the vascular network of the circulatory system plays a vital role in maintaining homeostasis in the human *** this paper,a novel modular microfluidic system with a vertical two-layered configuration is developed to generate largescale perfused microvascular networks in *** two-layer polydimethylsiloxane(PDMS)configuration allows the tissue chambers and medium channels not only to be designed and fabricated independently but also to be aligned and bonded *** method can produce a modular microfluidic system that has high flexibility and scalability to design an integrated platform with multiple perfused vascularized tissues with high *** medium channel was designed with a rhombic shape and fabricated to be semiclosed to form a capillary burst valve in the vertical direction,serving as the interface between the medium channels and tissue *** and anastomosis at the vertical interface were successfully achieved by using different combinations of tissue chambers and medium *** large-scale microvascular networks were generated and quantified in terms of vessel length and *** leakage of the perfused 70-kDa FItC-dextran confirmed the lumenization of the microvascular networks and the formation of tight vertical interconnections between the microvascular networks and medium channels in different structural *** platform enables the culturing of interconnected,large-scale perfused vascularized tissue networks with high density and scalability for a wide range of multiorgan-on-a-chip applications,including basic biological studies and drug screening.

Wedelolactone from Eclipta prostrata(L)*** inflammation and improves insulin resistance

Objective:to evaluate the effects of wedelolactone,a major flavonoid from Vietnamese Eclipta prostrata(L)L.,on inflammation and insulin ***:Wedelolactone was extracted from the leaves of Vietnamese Eclipta prostrata(L.)*** methanol by *** effects of wedelolactone on lipopolysaccharide(LPS)-induced cytokine production,reactive oxygen species(ROS)generation,and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase activities in Raw 264.7 cells were measured by enzyme-linked immunosorbent assay(ELISA),specific immunofluorescent dyes and luminometric analysis,*** addition,its effects on glucose uptake and the protein expression of insulin receptor substrate 1(IRS1)and glucose transporter 4(GLUt4)were examined in 3t3-L1 cells by immunofluorescent dyes and Western ***:Wedelolactone at 30μg/mL significantly inhibited LPS-induced production of tumor necrosis factor-α,interleukin(IL)-6,and IL-8(Pth no noticeable effects on IL-10 *** also reduced ROS generation and NADPH oxidase activities in LPS-stimulated Raw 264.7 cells(Pthermore,wedelolactone showed anti-insulin resistance activity,as evidenced by improved glucose uptake and the upregulated expression of IRS1 and GLUt4 in 3t3-L1 cells(Ptone exhibits anti-inflammation and anti-insulin resistance effects,which may be used for the treatment of diabetes and inflammation-associated diseases.

A hierarchical system to predict behavior of soil and cantilever sheet wall by data-driven models

the study proposes a framework combining machine learning(ML)models into a logical hierarchical system which evaluates the stability of the sheet wall before other *** study uses the hardening soil(HS)model to develop a 200-sample finite element analysis(FEA)database,to develop the ML ***,a system containing three trained ML models is proposed to first predict the stability status(random forest classification,RFC)followed by 1)the cantilever top horizontal displacement of sheet wall(artificial neural network regression models,RANN1)and 2)vertical settlement of soil(RANN2).the uncertainty of this data-driven system is partially investigated by developing 1000 RFC models,based on the application of random sampling technique in the data splitting *** on the distribution of the evaluation metrics reveals negative skewed data toward the 1.0000 *** implies a high performance of RFC on the database with medians of accuracy,precision,and recall,on test set are 1.0000,1.0000,and 0.92857,*** regression ANN models have coefficient of determinations on test set,as high as 0.9521 for RANN1,and 0.9988 for RANN2,*** parametric study for these regressions is also provided to evaluate the relative insight influence of inputs to output.

Poroelastodynamic responses and elastic moduli of a transversely isotropic porous cylinder under forced deformation test

Existing transversely isotropic poroelastodynamics solutions are limited to infinite domains and without experimental validation. Furthermore, there is a lack of analytical simulations for the elastic moduli dispersion of fluid-saturated porous cylinders. to address these three limitations and investigate the mechanisms of moduli dispersion, we present the analytical solutions of the poromechanical responses and the elastic moduli dispersion of a transversely isotropic, fluid-saturated, finite porous cylinder subjected to a forced deformation test. through an example, we demonstrate the effects of loading frequency, boundary conditions, and material's anisotropy, dimension, and permeability on the responses of pore pressure,force, displacement, and dynamic elastic moduli of the cylinder. the specimen's responses are significantly influenced by the frequency of the applied load, resulting in a drained state at low frequencies and an undrained state at high frequencies. At high frequencies, the sample behaves identically for an open or a closed lateral boundary, and permeability has insignificant effects. the dynamic elastic moduli are mainly controlled by the loading frequency and the ratio of the sample's radius to its height. Lastly,we show excellent matches between the newly derived analytical solution and laboratory measurements on one clay and two shale samples from Mont terri.

Recent advancements in stem cell technology open a new door for patients suffering from diseases and disorders that have yet to be *** cell-based therapy,including human pluripotent stem cells(hPsCs)and multipotent mesenchymal stem cells(MSCs),has recently emerged as a key player in regenerative *** are defined as self-renewable cell types conferring the ability to differentiate into various cellular phenotypes of the human body,including three germ *** are multipotent progenitor cells possessing self-renewal ability(limited in vitro)and differentiation potential into mesenchymal lineages,according to the International Society for Cell and Gene therapy(ISCt).this review provides an update on recent clinical applications using either hPSCs or MSCs derived from bone marrow(BM),adipose tissue(At),or the umbilical cord(UC)for the treatment of human diseases,including neurological disorders,pulmonary dysfunctions,metabolic/endocrine-related diseases,reproductive disorders,skin burns,and cardiovascular ***,we discuss our own clinical trial experiences on targeted therapies using MsCs in a clinical setting,and we propose and discuss the MSC tissue origin concept and how MSC origin may contribute to the role of MSCs in downstream applications,with the ultimate objective of facilitating translational research in regenerative medicine into clinical *** mechanisms discussed here support the proposed hypothesis that BM-MSCs are potentially good candidates for brain and spinal cord injury treatment,At-MSCs are potentially good candidates for reproductive disorder treatment and skin regeneration,and UC-MsCs are potentially good candidates for pulmonary disease and acute respiratory distress svndrometreatment.

Collagen provides tissue strength and structural *** fication of the orientated dispersion of collagen fibers is an important factor when studying the mechanical properties of the *** this study,for the first time,a new method for rapid characterization of the collagen fiber orientations of the cervix using linearly polarized light colposcopy is presented.A total of 24 colposcopic images were captured using a cross-polarized imaging system with white LED light *** the preprocessing stage,the Red channel of the RGB image was chosen,which contains no information of the blood vessels because of the low-absorption of blood cells in the red ***,which is an ImageJ plug-in,was used to estimate the local orientation of the collagen *** result shows that in the nonpregnant cervix,the middle zone(Zone 2)has circumferentially aligned collagen fibers while the inner zone(Zone 1)has randomly *** collagen fiber dispersion in Zone 2 is much smaller than that in Zone 1 at all four quadrants region(anterior,posterior,left,and right quadrant).this new analysis technique could potentially combine with diagnostic tools to provide a quantitative platform of collagen fibers in the clinic.

BACKGROUND twin reversed arterial perfusion(tRAP) sequence is an extremely rare congenital anomaly in monochorionic(MC) *** condition is characterized by a malformed fetus(acardiac twin) without cardiac activities being perfused by a structurally normal one(pump twin) via an artery-to-artery anastomosis in a reverse *** SUMMARY We described the first case of tRAP to receive laser surgery in *** 26-wk pregnancy was originally misdiagnosed in another hospital as MC twins with single intrauterine fetal *** admission to our center,the diagnosis was amended to a 26-wk tRAP sequence stage *** acardiac twin was 7.5 cm at the longest length,the ratio of the weight of the acardiac twin to the weight of the pump twin was more than 90%,the pump twin showed fetal distress with absent diastolic flow in umbilical artery of pump twin,and the peak systolic velocity in the middle cerebral artery = 1.6 Mo *** performed emergency laser photocoagulation of the acardiac twin’s umbilical *** surgery,we successfully maintained the pregnancy for 8 wk and ended it electively by cesarean section at 34 wk of gestation due to rupture of *** tRAP should be appropriately diagnosed and treated early to avoid complications of the pump *** laser photocoagulation is a new and effective treatment for this condition.

Path planning is a prevalent process that helps mobile robots find the most efficient pathway from the starting position to the goal position to avoid collisions with *** this paper,we propose a novel path planning algorithm-Intermediary RRt*-PSO-by utilizing the exploring speed advantages of Rapidly exploring Random trees and using its solution to feed to a metaheuristic-based optimizer,Particle swarm optimization(PSO),for fine-tuning and *** Phase 1,the start and goal trees are initialized at the starting and goal positions,respectively,and the intermediary tree is initialized at a random unexplored region of the search *** trees were grown until one met the other and then merged and re-initialized in other unexplored *** the start and goal trees merge,the first solution is found and passed through a minimization process to reduce unnecessary *** 2 begins by feeding the minimized solution from Phase 1 as the global best particle of PSO to optimize the *** simulating two special benchmark configurations and six practice configurations with special cases,the results of the study concluded that the proposed method is capable of handling small to large,simple to complex continuous environments,whereas it was very tedious for the previous method to achieve.
