为满足建筑师对北京CBD核心区Z15地块中国尊大楼外形的多方案比选及工程师对结构性能优化的需要,在设计中引入了结构参数化设计。通过对塔楼整体结构体系几何逻辑的定义,结合参数化插件Grasshopper和图形软件Rhinoceros,实现了不同建筑条件下几何模型的自动化建立。再通过奥雅纳二次开发工具将构件截面尺寸、材料属性、荷载形式及大小等信息赋予图形软件Rhinoceros中的几何单元,并导入GSA,ETABS,SAP2000等软件中,生成计算分析模型。以上模型成果可进一步应用到与建筑和机电专业的信息共享、CAD出图、有限元节点分析和BIM设计之中,实现了短时间内结构性能的优化和整体工作效率的提高。

Manifestation of Diabetes Mellitus Type 1: A Questionnaire to Evaluate the Acceptance of Initial Intravenous Therapy

For families suddenly confronted with the diagnosis of a lifelong chronic disease, implementing a continuous iv. infusion represents restriction of movement and possibly also a psychological burden. We designed a questionnaire to evaluate the perception of pediatric patients and their parents for this treatment as part of a diabetes manifestation. Patients and their parents treated in the diabetes outpatient clinic at children’s university hospital in Mainz were asked for their opinion about “iv. infusion” and “frequency of blood taking” during their first stay in the hospital upon manifestation of diabetes. They assigned scores from 1 to 10 for the following parameters: “delivery of diagnosis”, “communication by doctor”, “execution of training”, “atmosphere during diabetes training”, “frequency of blood taking”, “iv. infusion” and “start of sc. insulin application”. Parents gave relatively high ratings. Parameters such as “frequency of blood taking” were rated with 6.1 on average. Ultimately, “frequency of blood taking”, “delivery of diagnosis” (6.11) and “iv. infusion of insulin” (6.5) scored lower than for instance “atmosphere of diabetes training” (7.95). Children awarded scores of 3.72 for “iv. infusion”, on average, whereas they scored 7.15 for the “doctor’s communication” and 7.28 for “diabetes training”. Asked if a decision in favor of a subcutaneous insulin injection right at the beginning of the therapy would have been preferable, parents were undecided. Therefore it cannot be concluded that an instant subcutaneous therapy has psychological advantages or disadvantages. The patients themselves (aged 12 - 17 years) were undecided, too, (58.3%) when asked for their preference of the subcutaneous insulin regime right at the beginning of the therapy. Nevertheless, the continuous infusion of insulin iv. was rated poorly by them. Transcription factor TFIIA is controlled by complex regulatory networks including proteolysis by the protease Taspase 1, though the full impact of cleavage remains elusive. Here, we demonstrate that in contrast to the general assumption,de novoproduced TFIIA is rapidly confined to the cytoplasm via an evolutionary conserved nuclear export signal (NES, amino acids21VINDVRDIFL30), interacting with the nuclear export receptor Exportin-1/chromosomal region maintenance 1 (Crm1). Chemical export inhibition or genetic inactivation of the NES not only promotes TFIIA’s nuclear localization but also affects its transcriptional activity. Notably, Taspase 1 processing promotes TFIIA’s nuclear accumulation by NES masking, and modulates its transcriptional activity. Moreover, TFIIA complex formation with the TATA box binding protein (TBP) is cooperatively enhanced by inhibition of proteolysis and nuclear export, leading to an increase of the cell cycle inhibitor p16INK, which is counteracted by prevention of TBP binding. We here identified a novel mechanism how proteolysis and nuclear transport cooperatively fine-tune transcriptional programs.

Effects of Emotional State on Processing Conceptual Mismatches: ERPs in a Picture-Sentence Matching Task

Event-related potential (ERP) studies demonstrated that emotional state immediately affects the processing of different linguistic mismatches (e.g., semantic and syntactic mismatches) in sentences. Furthermore, mood has been shown to impact discourse processing. In these studies, a strong expectancy was evoked by a linguistic context. In the present study, a strong expectancy was induced by a visual (non-linguistic) context paired with a sentence. A spatial array was followed by a sentence that matched (□○—The square stands in front of the circle) or mismatched the picture (intra-dimensional mismatch: □○—The square stands behind the circle;extra-dimensional mismatch: □○—The square stands above the circle). The main question was whether mood effects on the processing of visually induced expectancies are similar or different from mood effects on the processing of linguistically induced expectancies. To this aim, we presented conceptual (mis)matches that differed in saliency: intra-dimensional vs. extra-dimensional mismatches which are both reported to elicit an N2b/N400 and P600, reflecting reanalysis. EEG was recorded while participants read for comprehension. Mood (happy vs. sad) was effectively induced by film clips. Neither for N2b/N400 nor for P600, an interaction between mood and match was observed. Thus, in contrast with studies investigating the effects of pure linguistic mismatches (linguistic context paired with unexpected lexical item), mood did not modulate the processing of conceptual mismatches. However, a main effect of match revealed different ERP patterns for the two mismatches. While an increase in N2b/ N400 occurred for the intra-dimensional and extra-dimensional mismatches, a P600 only occurred to the extra-dimensional (more salient) mismatches. This finding is taken as support that being in an emotional state (positive or negative mood) influences processes of reanalysis.
