Nerve allograft regeneration and immunological tolerance of rats with sciatic nerve transplantation after intragastric pretreatment with ultraviolet B-irradiated donor splenocytes

BACKGROUND： Nerve allograft rejection is an unavoidable problem for nerve allografts. Traumatic peripheral nerve injuries are commonly reconstructed using autogenous nerve grafts. However, this form of reconstruction is limited by insufficient autologous nerves for large gap repairs and by morbidity at the nerve donor site. OBJECTIVE： To examine sciatic nerve regeneration and immune tolerance reaction after intragastric administration of ultraviolet B-irradiated （UVB） donor splenocytes. DESIGN, TIME AND SETTING： A complete randomized grouping design and controlled experiment. The experiments were conducted in the Department of Orthopedics, the First Affiliated Hospital to Shanxi Medical University, China, between March and October 2007. MATERIALS： Fourteen adult male SD rats and fourteen male Wistar rats, weighing 250-300 g, were randomly matched as donors and acceptors. A further seven male SD rats （weight 250-300 g, age 12-16 weeks） were used for nerve isografts. Immune preparations and the Epics XL flow cytometer were purchased from B-D Company, USA. A computer-assisted electromyograph machine was provided by Keypoint P, Dantel Company, Denmark. METHODS： Splenocytes from Wistar rats were isolated, purified, and cultured, and then irradiated with ultraviolet B. In the first control group （Group 1）, the SD rats received a syngeneic SD nerve isograft. In the second control group （Group 2）, the SD rats received a nerve allograft from Wistar rats without pretreatment. In the oral-tolerance treated group （Group 3）, the SD recipient rats were inoculated with 2.5×10^7 Lewis UVB-irradiated donor splenocyte cells by intestinal tract administration at seven days before transplantation. MAIN OUTCOME MEASURES：（1） The recent end and the middle and distal end of the transplanted nerve were cut at 8 and 12 weeks after operation. Recovery of nerve regeneration was measured with HE staining using the total number, density, and diameter of the nerve fibers. （2） The level of CD25＋T

Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2

Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and ***, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. Together, our study unravels a novel mTORC2-PPAR-γ-PKM2 pathway in immune-metabolic regulation of early thymocyte development.

Proper control of B cell growth and metabolism is crucial for B-cell-mediated immunity,but the underlying molecular mechanisms remain incompletely *** this study,Sin1,a key component of mTOR complex 2(mTORC2),specifically regulates B cell growth and *** ablation of Sin1 in B cells reduces the cell size at either the transitional stage or upon antigen stimulation and severely impairs ***1 deficiency also severely impairs B-cell proliferation,antibody responses,and anti-viral *** the molecular level,Sin1 controls the expression and stability of the c-Myc protein and maintains the activity of mTORC1 through the Akt-dependent inactivation of GSK3 and TSC1/2,***,our study reveals a novel and specific role for Sin1 in coordinating the activation of mTORC2 and mTORC1 to control B cell growth and metabolism.

Absorbable bandage wrapping in treatment of severe blast liver injury： a miniature swine model

Background The aim of this research was to evaluate the feasibility and efficacy of absorbable bandage wrapping in the treatment of cases of severe liver trauma. Methods Electric firecrackers were detonated in 16 miniature swine to produce a severe blast liver injury. After fluid resuscitation, the animals were randomly divided into two groups （n=-8 each） and were either treated with absorbable bandage wrapping of the injured lobe of liver （Group B） or hepatic Iobectomy （Group H）. Time to hemostasis, blood loss during the treatment period, and other parameters were compared, including postoperative serum total bilirubin （TB）, alanine aminotransferase （ALT）, and aspartate aminotransferase （AST）. Results Blood loss during the treatment period was significantly lower in Group B than that in Group H （（81.3±26.0） ml vs. （130.8±2.9.5） ml, P=0.0031）. Serum AST and ALT were transiently increased post-surgically. These transient increases were significantly higher in Group B. No difference in time to hemostasis was noted （（8.70±2.27） minutes vs. （10.28±1.93） minutes, P=-0.1559） in Groups B and H, respectively. Two pigs were humanely euthanized 28 days post-surgically and the wrapped liver lobes appeared atrophies. Microscopically, there was evidence of emerging and mature fibrous tissue. Conclusion Absorbable bandaqe wrappinQ is both feasible and effective in the treatment of severe blast liver injury.

在水为介质的条件下,以三聚氯氰(TCT)与赖氨酸(Lys)为原料,经缩合反应制备N,N?-二-(4,6-二氯-[1,3,5]-三嗪基)-2,6-二氨基己酸(简称DTDHA),利用FTIR和1HNMR对其结构进行了表征。通过正交实验考察了原料物质的量之比、缚酸剂种类、反应时间对产物收率的影响,得出较适宜的反应条件为:n(三聚氯氰)∶n(赖氨酸)=3.0∶1.0,KOH做缚酸剂,反应时间6 h,在该条件下DTDHA产率为91%。并将产物应用于生皮鞣制,以鞣制坯革的收缩温度为考察指标,采用单因素实验对鞣制工艺条件进行了优化。得到适宜的鞣制工艺条件为:鞣剂用量20%(以灰皮质量为基准,下同),温度从25℃开始每隔2 h阶梯上升15℃,最终达到55℃,鞣制总时间8h,在该条件下,坯革的收缩温度为85.6℃。该鞣制工艺下合成产物具有良好的鞣制效应,是一种新型的有机合成鞣剂。
