Potential mechanism of Xuanfei Baidu Formula in treating new coronavirus pneumonia based on network pharmacology and molecular docking

ω-3多不饱和脂肪酸对大鼠慢性萎缩性胃炎的保护作用研究

目的探讨ω-3多不饱和脂肪酸(ω-3 PUFA)对大鼠慢性萎缩性胃炎的影响。方法用综合法建立慢性萎缩性胃炎大鼠模型。将造模成功的大鼠随机分为低、中、高剂量实验组、模型组、阳性对照组。另取正常大鼠作为正常组,每组9只。低、中、高剂量实验组每日分别灌胃3、6、12 mL·kg^(-1)ω-3 PUFA。阳性对照组每日灌胃0.24 g·kg^(-1)的维酶素混悬液。正常组和模型组每日灌胃等量0.9%NaCl。持续给药12周。用酶联免疫吸附试验(ELISA)法检测血清白细胞介素-1β(IL-1β)水平,用原位末端标记(TUNEL)染色法检测胃黏膜组织细胞凋亡情况,用试剂盒检测超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果正常组、模型组、低剂量实验组、中剂量实验组、高剂量实验组、阳性对照组的IL-1β水平分别为(58.96±8.23)、(140.02±19.65)、(109.81±14.35)、(98.72±12.64)、(85.31±11.42)和(77.64±10.23)pg·mL^(-1),凋亡指数分别为(7.89±1.36)%、(77.12±10.05)%、(42.33±6.87)%、(31.05±5.29)%、(22.76±4.16)%和(16.89±3.05),SOD水平分别为(398.71±58.24)、(170.25±29.81)、(249.81±34.26)、(268.04±34.11)、(322.15±46.36)和(276.42±29.81)U·mg^(-1),MDA水平分别为(3.55±0.87)、(11.02±2.15)、(8.02±1.50)、(6.92±1.23)、(5.98±1.27)和(6.67±1.32)U·mg^(-1)。模型组的上述指标与正常组比较,在统计学上差异均有统计学意义(均P<0.01);低、中、高剂量实验组、阳性对照组的上述指标与模型组比较,在统计学上差异均有统计学意义(均P<0.01)。结论ω-3 PUFA可改善大鼠慢性萎缩性胃炎,其机制可能与ω-3 PUFA的抗炎、抗凋亡和抗氧化应激作用有关。

Rare earth giant magnetostrictive materials(GMMs)Tb_(1-x)Dy_(x)Fe_(2±δ)(Tb-Dy-Fe)have been successfully employed in many microelectromechanical devices due to their excellent magnetostrictive properties at room ***,Tb-Dy-Fe still shows a relatively large coercivity with high hysteresis,which inevitably limits its application ***,micromagnetic simulations are performed to investigate the size effect of precipitated phase(α-Fe)on coercivity in *** results demonstrate that the coercivity is reduced from 31.46 to 12.48 mT with increasing the size ofα-Fe from 4 to 50 nm in Tb-Dy-Fe since the precipitated phase ofα-Fe can act as a magnetization reversal *** decreasing trend of coercivity can be well fitted with an inverse square relationship,which agrees well with the nucleation *** study highlights that the coercivity of Tb-Dy-Fe can be tailored by tuning the size ofα-Fe precipitation.

烟草生物碱性状的QTL定位

生物碱是烟草的重要化学成分。为明确烟草生物碱的遗传结构,发掘控制相关性状的主效位点,以烟草品种Y3、K326为亲本,构建大小为271的重组自交系群体。分别于2018、2019和2020年在云南省昆明市石林、玉溪市研和种植群体材料,检测总植物碱(TPA)、烟碱(NIC)、降烟碱(NOR)、假木贼碱(ANAB)和新烟草碱(ANAT)5种生物碱表型。对群体进行基因组测序,构建包含46,129个标记的遗传连锁图谱。利用基于混合线性模型的QTL定位方法及软件QTLNetwork2.0,进行QTL定位分析。共定位15个具有显著加性效应的QTL,加性效应对表型贡献率为0.58%~11.57%。其中4个主效QTL即控制总植物碱的qTPA14、烟碱的qNIC14、假木贼碱的qANAB14和新烟草碱的qANAT14,均可以解释相应性状10%以上的表型变异,且均位于14号连锁群上。6个QTL具有显著的加性与环境互作效应,对表型贡献率为0.80%~1.81%。5对QTL具有显著加性-加性上位性效应,对表型的贡献率为0.15%~2.31%。2对QTL具有显著的上位性与环境互作效应,对表型的贡献率为0.81%~1.16%。研究结果为进一步分离候选基因、解析遗传机理和促进烟草生物碱性状分子改良奠定了基础。

Removal of refractory organics and heavy metals in landfill leachate concentrate by peroxi-coagulation process

Landfill leachate is a complex effluent and it is difficult to deal with. Electrochemical methods have been considered as a promising alternative technology for treatment of landfill leachate with refractory organic contaminants and heavy metals. Peroxi-coagulation(PC)process with iron anode and modified graphite felt cathode was developed for efficient landfill leachate concentrate treatment. Compared to electro-Fenton(EF) and electrocoagulation(EC) processes, the PC process was more cost-effective due to the combined action of·OH oxidation and iron hydroxides coagulation. A maximal TOC removal of 77.2% ± 1.4% was obtained after 360 min at initial p H = 5.0 and current density of 10 m A/cm^(2). After the PC process, concentrations of all seven heavy metals in the final effluents were below the allowable emission limits given by the present regulatory standards. The method preference for heavy metal removal was PC > EC > EF. Based on the three-dimensional fluorescence spectroscopy coupled with regional integration analysis during the PC treatment, the florescence peaks of both humic acids and fulvic acids disappeared after treatment for 360min. Decreasing trends were observed in the fluorescent regional standard volumes for aromatic protein Ⅰ(31.4%), aromatic protein Ⅱ(63.7%), fulvic acid-like(69.5%), soluble microbial by-product-like(75%) and humic acid-like regions(76.3%). The results indicate that comparing to the EF and EC process, the PC process provide a promising and more cost-effective alternative for the treatment of landfill leachate concentrate.

基于时序InSAR技术的中贵天然气管道天水市段沿线滑坡隐患识别与形变分析

中贵天然气管道天水市段沿线地处黄土高原,地貌类型多样、构造活跃、复杂地质条件下发育多种具有隐蔽性和潜伏性的地质灾害,其中滑坡灾害对中贵天然气管道的安全危害极大,因此,对中贵天然气管道天水段沿线滑坡隐患进行有效识别与分析具有重要意义。采用Sentinel-1A升降轨卫星数据,基于时序合成孔径雷达干涉测量(synthetic aperture radar interferometry,InSAR)对中贵天然气天水段沿线滑坡隐患进行了解译识别、现场复核、发育特征与典型滑坡形变分析。研究区共识别17处滑坡隐患点,现场复核最终确定13处,其余4处为人类工程活动区,其中常沟村和磨峪沟村滑坡形变受降雨影响。统计发现研究区管道沿线滑坡多发生在坡度40°~50°,高差400~600 m,东北坡向和距管道100~150 m范围内,且岩土体强度较低,地层以第四系全新统冲洪积层(Q_(4)^(a1+p1))为主。结果表明:联合升降轨的时序InSAR技术可以有效识别管道沿线滑坡隐患,为油气管线的安全运营及今后油气管道选线提供重要的科学依据和参考。

Non-invasive chromosome screening for embryo preimplantation using cell-free DNA

Preimplantation genetic testing (PGT) is a widely adopted screening method that can be performed to identify and select embryos with normal ploidy;however, PGT relies on embryo biopsy, that is, polar body, embryo cells, or trophectoderm biopsy, to obtain embryonic DNA, increase its technical limitations. Studies have indicated that biopsy may have an influence on the quality and development of embryos, and increase the chance of abnormal epigenetic modifications. Therefore, non-invasive PGT (niPGT) detection of cell-free DNA (cfDNA) has gradually become a hot research topic in the field of assisted reproduction. Studies showed cfDNA could be detected in blastocyst fluid and spent culture medium (SCM) in vitro cultured embryos. The cfDNA collection requires less skill and makes lower risk to embryos. Some studies have been conducted to evaluate the feasibility of SCM-based niPGT approaches. When comparing the ploidy consistency of cfDNA in SCM, its consistency to the conventional PGT for aneuploidies results fluctuated widely, it is critical to recognize the factors influencing accuracy. These contradictory results may be related to factors such as the difference in SCM sampling methods and sampling time, and the definition of consistency. In this review, we aimed to comprehensively summarize how researchers use embryonic cfDNA to conduct niPGT detection. It also systematically reviews the factors affecting the accuracy of the test and its underlying issues, as well as prospective applications. We hope to provide a basis for future niPGT research and a useful reference for the standardized operation of niPGT that can be widely applied in clinical practice.

Comparison of cell wall changes of two different types of apple cultivars during fruit development and ripening

Fruit development and ripening is a complex procedure(Malus×domestica Borkh.)and can be caused by various factors such as cell structure,cell wall components,and cell wall hydrolytic *** our study,we focused on the variations in fruit firmness,cell wall morphology and components,the activity of cell wall hydrolytic enzymes and the expression patterns of associated genes during fruit development in two different types of apple cultivars,the hard-crisp cultivar and the loose-crisp *** this paper,the aim was to find out the causes of the texture variations between the different type *** wall materials(CWMs),hemicellulose and cellulose content were strongly associated with variations in fruit firmness during the fruit *** content of water soluble pectin(WSP)and chelator soluble pectin(CSP)gradually increased,while the content of ionic soluble pectin(ISP)showed inconsistent trends in the four *** activities of polygalacturonase(PG),β-galactosidase(β-gal),cellulase(CEL),and pectate lyase(PL)gradually increased in four *** the activities of PG,β-gal,and CEL were higher in‘Fuji’and‘Honeycrisp’fruit with the fruit development,while the activity of PL of‘Fuji’and‘Honeycrisp’was lower than that of‘ENVY’and‘Modi’.Both four cultivars of fruit cells progressively became bigger as the fruit expanded,with looser cell arrangements and larger cell *** to the qRT-PCR,the relative expression levels of MdACO and Mdβ-gal were notably *** study showed that there were large differences in the content of ISP and hemicellulose,the activity of PL and the relative expression of Mdβ-gal between two different types of apple cultivars,and these differences might be responsible for the variations in the texture of the four cultivars.
