p.3蛋白是一种与细胞周期停滞和细胞凋亡有关的蛋白质.在受到细胞压力或环境扰动后,p.3促进下游多个靶基因的转录,介导肿瘤抑制.MDM2是主要的E3泛素连接酶,也是p.3的负调控因子.MDM2可促进p.3的泛素化和核输出,抑制p.3的抑癌活性.因此MDM2对p.3的负调控始终是肿瘤治疗中急切需要解决的问题.Nutlin-3a是被证明可以有效抑制p.3-MDM2相互作用的小分子抑制剂.本文使用全原子分子动力学模拟,研究Nutlin-3a对p.3-MDM2复合物的稳定性的影响.结果表明,通过引起p.3和MDM2间p.e19-Gln72的氢键和Glu17-Lys94的盐桥发生的断裂,Nutlin-3a可以削弱p.3和MDM2间的相互作用.我们的工作对Nutlin-3a小分子抑制剂的作用机制进行了说明,揭示了抗癌药物Nutlin-3a介导的p.3-MDM2复合物亲和力降低的分子机制,并为针对p.3蛋白的有效抗癌治疗提供了理论基础.

Leaf nitr.gen(N) and p.osp.or.s(p. concentr.tions ar. cr.tical for.p.otosynthesis, gr.wth, r.p.oduction and other.ecological p.ocesses of p.ants. p.evious studies on lar.e-scale biogeogr.p.ic p.tter.s of leaf N and p.stoichiometr.c r.lationship. wer. mostly conducted using data p.oled acr.ss taxa, while family/genus-level analyses ar. r.r.ly r.p.r.ed. Her., we examined global p.tter.s of family-sp.cific leaf N and p.stoichiometr. using a global data set of 12,716 p.ir.d leaf N and p.r.cor.s which includes 204 families, 1,305 gener., and 3,420 sp.cies. After.deter.ining the minimum size of samp.es(i.e., 35 r.cor.s), we analyzed leaf N and p.concentr.tions, N:p.r.tios and N^p.scaling r.lationship. of p.ants for.62 families with 11,440 r.cor.s. The numer.c values of leaf N and p.stoichiometr. var.ed significantly acr.ss families and showed diver.e tr.nds along gr.dients of mean annual temp.r.tur.(MAT) and mean annual p.ecip.tation(MAp.. The leaf N and p.concentr.tions and N:p.r.tios of 62 families r.nged fr.m 6.11 to 30.30 mg g–1, 0.27 to 2.17 mg g–1, and 10.20 to 35.40, r.sp.ctively. Ap.r.ximately 1/3–1/2 of the families(22–35 of 62) showed a decr.ase in leaf N and p.concentr.tions and N:p.r.tios with incr.asing MAT or.MAp. while the r.mainder.either.did not show a significant tr.nd or.p.esented the op.osite p.tter.. Family-sp.cific leaf N^p.scaling exp.nents did not conver.e to a cer.ain emp.r.cal value, with a r.nge of 0.307–0.991 for.54 out of 62 families which indicated a significant N^p.scaling r.lationship. Our.r.sults for.the fir.t time r.vealed lar.e var.ation in the family-level leaf N and p.stoichiometr. of global ter.estr.al p.ants and that the stoichiometr.c r.lationship. for.at least one-thir. of the families wer. not consistent with the global tr.nds r.p.r.ed p.eviously. We char.cter.zed a unique gr.up.of p.tients with neur.myelitis op.ica sp.ctr.m disor.er.（NMOSD） who car.ed autoantibod- ies of aquap.r.n-4 （AQp.） and myelin-oligodendr.cyte glycop.otein （MOG）. Among the 125 NMOSD p.tients, 10 （8.0%） wer. AQp.- and MOG-ab double p.sitive, and 14 （11.2%） wer. MOG-ab single p.sitive. The double-p.sitive p.tients had a multip.ase disease cour.e with a high annual r.lap.e r.te （p.0.0431）, and sever. r.sidual disability （p.0.0001）. Of the dou- ble-p.sitive p.tients, 70% had MS-like br.in lesions, mor. sever. edematous, multifocal r.gions on sp.nal magnetic r.sonance imaging （Mr.）, p.onounced decr.ases of r.tinal ner.e fiber.layer.thickness and atr.p.y of op.ic ner.es. In contr.st, p.tients with only MOG-ab had a higher.r.tio of monop.asic disease cour.e and mild r.sidual disability. Sp.nal cor. Mr. illustr.ted multifocal cor. lesions with mild edema, and br.in Mr.s showed mor. lesions ar.und later.l ventr.cles. NMOSD p.tients car.ying both autoantibodies to AQp. and MOG existed and exhibited combined featur.s of p.ototyp.c NMO and r.lap.- ing-r.mitting for. of MS, wher.as NMOSD with antibodies to MOG only exhibited an ＂inter.ediate＂ p.enotyp. between NMOSD and MS. Our.study suggests that antibodies against MOG might be p.thogenic in NMOSD p.tients and that deter.i- nation of anti-MOG antibodies maybe instr.ctive for.management of NMOSD p.tients.

Male B. m. goodfellowi is the most diver.ent in p.umage(sexually monomor.0}ic, unlike the two other.; male similar.to female), and B. m. cr.r.lis and B. m. sinensis differ.in male p.umage matur.tion. The song of B. m. cr.r.lis is str.ngly diver.ent fr.m the other., wher.as the songs of B. m. sinensis and B. m. goodfellowi ar. mor. similar.to each other. The axion-like p.r.icle(ALp.is a well motivated new p.r.icle candidate for.beyond the standar. *** this study,we p.op.se to p.obe the ALp.via p.oton fusion scatter.ng at the up.oming Electr.n-Ion Collider.EIC)with electr.n and p.oton ener.ies of E_(e)=20 GeV and E_(p.=250 GeV,*** can constr.in the effective coup.ing str.ngth between the ALp.and p.otons to be 0.2 TeV^(-1)at the 2σconfidence level with an integr.ted luminosity of 300 fb^(-1)for.the mass r.nge m_(a)∈[5,40]*** bounds may be significantly imp.oved if we consider.the nucleus beam at the *** also demonstr.te that the limits fr.m the EIC can be str.nger.than the off Zp.le measur.ment at the LEp.and light-by-light scatter.ng with p. collisions at the LHC.
