The maturation and function of osteoblasts(OBs)rely heavily on the reversible phosphorylation of signaling *** date,most of the work in OBs has focused on phosphorylation by tyrosyl kinases,but little has been revealed about dephosphorylation by protein tyrosine phosphatases(PTPases).SHP2(encoded by PTPN11)is a ubiquitously expressed ***11 mutations are associated with both bone and cartilage manifestations in patients with Noonan syndrome(NS)and metachondromatosis(Mc),although the underlying mechanisms remain ***,we report that SHP2 deletion in bone gamma-carboxyglutamate protein-expressing(Bglap+)bone cells leads to massive osteopenia in both trabecular and cortical bones due to the failure of bone cell maturation and enhanced osteoclast activity,and its deletion in Bglap+chondrocytes results in the onset of enchondroma and osteochondroma in aged mice with increased tubular bone ***,SHP2 was found to be required for osteoblastic differentiation by promoting RUNX2/OSTERIX signaling and for the suppression of osteoclastogenesis by inhibiting STAT3-mediated RANKL production by osteoblasts and *** findings are likely to explain the compromised skeletal system in NS and Mc patients and to inform the development of novel therapeutics to combat skeletal disorders.

chondrocytes and osteoblasts differentiate from a common mesenchymal precursor, the osteochondroprogenitor（OcP）, and help build the vertebrate skeleton. The signaling pathways that control lineage commitment for OcPs are incompletely understood. We asked whether the ubiquitously expressed protein-tyrosine phosphatase SHP2（encoded by Ptpn11） affects skeletal lineage commitment by conditionally deleting Ptpn11 in mouse limb and head mesenchyme using ＂cre-lox P＂-mediated gene ***2-deficient mice have increased cartilage mass and deficient ossification, suggesting that SHP2-deficient OcPs become chondrocytes and not osteoblasts. consistent with these observations, the expression of the master chondrogenic transcription factor SOX9 and its target genes Acan, col2a1, and col10a1 were increased in SHP2-deficient chondrocytes, as revealed by gene expression arrays, q RT-PcR, in situ hybridization, and immunostaining. Mechanistic studies demonstrate that SHP2 regulates OcP fate determination via the phosphorylation and SUMOylation of SOX9, mediated at least in part via the PKA signaling pathway. Our data indicate that SHP2 is critical for skeletal cell lineage differentiation and could thus be a pharmacologic target for bone and cartilage regeneration.

Bats are the second-most diverse group of mammals in the world,and bat flies are their main ***,significant knowledge gaps remain regarding these antagonistic interactions,especially since diverse factors such as seasonality and host sex can affect their network ***,we explore the influence of such factors by comparing species richness and composition of bat flies on host bats,as well as specialization and modularity of bat–bat fly interaction networks between seasons and adult host *** captured bats and collected their ectoparasitic flies at 10 sampling sites in the savannahs of AmapáState,northeastern region of the Brazilian *** female bats being more parasitized and recording greater bat fly species richness in the wet season,neither relationship was statistically *** pooled network could be divided into 15 compartments with 54 links,and all subnetworks comprised>12 *** total number of links ranged from 27 to 48(for the dry and wet seasons,respectively),and female and male subnetworks had 44 and 41 links,*** values were very low for the pooled network and for all *** results revealed higher bat fly species richness and abundance in the wet season,whereas specialization and modularity were higher in the dry ***,the subnetwork for female bats displayed higher specialization and modularity than the male ***,both seasonality and host sex contribute in different ways to bat–bat fly network *** studies should consider these factors when evaluating bat–bat fly interaction networks.

Effects of Papyrus Plants (cyperus papyrus) on the Physicochemical Parameters and Nutrient Levels of Water and Sediments in Yala Swamp Wetland in Western Kenya

Yala swamp wetland is the largest fresh water wetland ecosystem in Kenya supporting a broad biodiversity. It comprises of River Yala, the Yala swamp, Lakes Kanyaboli, Namboyo and Sare, and a portion of Lake Victoria neighboring the swamp. Approximately 2300 ha of land have been reclaimed and has been used for large-scale agriculture resulting in mass destruction of papyrus to create room for framing. Papyrus are known to be important in phytoremediation but despite this role information is limited, lacks supportive evidence and the empirical aspect on the levels of these pollutants in relation to the papyrus biomass is limited. The study is aimed at determining the effects of cyperus papyrus on the water and sediment quality in Yala Swamp wetland. Six sampling sites were purposefully selected to monitor the variations of the physicochemical parameters (temperature, dissolved oxygen: DO, pH, biological oxygen demand: BOD, total suspended solids: TSS, turbidity, electrical conductivity: Ec and total dissolved solids: TSS) and the levels selected nutrients (phosphorus and nitrates) in water and sediments as River Yala flows through Dominion Farms, Lake Sare which surrounded by cyperus papyrus and finally into Lake Victoria. The samples were collected in triplicates during the wet and dry seasons (May and September 2015 respectively). The physicochemical parameters were measured in situ using a Hydrolab muiltimeter while the nutrients were analyzed using UV/Vis spectroscopy. Statistical analysis was done using SAS V9.0 software. The mean temperature was 26.19˚c ± 0.71˚c, DO: 3.72 ± 1.02 Mg/l, BOD: 3.9 ± 0.32 Mg/l, pH: 7.52 ± 0.17, TDS: 109 ± 86.33, Ec: 173.26 ± 13.8 μS/cm, TSS: 12.42 ± 18.51 Mg/l and Turbidity: 12.29 ± 10.03 NTU. The values varied significantly at P 0.05 among all the sites. The results show that papyrus is useful in maintaining the required levels of physicochemical parameters. Forest composition and red oak (Quercus sp.) response to elevation gradients across greentree reservoirs

Dialectical Thermodynamics’Solution to the conceptual Imbroglio That Is the Reversible Path

According to the second law of thermodynamics, as currently understood, any given transit of a system along the reversible path proceeds with a total entropy change equal to zero. The fact that this condition is also the identifier of thermodynamic equilibrium, makes each and every point along the reversible path a state of equilibrium, and the reversible path, as expressed by a noted thermodynamic author, “a dense succession of equilibrium states”. The difficulties with these notions are plural. The fact, for example, that systems need to be forced out of equilibrium via the expenditure of work, would make any spontaneous reversible process a consumer of work, this in opposition to common thermodynamic wisdom that makes spontaneous reversible processes the most efficient transformers of work-producing-potential into actual work. The solution to this and other related impasses is provided by Dialectical Thermodynamics via its previously proved notion assigning a negative entropy change to the energy upgrading process represented by the transformation of heat into work. The said solution is here exemplified with the ideal-gas phase isomerization of butane into isobutane.
