the organic phase can be effectively stripped by distilled water.For comparison,rare earth cations are barely extracted under the similar conditions,suggesting that the mixtures can be applied to separate thorium from rare earths.A cascade extraction process was developed based on the synergistic extraction system to separate thorium from the hydrochloric acid leaching of bastnaesite.The content of thorium in the leaching solution decreases obviously from 19.90 mg/L to1.4μg/L by 3 stages of extraction,which is superior to sole Cextrant 230 or Cyanex 923.The introduction of Cextrant 230 into the extraction system not only lowers the usage of Cyanex 923 but also enhances the selective extraction of thorium at low acidity,implying that the synergistic extraction system can selectively extract thorium more efficiently and economically than the sole systems.

Practical cloud storage auditing using serverless computing

Cloud storage auditing research is dedicated to solving the data integrity problem of outsourced storage on the cloud. In recent years, researchers have proposed various cloud storage auditing schemes using different techniques. While these studies are elegant in theory, they assume an ideal cloud storage model;that is, they assume that the cloud provides the storage and compute interfaces as required by the proposed schemes. However, this does not hold for mainstream cloud storage systems because these systems only provide read and write interfaces but not the compute interface. To bridge this gap, this work proposes a serverless computing-based cloud storage auditing system for existing mainstream cloud object storage. The proposed system leverages existing cloud storage auditing schemes as a basic building block and makes two adaptations. One is that we use the read interface of cloud object storage to support block data requests in a traditional cloud storage auditing scheme. Another is that we employ the serverless computing paradigm to support block data computation as traditionally required. Leveraging the characteristics of serverless computing, the proposed system realizes economical, pay-as-you-go cloud storage auditing. The proposed system also supports mainstream cloud storage upper layer applications(e.g., file preview) by not modifying the data formats when embedding authentication tags for later auditing. We prototyped and open-sourced the proposed system to a mainstream cloud service, i.e., Tencent Cloud. experimental results show that the proposed system is efficient and promising for practical use. For 40 GB of data, auditing takes approximately 98 s using serverless computation. The economic cost is 120.48 CNY per year, of which serverless computing only accounts for 46%. In contrast, no existing studies reported cloud storage auditing results for real-world cloud services.

Improving benzene catalytic oxidation on Ag/Co_(3)O_(4) by regulating the chemical states of Co and Ag

Silver(9 wt.%)was loaded on Co_(3)O_(4)-nanofiber using reduction and impregnation methods,respectively.Due to the stronger electronegativity of silver,the ratios of surface Co^(3+)/Co^(2+) on Ag/Co_(3)O_(4) were higher than on Co_(3)O_(4),which further led to more adsorbed oxygen species as a result of the charge compensation.Moreover,the introducing of silver also obviously improved the reducibility of Co_(3)O_(4).Hence the Ag/Co_(3)O_(4) showed better catalytic performance than Co_(3)O_(4) in benzene oxidation.Compared with the Ag/Co_(3)O_(4) synthesized via impregnation method,the one prepared using reduction method(named as Ag Co-R)exhibited higher contents of surface Co^(3+) and adsorbed oxygen species,stronger reducibility,as well as more active surface lattice oxygen species.Consequently,Ag Co-R showed lowest T_(90) value of 183℃,admirable catalytic stability,largest normalized reaction rate of1.36×10^(-4)mol/(h·m^(2))(150℃),and lowest apparent activation energy(e_(a))of 63.2 kJ/mol.The analyzing of in-situ DRIFTS indicated benzene molecules were successively oxidized to phenol,o-benzoquinone,small molecular intermediates,and finally to CO_(2) and water on the surface of Ag Co-R.At last,potential reaction pathways including five detailed steps were proposed.

Base editor-mediated large-scale screening of functional mutations in bacteria for industrial phenotypes

Base editing,the targeted introduction of point mutations into cellular DNA,holds promise for improving genome-scale functional genome screening to single-nucleotide resolution.Current efforts in prokaryotes,however,remain confined to loss-of-function screens using the premature stop codons-mediated gene inactivation library,which falls far short of fully releasing the potential of base editors.Here,we developed a base editor-mediated functional single nucleotide variant screening pipeline in escherichia coli.We constructed a library with 31,123 sgRNAs targeting 462 stress response-related genes in e.coli,and screened for adaptive mutations under isobutanol and furfural selective conditions.Guided by the screening results,we successfully identified several known and novel functional mutations.Our pipeline might be expanded to the optimization of other phenotypes or the strain engineering in other microorganisms.

Optimal Stochastic Scheduling Strategy of Multi-vector energy Complex Integrated with Full-blown Power-to-biomethane Model

We propose an optimal stochastic scheduling strategy for a multi-vector energy complex(MeC), considering a fullblown model of the power-to-biomethane(Pt M) process. Unlike conventional optimization that uses a simple efficiency coefficient to coarsely model energy conversion between electricity and biomethane, a detailed Pt M model is introduced to emphasize the reactor kinetics and chemical equilibria of methanation. This model crystallizes the interactions between the Pt M process and MeC flexibility, allowing to adjust the operating condition of the methanation reactor for optimal MeC operation in stochastic scenarios. Temperature optimization and flowsheet design of the Pt M process increase the average selectivity of methane(i.e., ratio between net biomethane production and hydrogen consumption) up to 83.7% in the proposed synthesis flowsheet. Simulation results can provide information and predictions to operators about the optimal operating conditions of a Pt M unit while improving the MeC flexibility.
