Antimicrobial resistance(AMR) represents a substantial threat to global public health, complicating the treatment of.common inf.ctions and leading to prolonged illness and escalated healthcare expenses. To ef.ectively combat AMR, timely and accurate detection is crucial f.r AMR surveillance and individual-based therapy. Phenotypic antibiotic resistance testing(AST) has long been considered the gold standard in clinical applications, serving as the f.undation f.r clinical AMR diagnosis and optimized therapy. It has signif.cantly contributed to ensuring patients′ health and the development of.novel antimicrobials. Despite advancements in automated culture-based AST technologies,inherent limitations impede the widespread use of.phenotypic AST in AMR surveillance. Genotypic AST technologies of.er a promising alternative option, exhibiting advantages of.rapidity, high sensitivity, and specif.city. With the continuous advancement and expanding applications of.genotypic AST technologies, such as microf.uidics, mass spectrometry, and high-resolution melting curve analysis, new vigor has been injected into the development and clinical implementation of.genotypic AST technologies. In this narrative review, we discuss the principles, applications, and advancements of.emerging genotypic AST methods in clinical settings. The comprehensive review aims to highlight the signif.cant scientif.c potential of.emerging genotypic AST technologies in clinical AMR diagnosis, providing insights to enhance existing methods and explore novel approaches.

In the pathogenesis of.major depressive disorder, chronic stress-related neuroinf.ammation hinders f.vorable prognosis and antidepressant response. Mitochondrial DNA may be an inf.ammatory trigger, af.er its release f.om stress-induced dysf.nctional central nervous system mitochondria into peripheral circulation. This evidence supports the potential use of.peripheral mitochondrial DNA as a neuroinf.ammatory biomarker f.r the diagnosis and treatment of.major depressive disorder. Herein, we critically review the neuroinf.ammation theory in major depressive disorder, providing compelling evidence that mitochondrial DNA release acts as a critical biological substrate, and that it constitutes the neuroinf.ammatory disease pathway. Af.er its release, mitochondrial DNA can be carried in the exosomes and transported to extracellular spaces in the central nervous system and peripheral circulation. Detectable exosomes render encaged mitochondrial DNA relatively stable. This mitochondrial DNA in peripheral circulation can thus be directly detected in clinical practice. These characteristics illustrate the potential f.r mitochondrial DNA to serve as an innovative clinical biomarker and molecular treatment target f.r major depressive disorder. This review also highlights the f.ture potential value of.clinical applications combining mitochondrial DNA with a panel of.other biomarkers, to improve diagnostic precision in major depressive disorder.

The presence of.endogenous neural stem/progenitor cells in the adult mammalian brain suggests that the central nervous system can be repaired and regenerated af.er ***,whether it is possible to stimulate neurogenesis and reconstruct cortical layers II to VI in non-neurogenic regions,such as the cortex,remains *** this study,we implanted a hyaluronic acid collagen gel loaded with basic f.broblast growth f.ctor into the motor cortex immediately f.llowing traumatic *** f.ndings reveal that this gel ef.ectively stimulated the prolif.ration and migration of.endogenous neural stem/progenitor cells,as well as their dif.erentiation into mature and f.nctionally integrated ***,these new neurons reconstructed the architecture of.cortical layers II to VI,integrated into the existing neural circuitry,and ultimately led to improved brain *** f.ndings of.er novel insight into potential clinical treatments f.r traumatic cerebral cortex injuries.

Molecular materials showing synergetic coupling of.near-inf.ared （NIR） f.uorescence and spin-crossover （SCO） are promisingly applicable in in-vivo bioimaging,temperature sensing,and spintronic *** the related study f.ces f.rmidable *** this work we graf. the NIR f.uorophore xanthene onto the dipyridyl substituted triazole ligand and construct a new f.based SCO material,{f.Ⅱ（ddxc-abpt）2[N（CN）2]2}(1,ddxc-abpt:（E）-4-(((3,5-di（pyridin-2-yl）-4H-1,2,4-triazol-4-yl)imino)methyl)-N,N-diethyl-2,3-dihydro-1H-xanthen-6-amine).Magnetic studies indicate that compound 1 exhibits thermally induced SCO property in both the solid and solution ***-temperature f.uorescence emission spectra reveal a remarkable 7-f.ld increase in photoluminescence intensity upon the transition f.om the low-spin state at 110 K to the high-spin state at 280 K,which is the record of.all reported coupled f.uorescence-SCO ***-temperature ultraviolet-visible （UV-Vis）absorption spectra show a signif.cant change in absorption intensity within the f.uorescence emission window,consistent with the f.rster resonance energy transf.r （f.ET） ***-dependent density f.nctional theory （TD-Df.） calculations demonstrate that the strong coupling is attributed to disrupted energy transf.r between the xanthene f.uorophore and high-spin *** work provides a f.asible approach f.r expanding the synergetic SCO-f.uorescence materials to the NIR-Iwavelength region,and hopef.lly contributes to the applications of.tunable NIR-SCO molecule-based sensors and devices.

Immobilizing highly dispersed Au nanoparticles onto titanium silicalite-1 (TS-1) by a simple impregnation method under light is still a challenging *** this work,a high photostable Au precursor protected by thiol ligands was f.rstly synthesized,and the as-prepared Au precursor displayed excellent stability under light f.r over 30 days,while the Au precursor without thiol ligands f.rmed precipitates af.er just 2 h under ***,the Au precursor protected by thiol ligands was immobilized on the external surf.ce of.uncalcined TS-1 (i.e.,TS-1-B) by the incipient wetness impregnation method (IWI method) to prepare Au/TS-1-B *** activity of.as-prepared Au/TS-1-B catalyst in the hydro-oxidation of.propane to acetone was 1.4 times higher than that of.the catalyst prepared by the deposition-precipitation urea method (DPU method).The structures of.the Au/TS-1-B catalysts prepared by dif.erent methods were analyzed by multiple *** to the Au/TS-1-B catalyst prepared by the DPU method,the Au/TS-1-B catalyst synthesized by the IWI method exhibited a narrower size distribution of.Au nanoparticles and had more small Au particles ( f.r its superior ***,the ef.ects of.gold loadings,reaction temperature and Si/Ti molar ratio on the catalytic perf.rmance of.the Au/TS-1-B catalyst prepared by the IWI method were also *** addition,the Au/TS-1-B catalyst prepared by the IWI method also exhibited excellent stability f.r over 140 h.

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant inf.station, we developed a highly ef.icient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of.PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modif.cation of.PBQ and were used, through both protein pull-down assay and conf.cal laser scanning microscopy imaging, to identif. and conf.rm that phosphof.uctokinase(Pf.) in the apple snail is the primary target protein of.PBQ. f.rther studies revealed that PBQ successf.lly inhibits the activity,content, and expression of.Pf. in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its ef.icient eradication. These f.ndings of.er a promising direction f.r developing next-generation Pf.-targeted snail-killing pesticides. f.rthermore, the strategy of.combining pull-down assays with optical imaging used in this work provides a novel approach f.r the exploration of.mechanisms of.other pesticide candidates.

Metabolites and metabolism-related gene expression prof.les in skeletal muscle change dramatically under obesity, aging and metabolic disease. Since obese and lean pigs are ideal models f.r metabolic research. Here, we compared metabolome and transcriptome of.Longissimus dorsi(LD) muscle between Taoyuan black(TB, obese) and Duroc(lean) pigs at dif.erent ages. We def.ned the “window phase” of.intramuscular f.t(IMf. deposition in TB pig, which has signif.cantly higher IMf.than Duroc pig. Our results displayed discrepant lipid composition and dif.erent expression genes(DEGs) enriched in lipid metabolism, and both metabolome and transcriptome analyses revealed stronger energy expenditure and more active amino acid and protein metabolism in Duroc pig. 10 up-and 51 down-regulated biomarker metabolites with age-and breed-specif.city were identif.ed. Potential secretory metabolites, including organic acid(f.maric acid, succinate,malic acid, and gamma-aminobutyric acid), amino acid(L-lysine, and L-glutamic acid), lipid(2-hydroxyisovaleric acid, and L-carnitine) were demonstrated a signif.cant correlation with IMf.deposition. Our research highlights the huge dif.erence of.metabolic spectrum in skeletal muscle between obese and lean model and muscle-derived secretory metabolites might act as an ambassador of.intercellular communication to regulate systematic metabolism.

法治宣传教育对于实现全民守法、构建法治社会具有重要意义。尤其对于保障推进新时代全面依法治国,具有无可替代的思想启蒙、文化传播与创新引领作用。自1985年开始,我国法治宣传教育工作已经实施了近40年,陆续开展了8个五年普法规划,在推进实践工作的内容、形式、主体、机制等多个“形式创新”层面积累了诸多有益经验,有效提升了法治宣传教育的深度与广度,大幅提升了公民法治素养和水平。然而在更为抽象与深层的基础理论层面,围绕普法工作的本质属性、原则理念以及客观规律等问题,实践理论工作的“实质创新”仍然存在不足,一定程度上抑制了普法主体与学法对象的主动性、积极性与能动性。为持续有效地健全完善法治宣传教育,未来应坚持“形式创新”与“实质创新”相结合,在稳步推进“形式创新”的基础上,重点推进法治宣传教育在基础理论层面的“实质创新”。
