Genetic Engineered Ultrasound-Triggered Injectable Hydrogels for Promoting Bone Reconstruction

Genetic engineering technology can achieve specific gene therapy for a variety of diseases, but the current strategy still has some flaws, such as a complex system, single treatment, and large implantation trauma. Herein, the genetic engineering injectable hydrogels were constructed by ultrasonic technology for the first time to realize in vivo ultrasound-triggered in situ cross-linking and cell gene transfection, and finally complete in situ gene therapy to promote bone reconstruction. First, ultrasound-triggered calcium release was used to activate transglutaminase and catalyze the transamidation between fibrinogen. Simultaneously, liposome loaded with Zinc-finger E-box-binding homeobox 1 (ZEB1) gene plasmid (Lip-ZEB1) was combined to construct an ultrasound-triggered in situ cross-linked hydrogels that can deliver Lip-ZEB1. Second, ultrasound-triggered injectable hydrogel introduced ZEB1 gene plasmid into endothelial cell genome through Lip-ZEB1 sustained release, and then acted on the ZEB1/Notch signal pathway of cells, promoting angiogenesis and local bone reconstruction of osteoporosis through genetic engineering. Overall, this strategy provides an advanced gene delivery system through genetic engineered ultrasound-triggered injectable hydrogels.

Enhanced hepatic metabolic perturbation of polystyrene nanoplastics by UV irradiation-induced hydroxyl radical generation

The environmental behavior of and risks associated with nanoplastics(NPs)have attracted considerable ***,compared to pristine NPs,environmental factors such as ultraviolet(UV)irradiation that lead to changes in the toxicity of NPs have rarely been *** evaluated the changes in morphology and physicochemical properties of polystyrene(PS)NPs before and after UV irradiation,and compared their hepatotoxicity in *** results showed that UV irradiation caused particle size reduction and increased the carbonyl index(CI)and negative charge on the particle ***-aged PS NPs(aPS NPs)could induce the generation of hydroxyl radicals(·OH),but also further promoted the generation of·OH in the Fenton reaction *** pathological damage was more severe in mice exposed to aPS NPs,accompanied by a large number of vacuoles and hepatocyte balloon-like changes and more marked perturbations in blood glucose and serum lipoprotein,alanine aminotransferase and aspartate aminotransferase *** addition,exposure to PS NPs and aPS NPs,especially aPS NPs,triggered oxidative stress and significantly damaged the antioxidant capacity of mice *** with PS NPs,exposure to aPS NPs increased the number of altered metabolites in hepatic and corresponding metabolic pathways,especially glutathione *** research suggests that UV irradiation can disrupt the redox balance in organisms by promoting the production of·OH,enhancing PS NPs-induced liver damage and metabolic *** study will help us understand the health risks of NPs and to avoid underestimation of the risks of NPs in nature.

Histological,transcriptomic,and gene functional analyses reveal the regulatory events underlying gibberellin-induced parthenocarpy in tomato

Gibberellin(GA)is one of the major plant hormones that promote parthenocarpy,a highly valuable agronomic ***,we demonstrated that exogenous GA3application triggered the formation of parthenocarpic fruits with smaller size but unchanged shape in tomato(Solanum lycopersicum).These fruits exhibited a thicker pericarp,undeveloped ovules,and few jelly tissues,leading to smaller locules with empty *** investigation showed that GA treatment produced more cell layers with larger cells in the pericarp,suggesting its promotion in both cell division and *** analyses between GA-3and mock-treated unpollinated ovaries/fruits identified a large number of differentially expressed genes related to hormones,cell division,cell expansion,and transcription factors,implying that they coordinately regulated parthenocarpy conferred by *** particular,the downregulation of five reported repressors of tomato parthenocarpy,including two auxin signaling components,AUXIN RESPONSE FACTOR5(SlARF5)and Sl ARF7,and three MADS-box genes,TOMATO APETALA3(TAP3),TOMATO PISTILLATA(TPI),and AGAMOUS-LIKE6(SlAGL6),after GA treatment might play a key role in this ***,we found that the knockdown of a GA signaling factor SlMYB33,which was depressed by GA treatment,induced parthenocarpic fruit set in tomato,an effect that might have been achieved by enhancing GA biosynthesis and decreasing the expression of some repressors of tomato ***,our results provide a basis for understanding the regulatory mechanism of GA in tomato parthenocarpy.

Modeling of multiphase flow in low permeability porous media:Effect of wettability and pore structure properties

Multiphase flow in low permeability porous media is involved in numerous energy and environmental ***,a complete description of this process is challenging due to the limited modeling scale and the effects of complex pore structures and *** address this issue,based on the digital rock of low permeability sandstone,a direct numerical simulation is performed considering the interphase drag and boundary slip to clarify the microscopic water-oil displacement *** addition,a dual-porosity pore network model(PNM)is constructed to obtain the water-oil relative permeability of the *** displacement efficiency as a recovery process is assessed under different wetting and pore structure *** show that microscopic displacement mechanisms explain the corresponding macroscopic relative *** injected water breaks through the outlet earlier with a large mass flow,while thick oil films exist in rough hydrophobic surfaces and poorly connected *** variation of water-oil relative permeability is significant,and residual oil saturation is high in the oil-wet *** flooding is extensive,and the residual oil is trapped in complex pore networks for hydrophilic pore surfaces;thus,water relative permeability is lower in the water-wet *** the displacement efficiency is the worst in mixed-wetting systems for poor water *** negatively correlates with invading oil volume fraction due to strong capillary resistance,and a large microporosity corresponds to low residual oil *** work provides insights into the water-oil flow from different modeling perspectives and helps to optimize the development plan for enhanced recovery.

ST-LSTM-SA:A New Ocean Sound Velocity Field Prediction Model Based on Deep Learning

The scarcity of in-situ ocean observations poses a challenge for real-time information acquisition in the *** the crucial hydroacoustic environmental parameters,ocean sound velocity exhibits significant spatial and temporal variability and it is highly relevant to oceanic *** this study,we propose a new data-driven approach,leveraging deep learning techniques,for the prediction of sound velocity fields(SVFs).Our novel spatiotemporal prediction model,STLSTM-SA,combines Spatiotemporal Long Short-term Memory(ST-LSTM) with a self-attention mechanism to enable accurate and real-time prediction of *** circumvent the limited amount of observational data,we employ transfer learning by first training the model using reanalysis datasets,followed by fine-tuning it using in-situ analysis data to obtain the final prediction *** utilizing the historical 12-month SVFs as input,our model predicts the SVFs for the subsequent three *** compare the performance of five models:Artificial Neural Networks(ANN),Long Shortterm Memory(LSTM),Convolutional LSTM(ConvLSTM),ST-LSTM,and our proposed ST-LSTM-SA model in a test experiment spanning 2019 to *** results demonstrate that the ST-LSTM-SA model significantly improves the prediction accuracy and stability of sound velocity in both temporal and spatial *** ST-LSTM-SA model not only accurately predicts the ocean sound velocity field(SVF),but also provides valuable insights for spatiotemporal prediction of other oceanic environmental variables.

Potential evaluation and implementation strategy for pocket park construction in high-density urban areas: A case study in Dalian, China

As an important element of urban renewal in highly urbanized areas, pocket parks with small size, flexible layout and daily accessibility are becoming a major component of green infrastructure and the mainstay of outdoor recreation space expansion in high-density urban centers. Nevertheless, the absence of a comprehensive framework for evaluating the potential of pocket park construction (PPC), one that integrates diverse influencing factors on a macro scale, has resulted in the random installation of such spaces, often failing to optimize the utilization of urban land. Addressing this critical lacuna, we propose an approach to evaluate PPC potential from a city-scale perspective, which is used to support the determination of which land units should be prioritized for PPC. A complete and feasible workflow was also established to identify potential land units, construct an index system for PPC combining demand and supply levels, quantitatively calculate indices based on remote sensing (RS) and geographic information system (GIS), accurately evaluate PPC potential using an entropy-weighted TOPSIS model, and develop targeted renewal strategies. A case study in Dalian, China, demonstrated the applicability and implications of the workflow. The results showed that it is flexible and easy to adapt to different local contexts, allowing evaluators to introduce parameters considering the availability of local data, and will help decision makers to build pocket parks in the most effective plots, providing a strong reference for high-quality development in other high-density urban centers facing the contradiction between ecological construction and land scarcity.

Federated learning effectively addresses issues such as data privacy by collaborating across participating devices to train global ***,factors such as network topology and computing power of devices can affect its training or communication process in complex network *** and network convergence(CNC)of sixth-generation(6G)networks,a new network architecture and paradigm with computing-measurable,perceptible,distributable,dispatchable,and manageable capabilities,can effectively support federated learning training and improve its communication *** guiding the participating devices'training in federated learning based on business requirements,resource load,network conditions,and computing power of devices,CNC can reach this *** this paper,to improve the communication eficiency of federated learning in complex networks,we study the communication eficiency optimization methods of federated learning for CNC of 6G networks that give decisions on the training process for different network conditions and computing power of participating *** simulations address two architectures that exist for devices in federated learning and arrange devices to participate in training based on arithmetic power while achieving optimization of communication efficiency in the process of transferring model *** results show that the methods we proposed can cope well with complex network situations,effectively balance the delay distribution of participating devices for local training,improve the communication eficiency during the transfer of model parameters,and improve the resource utilization in the network.

《孝经》在宋至明初受到冷遇，但在晚明时期再度兴盛。《孝经》复兴之初，是通过与《大学》同出于孔、曾授受的关系，与《大学》相结合，进而在义理上与“四书”形成互为表里的关系。但这仍局限于“四书”体系，真正对宋明理学形成突破，是从《孝经》与《春秋》相配开始。基于孔子“志在《春秋》,行在《孝经》”言论，明代中后期《春秋》与《孝经》再度结合，形成刑书与德教的关系。由此，《孝经》进一步与“六经”相配，回归“六经之总会”的经典地位。同时，这也是《孝经》摆脱蒙书、回归孝治经典的过程。这一过程始于对王安石罢黜《孝经》的批判，以及对“孝”的概念范围的反思，目的是使《孝经》超越家庭内部的事亲仪节，回归先王以孝治天下的大经大法，体现了晚明儒者对孝德的高度肯定，隐含着对“二帝三王”之治的畅想。

Quantifying freeze-melt dynamics of lakes on the Tibetan Plateau using Sentinel-1 synthetic aperture radar imagery

The ice phenology of alpine lakes on the Tibetan Plateau(TP)is a rapid and direct responder to climate changes,and the variations in lake ice exhibit high temporal frequency *** and passive microwave data are widely used to monitor lake ice changes with high temporal ***,the low spatial resolutions make it difficult to effectively quantify the freeze-melt dynamics of *** work used Sentinel-1 synthetic aperture radar(SAR)data to derive high-resolution ice maps(about 6 days),then with the aid of Sentinel-2 optical images to quantify freeze-melt processes in three typical lakes on the TP(*** Co,Ayakekumu Lake,and Nam Co).The results showed that three lakes had an average annual ice period of 125-157 days and a complete ice cover period of 72-115 days,from 2018 to *** exhibit different ice phenology *** Co is characterized by repeated episodes of freezing,melting,and refreezing,resulting in a prolonged freeze-up ***,the break-up period of Nam Co lasts for a longer duration(about 19 days),and the break-up exhibits a smooth ***,Ayakekumu Lake showed more significant inter-annual fluctuations in the freeze-up period,with deviations of up to 28 days observed among different *** to the other two lakes,Selin Co experienced a relatively short freeze-up and break-up *** short,Sentinel-1 SAR data can effectively monitor the weekly and seasonal variations in lake ice on the ***,this data facilitates quantification of the freeze-melt dynamics.
