Emerging role of liquid biopsy in rat sarcoma virus mutated metastatic colorectal cancer:A case report

BACKGROUND In patients with metastatic colorectal cancer(mCRC),the treatment options are limited and have been proved to be affected by rat sarcoma virus(RAS)mutational *** RAS wild-type(wt)patients,the combination of antiepidermal growth factor receptor(EGFR)monoclonal antibodies with chemotherapy(CT)is more effective than CT *** the other hand,RAS-mutated patients are not eligible for treatment with anti-EGFR *** SUMMARY Eleven patients with initially RAS-mutated mCRC were followed from diagnosis to may *** the time of cell-free DNA determination,five patients had undergone one CT line,five patients had undergone two CT lines,and one patient had undergone three CT lines(all in combination with bevacizumab).At the second and third treatment lines[second line(2L),third line(3L)],patients with neo-RAS wt received a combination of CT and *** neo-RAS wt patients treated with anti-EGFR,our findings indicated an increase in progression-free survival for both 2L and 3L(14.5 mo,p=0.119 and 3.9 mo,p=0.882,respectively).Regarding 2L overall survival,we registered a slight increase in neo-RAS wt patients treated with anti-EGFR(33.6 mo vs 32.4 mo,p=0.385).At data cut-off,two patients were still alive:A RAS-mutated patient undergoing 3L treatment and a neo-RAS wt patient who received 2L treatment with anti-EGFR(ongoing).CONCLUSION Our case series demonstrated that monitoring RAS mutations in mCRC by liquid biopsy may provide an additional treatment line for neo-RAS wt patients.

Indonesian peatlands are a fragile ecosystem,and to protect it,growing Liberica coffee is a promising way for both the environment and the *** study aimed to evaluate the performance of the liberica coffee on peatland with different water tables and develop an improved cultivation system in this *** study area was in Tanjung Jabung Barat District,Jambi province,*** plant age is greater than 15 years and the average plant density was about 1000 trees/*** study was conducted in two *** 1st stage was a survey to identify and characterize smallholder liberica coffee farming at three peatland zones,namely 0-100 m,200-300 m,and>400 m from the principal drainage canal,and the 2nd stage was a field experiment to test the effectiveness of amendments in improving liberica coffee growth and improving degraded *** treatments were arranged in a randomized complete block design with four replications,including manure(M),at the rate of 10 t/hm^(2);lime(L),3 t/hm^(2);peat surface elevation(p),and a farmer’s practice(Control)without manure and/or lime as a *** research revealed that liberica coffee can not tolerate high water table as it may stimulate disease incidence and cause low-quality ***,recycling of organic C of about 17.14 t/(hm^(2)·a)through cherry residue application and litter compensated part of the loss of organic C through CO_(2)emissions,coffee bean removal,and maintaining peat fertility for sustainable *** low yield of(0.70±0.12)t/(hm^(2)∙a)coffee bean could be increased to(0.87±0.24)and(0.94±0.14)t/(hm^(2)∙a)by adding 3 t/(hm^(2)∙3a)of lime or 10 t/(hm^(2)∙a)of manure,*** research revealed that water table management and amendments are two main factors in liberica coffee farming on *** is of great significance to study the cultivation technology of coffee in peatland.

Since the time of Darwin(1859)and Wallace(1869),islands have been regarded by scientists as a prime target for scrutinizing the forces that may influence evolution and diversification and important elements in biogeographic *** research aims to scrutinize whether and to what extent the composition and structure of past mammal insular faunas and their changes through time may provide sound clues for inferring the paleogeographical evolution of a *** a case study,I critically analyzed the dynamics shown by the plio-pleistocene mammalian fauna of 3 Western Mediterranean insular districts,the Balearic Islands,Sardinia and Sicily,each characterized by its own peculiar paleobiogeographical evolutionary *** revision of faunas and the critical analysis of the dispersal ability of the ancestors of island settlers have allowed hypothesizing on the time and mode of island *** results obtained confirm that the early isolation of the Balearic Islands from the mainland led to the establishment of an endemic fauna since the pre-Messinian Miocene(?Astaracian European Land Mammal Age,MN7/8),and that Sardinia has definitely been isolated since the pliocene,although dispersal events led to some faunal turnovers during the *** addition,the results suggest for Sicily a complex,still imperfectly disentangled history of alternate phases of complete separation and sporadic,more or less difficult connections with southern Italy.

The research describes for the first time a possible case of pituitary gigantism in fossil mammals,precisely in *** pathology was detected in 2 long bones(tibia and metatarsus)belonging to an individual of an unusual large size found at the Bate cave(Rethymnon,Northern Crete).It formed the basis of Candiacervus major,the largest among the endemic deer species recorded in the pleistocene-Early Holocene of *** and histomorphological examinations highlighted a reduction in cortical bone thickness and the presence of wide lacunae inside of the bone *** pathological conditions suggest a pituitary gigantism diagnosis also supported by some morphological evidence,such as the extremely elongated distal part of the metatarsal diaphysis,the proportionally small proximal epiphysis,and some bone *** diagnosis of a case of pituitary gigantism as presumed responsible for the extraordinary elongation of the tibia and the metatarsal bone is intriguing as they are,respectively,the paratype and the holotype of the *** species represents a case of a deviation from the“island rule”in pleistocene large *** new evidence recommends a taxonomic and nomenclatural revision of this *** main outcomes of this research are as follows:(i)a case of pituitary gigantism is described for the first time in an extinct mammal;(ii)it is underlined that paleohistology may provide interesting clues for disentangling taxonomic and nomenclatural issues;(iii)one of the very few cases of gigantism in insular mammals is being questioned.

Given the failure to develop disease-modifying therapies for Alzheimer’s disease(AD),strategies aiming at preventing or delaying the onset of the disease are being *** the debate regarding whether depression is an etiological risk factor or a prodrome of AD rages on,a key determining factor may be the timing of depression onset in older *** is increasing evidence that untreated early-onset depression is a risk factor and that late-onset depression may be a catalyst of cognitive *** from animal studies have shown a beneficial impact of selective serotonin reuptake inhibitors on pathophysiological biomarkers of AD including amyloid burden,tau deposits and *** humans,studies focusing on subjects with a prior history of depression also showed a delay in the onset of AD in those treated with most selective serotonin reuptake ***,which has strong anticholinergic properties,was associated with increased mortality and mixed effects on amyloid and tau deposits in mice,as well as increased odds of developing AD in *** most of the data regarding selective serotonin reuptake inhibitors is promising,findings should be interpreted cautiously because of notable methodological heterogeneity between *** is thus a need to conduct large scale randomized controlled trials with long follow up periods to clarify the dose-effect relationship of specific serotonergic antidepressants on AD prevention.

A wave–tide–circulation coupled model is used to simulate upwelling off the south coast of Java, Indonesia. The results show that the vertical velocity off East Java is stronger than other parts in this area. The strongest vertical velocity is located approximately at 80 m *** annual averaged values of upwelling are 2.3 9 10-6and 1.06 9 10-5m/s for south of West Java and south of East Java, respectively. The vertical velocity from the model shows that upwelling off West Java has seasonal variability, while it is quite steady and strong off East *** numerical experiments show that the wind is not the dominant factor for the steady upwelling off the southern part of East Java. It is then hypothesized that the Indonesian Throughflow(ITF) may be responsible for the upwelling. To test this hypothesis, two scenarios are implemented, both of which block the outflow of the *** study shows that the ITF plays a key role in the formation of East Java upwelling. The effect of the ITF can account for about 55 %–65 % of the upwelling.

Frailty is an issue of paramount importance for cardiologists,because of the aging of patients admitted to hospital for acute coronary syndrome(ACS)and the straight relationship between aging and *** tools have been provided in this setting,in order to objectively assess frailty status,but important questions are still *** are conflicting data about a unique definition of frailty in subjects with cardiovascular diseases,the timing to perform a frailty evaluation in the context of an acute myocardial infarction,the mean to assess frailty in these patients and the usefulness of the information derived from the frailty *** results from the analysis of several items and a multidomain evaluation including laboratory values,clinical data and physical performance assessment is required for a comprehensive frailty ***,regardless of the frailty tool,the prevalence of frailty in older ACS patients is high and it could add important information to the decision-making process about invasive strategy,the multivessel disease management,dual antiplatelet therapy and secondary prevention *** present overview tries to summarize the current knowledge about the definition and prevalence of frailty in older adults admitted to hospital for ACS,suggesting how frailty assessment may improve the management of older ACS patients.
