Downregulation of NADPH-cytochrome P450 reductase via RNA interference increases the susceptibility of Acyrthosiphon pisum to desiccation and insecticides

Nicotinamide adenine dinucleotide phosphate(NADPH)-cytochrome P450 reductase(CPR)is involved in the metabolism of endogenous and exogenous substances,and detoxification of insecticides.RNA interference(RNAi)of CPR in certain insects causes developmental defects and enhanced susceptibility to insecticides.However,the CPR of Acyrthosiphon pisum has not been characterized,and its function is still not understood.In this study,we investigated the biochemical functions of A.pisum CPR(ApCPR).ApCPR was found to be transcribed in all developmental stages and was abundant in the embryo stage,and in the gut,head,and abdominal cuticle.After optimizing the dose and silencing duration of RNAi for downregulating ApCPR,we found that ApCPR suppression resulted in a significant decrease in the production of cuticular and internal hydrocarbon contents,and of cuticular waxy coatings.Deficiency in cuticular hydrocarbons(CHCs)decreased the survival rate of A.pisum under desiccation stress and increased its susceptibility to contact insecticides.Moreover,desiccation stress induced a significant increase in ApCPR mRNA levels.We further confirmed that ApCPR participates in CHC production.These results indicate that ApCPR modulates CHC production,desiccation tolerance,and insecticide susceptibility in A.pisum,and presents a novel target for pest control.

Multiplexing modulation design optimization and quality evaluation of BDS-3 PPP service signal

The Precise Point Positioning(PPP)service of BeiDou-3 Navigation Satellite System(BDS-3)is implemented on its Geostationary Earth Orbit(GEO)satellites.However,its signal design is limited by the actual power of satellite and other conditions.Furthermore,the design needs to fully consider the compatibility of diferent service phases.Starting from the actual state of the BDS-3 GEO satellite,this paper studies the multiplexing modulation of the BDS PPP service signal that is based on the Asymmetric Constant Envelope Binary Ofset Carrier(ACE-BOC)technique and proposes several feasible schemes for this signal.Comparison and optimization of these techniques are made from the aspects of transmission efciency,multiplexing efciency,and service forward compatibility.Based on the Type-III ACE-BOC multiplexing modulation technique,phase rotation and intermodulation reconstruction techniques are proposed to suppress the intermodulation interference issue.Finally,a signal based on improved ACE-BOC multiplexing is designed.The quality of the proposed signal was continuously monitored and tested using large-diameter antennas.The evaluation results show that the power spectrum deviation of the signal is 0.228 dB,the correlation loss is 0.110 dB,the S-curve slope deviation is 1.558%on average,the average length diference between the positive/negative chip and the ideal chip is only 0.0006 ns,and the coherence between the carrier and the pseudo code is 0.082°.All quality indicators are satisfactory,indicating that the proposed signal multiplexing modulation technique is an ideal solution that meets all the requirements of the design constraints,and can achieve efcient information broadcasting and forward compatibility of the BDS PPP service.

The M_(S)6.9 Menyuan earthquake in Qinghai Province,west China is the largest earthquake by far in 2022.The earthquake occurs in a tectonically active region,with a background b-value of 0.87 within 100 km of the epicenter that we derived from the unified catalog produced by China Earthquake Networks Center since late 2008.Field surveys have revealed surface ruptures extending 22 km along strike,with a maximum ground displacement of 2.1 m.We construct a finite fault model with constraints from In SAR observations,which showed multiple fault segments during the Menyuan earthquake.The major slip asperity is confined within 10 km at depth,with the maximum slip of 3.5 m.Near real-time back-projection results of coseismic radiation indicate a northwest propagating rupture that lasted for~10 s.Intensity estimates from the back-projection results show up to a Mercalli scale of IX near the ruptured area,consistent with instrumental measurements and the observations from the field surveys.Aftershock locations(up to January 21,2022)exhibit two segments,extending to~20 km in depth.The largest one reaches M_(S)5.3,locating near the eastern end of the aftershock zone.Although the location and the approximate magnitude of the mainshock had been indicated by previous studies based on paleoearthquake records and seismic gap,as well as estimated stressing rate on faults,significant surfacebreaching rupture leads to severe damage of the high-speed railway system,which poses a challenge in accurately assessing earthquake hazards and risks,and thus demands further investigations of the rupture behaviors for crustal earthquakes.

Determination and comparison of M， M， m， M， M， M， and M（GNSS） for the 22 May 2021 Mw 7.3 Madoi， Qianghai， China earthquake

The 2021 Madoi M7.3 earthquake in Qinghai is a major earthquake that occurred in the Bajankara block of Qinghai-Tibet Plateau in the past 30 years， which spatially filled the seismogenic gap in the eastern section of the northern boundary of the block. Here we determined the values of M， M， m， M， M， M， and M （GNSS） by abundant regional and global seismic and geodetic observations， which is 6.61， 7.43， 7.18， 7.33， 7.43， and 7.38， respectively. To compare the time efficiency and stability of different magnitude scales， we generated a real-time environment， to iteratively determine the magnitudes over elapsed times. Some methods such as m， M， Mgave considerable variations of as large as 0.5 units for the determined magnitudes with elapsed time， as more data were included. Others such as Mand Mwere very stable with increasing data over time. The systematic calculations of various magnitude scales in this study quantitively evaluated the stability and accuracy of those methods， shading light on the adaptability and applicability of different magnitude scales.

Water resource utilization characteristics and driving factors in the Hainan Island

The scarcity of water resources caused by the unique topography and uneven rainfall distribution in Hainan Island has become a major factor restricting local development. In order to provide effective and scientific reference basis for the overall water resource utilization status and solving this problem, this study calculated the water resource utilization situation of Hainan Island from 2017 to 2021 in detail using methods including water resource ecological footprint analysis. Furthermore, a spatial correlation analysis was conducted to examine the island's water resource utilization characteristics, and the driving factors behind the changes in water resource utilization over the past five years were analyzed using the LMDI model. The results show that:(1) During the study period, the water resource ecological footprint in Hainan Island exhibited a slow growth trend, while the ecological carrying capacity showed a downward tendency.The per capita ecological deficit of water resources remains relatively high, and the water consumption per 10 000 yuan GDP in the whole land continues to decrease, indicating that the overall pressure on water resource demand remains high with significant regional differences accompanied by the efficiency of water resource utilization steadily improving at the same time;(2) Agricultural water use accounts for the highest proportion in the entire water use structure, while ecological water use represents the smallest share, with a year-on-year increase, indicating that Hainan Island highlights the agricultural development and is increasingly conscious of the ecological environment;(3) Significant spatial differentiation in water resource utilization characteristics exists in Hainan Island, with the western region being a hot spot aggregation area for per capita water resource ecological footprint, per capita ecological carrying capacity of water resources, water consumption per 10 000 yuan GDP, while it is a cold spot cluster area for per capita ecological deficit of water resourc

为降低六硝基六氮杂异伍兹烷(CL-20)的机械感度,提高其综合性能,以3-氨丙基三乙氧基硅烷(APS)为改性剂对CL-20改性,以氧化石墨烯(GO)为包覆材料,利用静电自组装方法对改性CL-20炸药进行表面包覆,制备得到CL-20@GO核壳复合材料;采用水接触角测试、扫描电子显微镜(SEM)和X射线光电子能谱(XPS)等对样品进行形貌表征;利用差示扫描量热仪(DSC)测试其热性能,并测试了其机械感度。结果表明,经质量分数5%的APS溶液改性的CL-20成功引入氨基基团,亲水性效果最佳;GO层包覆改性CL-20完整,致密性强;与原料CL-20相比,CL-20@GO核壳复合材料的活化能提高了63.0kJ/mol,撞击感度(H 50)由13.0cm提升至23.5cm,摩擦感度由100%降至24%,表明采用静电自组装的GO涂层可以明显降低CL-20的感度。

Insights into the Temporal Gene Expression Pattern in Lymantria dispar Larvae During the Baculovirus Induced Hyperactive Stage

Baculoviruses are effective biological control agents for many insect pests. They not only efficiently challenge the host immune system but also make them hyperactive for better virus dispersal. Some investigations have focused on the viral mechanisms for induction of such altered response from the host. However, there are no current studies monitoring changes in gene expression during this altered phenotype in infected larvae. The L. dispar multiple nucleopolyhedrovirus(Ld MNPV) induces hyperactivity in third instar L. dispar larvae at 3-days post infection(dpi), to continued till 6 dpi. The transcriptome profiles of the infected and uninfected larvae at these time points were analyzed to provide new clues on the response of the larvae towards infection during hyperactivity. Gene ontology enrichment analysis revealed, most of the differentially expressed genes(DEGs) were involved in proteolysis, extracellular region, and serine-type endopeptidase activity. Similarly, Kyoto Encyclopedia of Genes and Genome enrichment analysis showed maximum enrichment of 487 genes of the signal transduction category and neuroactive ligand–receptor interaction sub-category with 85 annotated genes. In addition, enrichment map visualization of gene set enrichment analysis showed the coordinated response of neuroactive ligand–receptor interaction genes with other functional gene sets, as an important signal transduction mechanism during the hyperactive stage. Interestingly all the DEGs in neuroactive ligand–receptor interactions were serine proteases, their differential expression during the hyperactive stage correlated with their conceivable involvement in disease progression and the resulting altered phenotype during this period. The outcome provides a basic understanding of L. dispar larval responses to Ld MNPV infection during the hyperactive stage and helps to determine the important host factors involved in this process.

Imaging the rupture process of the 10 January 2018 Mw7.5 Swan island, Honduras earthquake

The 10 January 2018 Mw7.5 Swan island,Honduras earthquake occurred on the Swan island fault,which is a transform plate boundary between the North American and Caribbean plates.Here we back-project the rupture process of the earthquake using dense seismic stations in Alaska,and find that the earthquake ruptured at least three faults(three stages)for a duration of~40 s.The rupture speed for the longest fault(stage 3)is as fast as 5 km/s,which is much faster than the local shear wave velocity of~4 km/s.Supershear rupture was incidentally observed on long and straight strike-slip faults.This study shows a supershear rupture that occured on a strike-slip fault with moderate length,implying that supershear rupture might commonly occur on large strike-slip earthquakes.The common occurrence of supershear rupture on strike-slip earthquakes will challenge present understanding of crack physics,as well as strong ground motion evaluation in earthquake engineering.

Magnitude of the 23 January 2018 M7.9 Alaska Earthquake Estimated from Local Dense Seismic Records in Alaska

We apply a novel method to estimate the magnitude of the 23 January 2018 M7.9 Alaska earthquake using seismic stations recorded at local to regional distances in Alaska, US. We determine the source duration from back-projection results derived from the Alaska stations in a relatively compact azimuth range. Then we calculate the maximum P-wave displacements recorded on a wide azimuth range at distances of 8 to 15 degrees. Combining the source duration and the maximum P-wave displacements, we obtain magnitudes of 7.86–8.03 for the 23 January 2018 earthquake in 3–5 min, very close to the Mw 7.9 determined by the USGS and GCMT. This example validates the new approach for determining magnitude of large earthquakes using local to regional stations, and its time efficiency that magnitudes of large earthquakes can be accurately estimated within in 3–5 min after origin time. Therefore, further application of this new method would help accurate estimation of size of earthquakes that occur offshore and might cause tsunami hazards.
