AIM:To investigate whether the iron stores regulator hepcidin is implicated in colon cancer-associated anae- mia and whether it might have a role in colorectal car- cinogenesis. METHODS: Mass spectrometry (MALDI-TOF MS and SELDI-TOF MS) was employed to measure hepcidin in urine collected from 56 patients with colorectal cancer. Quantitative Real Time RT-PCR was utilised to determine hepcidin mRNA expression in colorectal cancer tissue. Hepcidin cellular localisation was determined using im- munohistochemistry. RESULTS: We demonstrate that whilst urinary hepcidin expression was not correlated with anaemia it was posi- tively associated with increasing T-stage of colorectal cancer (P < 0.05). Furthermore, we report that hepcidin mRNA is expressed in 34% of colorectal cancer tissue specimens and was correlated with ferroportin repres- sion. This was supported by hepcidin immunoreactivity in colorectal cancer tissue. CONCLUSION: We demonstrate that systemic hepcidin expression is unlikely to be the cause of the systemic anaemia associated with colorectal cancer. However, we demonstrate for the first time that hepcidin is expressed by colorectal cancer tissue and that this may represent a novel oncogenic signalling mechanism.

背景：肠系膜上动脉综合征（superior mesenteric artery syndrome，SMAS）是脊柱畸形矫形术后较常见的并发症。近年来，对此并发症易发因素的研究主要集中在患者的身高和体重方面。迄今为止，未见对脊柱侧凸患者的畸形严重程度、侧凸类型、侧凸纠正率与术后并发SMAS相关性的研究报告，而且所有文献中对SMAS的研究均缺乏对照组资料。本研究探讨脊柱畸形的纠正率与术后并发SMAS的相关性。我们的假设是畸形纠正率越高，术后并发SMAS的风险越大。方法：这是一项历时5年的病例对照性研究。主要评估指标为SMAS的发生。预测变量包括患者一般资料；术前身高、体重和体重指数；脊柱畸形参数，包括弧度大小、Lenke弯度分型和畸形纠正率；手术方面包括手术入路、估计手术失血量和有无术中低血压。结果：364例行脊柱畸形矫形手术的患者中，17例术后发生SMAS。从行脊柱畸形矫形手术但未发生SMAS的患者中随机抽取34例作为对照组。SMAS组中，8例行分期手术（对照组仅1例，P〈0．001），9例行前后路联合手术。SMAS组中，7例同时行胸廓成形术（对照组仅2例，P〈0．001）。SMAS组与对照组的一般资料比较，差异无统计学意义。与对照组相比，SMAS组患者身高更矮（平均为7．1cm，P=0．03），体重更轻（平均为11．5kg，P=0．001），体重指数更小（P=0．003）；凸侧侧屈片上胸弯残留的度数较大[平均高12°（SMAS组为45°，对照组为33°），P=0．015]，胸弯的纠正率较低（平均低11％，P=0．025），腰弯顶椎偏移更明显（88％的SMAS患者的Lenke腰弯矫正指数是B或C而不是A，而对照组为61％，P〈0．008）。多因素Logistic回归分析得出各可靠预测因素的优势比：分期手术次数（优势比，31．0）、腰弯矫正指数（优势比，9．06）、体重指数（优势比，7．75）和胸弯的僵硬性（优势比，6．67），可作为SMAS发生的最有价值的预测指标。结论：术前找出并发SMAS的危险因素，并尽可能改善患者的营养状况，可以减少脊柱畸形矫形术后SMAS的发生。可信水平：治疗性研究，旦级。进一步可信度参见作者介绍。

Chondrocytes and osteoblasts differentiate from a common mesenchymal precursor, the osteochondroprogenitor（OCP）, and help build the vertebrate skeleton. The signaling pathways that control lineage commitment for OCPs are incompletely understood. We asked whether the ubiquitously expressed protein-tyrosine phosphatase SHP2（encoded by Ptpn11） affects skeletal lineage commitment by conditionally deleting Ptpn11 in mouse limb and head mesenchyme using ＂Cre-lox P＂-mediated gene ***2-deficient mice have increased cartilage mass and deficient ossification, suggesting that SHP2-deficient OCPs become chondrocytes and not osteoblasts. Consistent with these observations, the expression of the master chondrogenic transcription factor SOX9 and its target genes Acan, Col2a1, and Col10a1 were increased in SHP2-deficient chondrocytes, as revealed by gene expression arrays, q RT-PCR, in situ hybridization, and immunostaining. Mechanistic studies demonstrate that SHP2 regulates OCP fate determination via the phosphorylation and SUMOylation of SOX9, mediated at least in part via the PKA signaling pathway. Our data indicate that SHP2 is critical for skeletal cell lineage differentiation and could thus be a pharmacologic target for bone and cartilage regeneration.
