The aim of the present study is to develop a surface plasmon resonance sensor for the detection of vitamin B2,vitamin B9,and vitamin B12 in food samples by using the molecular imprinting *** vitamin B2,vitamin B9,and vitamin B12 imprinted and the non-imprinted surface plasmon resonance sensor chip surfaces were characterized by using contact angle measurements,atomic force microscopy,ellipsometry,and Fourier transform infrared-attenuated total *** real-time detection of vitamin B2,vitamin B9,and vitamin B12 was analyzed by using aqueous solutions in the concentration range of 0.01 ng/mL−10 ng/mL for vitamin B2,0.1 ng/mL−8.0 ng/mL for vitamin B9,and 0.01 ng/mL−1.5 ng/mL for vitamin *** limit of detection values was calculated as 1.6×10^(−4) ng/mL for vitamin B2,13.5×10^(−4) ng/mL for vitamin B9,and 2.5×10^(−4) ng/mL for vitamin B12,*** experiments were performed by using vitamin B1 and vitamin *** reproducibility of surface plasmon resonance sensors was investigated both on the same day and on different days for four *** studies of the prepared surface plasmon resonance(SPR)sensors were performed by liquid chromatography-tandem mass spectrometry(LC-MS/MS).

Kefir is a fermented milk product produced by kefir grains traditionally or lyophilized starter *** quality characteristics of kefir produced at 22℃ and 30℃ using kefir grains after addition of microbial levan and pullulan were investigated for 14 ***,the survival of different Gram-positive and Gram-negative bacteria strains which were inoculated before and after fermentation were investigated during *** pH and titratable acidity were in the ranges of 4.67–4.39 and 1.10–0.87%,*** highest exopolysaccharide values(458.3–465.3 mg/kg)were found in the samples produced with the addition of 1%(w/v)of microbial levan at 30℃.Yeast counts were in the ranges of 6.73–5.11 log CFU/mL while the lactobacilli and lactic streptococci counts were in the ranges of 9.64–7.91 and 9.64–8.69 log CFU/mL,*** microbial polysaccharide addition did not show any significant differences in the enumeration of lactic acid bacteria in kefir(p>0.05).All Gram-positive and Gram-negative bacteria grew/survived during fermentation however Listeria monocytogenes was the most susceptible test bacterium to metabolites during *** highest reduction in the counts of *** was determined as 2.39 log units after 24 h of fermentation at 30℃ in the samples which were inoculated before fermentation and produced with the addition of *** samples produced at 30℃ without polysaccharide addition were considered as the best with respect to appearance,taste,consistency and overall acceptability.

In this study,the patulin imprinted and the non-imprinted nanoparticles are synthesized by the two-phase mini emulsion polymerization method and characterized by zeta-size analysis,Fourier transform infrared spectroscopy,and scanning electron ***,the patulin imprinted and the non-imprinted nanoparticles are attached on the surface of surface plasmon resonance(SPR)*** patulin imprinted and the non-imprinted SPR nanosensors are characterized by using atomic force microscope,ellipsometer,and contact angle *** studies for patulin detection are carried out in the concentration range of *** limit of detection and the limit of quantification values are obtained as 0.011 nmol and 0.036 nmol,*** all kinetic analysis,the response time is 13 min for equilibration,adsorption,and desorption *** selectivity studies of the patulin imprinted and the non-imprinted SPR nanosensors are determined in the presence of ochratoxin A and aflatoxin *** order to demonstrate the applicability,validation studies of the patulin imprinted SPR nanosensor are performed by liquid chromatography-tandem mass spectrometry(LC-MS).

Computational Prediction of Micro RnAs from Toxoplasma gondii Potentially Regulating the Hosts' Gene Expression

MicroRnAs （miRnAs） were discovered two decades ago, yet there is still a great need for further studies elucidating their genesis and targeting in different phyla. Since experimental discovery and validation of miRnAs is difficult, computational predictions are indispensable and today most computational approaches employ machine learning. Toxoplasma gondii, a parasite residing within the cells of its hosts like human, uses miRnAs for its post-transcriptional gene reg- ulation. It may also regulate its hosts＇ gene expression, which has been shown in brain cancer. Since previous studies have shown that overexpressed miRnAs within the host are causal for disease onset, we hypothesized that T. gondii could export miRnAs into its host cell. We computationally predicted all hairpins from the genome of T. gondii and used mouse and human models to filter possible candidates. These were then further compared to known miRnAs in human and rodents and their expression was examined for T. gondii grown in mouse and human hosts, respectively. We found that among the millions of potential hairpins in T. gondii, only a few thousand pass filtering using a human or mouse model and that even fewer of those are expressed. Since they are expressed and differentially expressed in rodents and human, we suggest that there is a chance that T. gondii may export miRnAs into its hosts for direct regulation.

Is the iron regulatory hormone hepcidin a risk factor for alcoholic liver disease?

Despite heavy consumption over a long period of time, only a small number of alcoholics develop alcoholic liver disease. This alludes to the possibility that other factors, besides alcohol, may be involved in the progression of the disease. Over the years, many such factors have indeed been identified, including iron. Despite being crucial for various important biological processes, iron can also be harmful due to its ability to catalyze Fenton chemistry. Alcohol and iron have been shown to interact synergistically to cause liver injury. Iron-mediated cell signaling has been reported to be involved in the pathogenesis of experimental alcoholic liver disease. Hepcidin is an iron-regulatory hormone synthesized by the liver, which plays a pivotal role in iron homeostasis. Both acute and chronic alcohol exposure suppress hepcidin expression in the liver. The sera of patients with alcoholic liver disease, particularly those exhibiting higher serum iron indices, have also been reported to display reduced prohepcidin levels. Alcohol-mediated oxidative stress is involved in the inhibition of hepcidin promoter activity and transcription in the liver. This in turn leads to an increase in intestinal iron transport and liver iron storage. Hepcidin is expressed primarily in hepatocytes. It is noteworthy that both hepatocytes and Kupffer cells are involved in the progression of alcoholic liver disease. However, the activation of Kupffer cells and TnF-α signaling has been reported not to be involved in the down-regulation of hepcidin expression by alcohol in the liver. Alcohol acts within the parenchymal cells of the liver to suppress the synthesis of hepcidin. Due to its crucial role in the regulation of body iron stores, hepcidin may act as a secondary risk factor in the progression of alcoholic liver disease. The clarification of the mechanisms by which alcohol disrupts iron homeostasis will allow for further understanding of the pathogenesis of alcoholic liver disease.

Changes in retinal vessel diameters in migraine patients during attack-free period

AIM：To evaluate the retinal vessel diameters in patients with migraine by optical coherence tomography（OCT）.METHODS：In this cross-sectional study,124 eyes of 62 patients with a diagnosis of unilateral migraine during attack-free period and 42 age-and sex-matched control subjects were included. Migraine patients were divided into the ≤2 migraine attacks per month group and the ≥5 migraine attacks per month group. All subjects underwent complete ophthalmological and neurological examinations before measurements. Retinal vessel diameters and choroidal thickness were examined with the Spectralis ***：The mean diameters of the arteries in the eyes on the headache side of control group,≥5 migraine attacks per month and ≤2 migraine attacks per month group at 480 μm from the optic disk（Raster 3）were 119.54±46.69,136.68±25.93 and 119.34±31.75 μm respectively with a steady decline to 105.57±32.15,118.18±31.87 and 108.05±38.77 μm at 1440 μm（Raster 7）,the last measurement point,respectively. The retinal artery diameter measurements were significantly increased in ≥5 migraine attacks per month patients at four out of five measured points compared to control group（P〈0.05）. There were no statistical differences at any of the points of vein measurements. The choroidal thickness measurements were significantly decreased in ≥5 migraine attacks per month patients at all measured points compared to control group（P〈0.05）.COnCLUSIOn：The retinal artery diameter is found to increase significantly and the choroidal thickness is found to decrease in the eyes on the headache side in ≥5 migraine attacks per month patients compared to control group.

Aim: To investigate the role of fexofenadine, a mast cell blocker, on semen quality in the treatment of infertile men. Methods: The study included 16 Turkish idiopathic infertile men with azoospermia or oligozoospermia who underwent testicular biopsy to examine mast cells containing tryptase. In all patients, a complete medical history, clinical examination, semen analysis and serum hormone assay were carried out. The biopsy specimens were immunohistochemically stained with antihuman tryptase for mast cells. The number of total mast cells per seminiferous tubule was calculated and recorded as mast cell index. The patients were divided into two groups according to their mast cell index: the higher (≥1, n=9) and the lower (n=7) index groups. Fexofenadine was administered orally at a dose of 180 mg/day for 4 to 9 months. Pre-and post-treatment semen parameters, including total motile sperm counts (TMC) were recorded and compared. Spontaneous pregnancies after the treatment were registered. Results: There was no statistically significant difference in TMC between the pre-treatment and post-treatment values in patients with higher and lower mast cell index (P≥0.05). In both groups, nobody had a significant response to the treatment and there was no spontaneous pregnancy after the treatment. Conclusion: Although testicular dysfunction is closely associated with increased number of testicular mast cells, fexofenadine, a mast cell blocker, appears not having any benefit in the treatment of Turkish infertile men with a significant increase in testicular mast cells. We study continuous data assimilation(CDA)applied to projection and penalty methods for the navier-Stokes(nS)*** and projection methods are more efficient than consistent ns discretizations,however are less accurate due to modeling error(penalty)and splitting error(projection).We show analytically and numerically that with measurement data and properly chosen parameters,CDA can effectively remove these splitting and modeling errors and provide long time optimally accurate solutions.

A Double-Sided View to Adult Vaccination:The Opinions and Attitudes of Patients and Health Workers

Objective: Although there are adult vaccination schemes in our country, there are serious deficiencies in the way that doctors direct the patients to this vaccination, but also patients have to make and demand these vaccinations. The aim of our study is to identify the shortcomings in this area and draw a roadmap for what arrangements should be made in terms of physicians and patients in order to increase adult immunization rates in primary care. Method: We conducted a two-phase, multicentered, descriptive clinical trial between October and December 2017. The first phase of the trial was carried out with patients from 3 Family Health Centers in Antalya, Istanbul and Osmaniye. Patients to be interviewed were selected voluntarily among Family Health Center’s applicants. The second phase of the trial was carried out with health workers, who were participated to trial from 26 different provinces of Turkey. In the process, a questionnaire of 19 questions was applied to primary health care workers by the internet. Results: 490 patients were included in the study. There was a significant difference between gender, age, education level and guideline follow-up and vaccination status of patients [p Conclusion: As a result of the studies including our study about adult vaccination, only 10% - 20% of the targeted groups in adults can be vaccinated. However, just like in childhood during adulthood, vaccinations protect individuals from diseases and provide economic benefits. Firstly, the knowledge level of physicians on adult immunization should be updated with in-service trainings, they should first be convinced to vaccinate because they are in risk group and then they should be recommended vaccination to patient to increase the vaccination rates.
