Causal genetic regulation of DNA replication on immune microenvironment in colorectal tumorigenesis: Evidenced by an integrated approach of trans-omics and GWAS

The interplay between DNA replication stress and immune microenvironment alterations is known to play a crucial role in colorectal tumorigenesis,but a comprehensive understanding of their association with and relevant biomarkers involved in colorectal tumorigenesis is lacking.To address this gap,we conducted a study aiming to investigate this association and identify relevant biomarkers.We analyzed transcriptomic and proteomic profiles of 904 colorectal tumor tissues and 342 normal tissues to examine pathway enrichment,biological activity,and the immune microenvironment.Additionally,we evaluated genetic effects of single variants and genes on colorectal cancer susceptibility using data from genome-wide association studies(GWASs)involving both East Asian(7062 cases and 195745 controls)and European(24476 cases and 23073 controls)populations.We employed mediation analysis to infer the causal pathway,and applied multiplex immunofluorescence to visualize colocalized biomarkers in colorectal tumors and immune cells.Our findings revealed that both DNA replication activity and the flap structure-specific endonuclease 1(FEN1)gene were significantly enriched in colorectal tumor tissues,compared with normal tissues.Moreover,a genetic variant rs4246215 G>T in FEN1 was associated with a decreased risk of colorectal cancer(odds ratio=0.94,95%confidence interval:0.90–0.97,P_(meta)=4.70×10^(-9)).Importantly,we identified basophils and eosinophils that both exhibited a significantly decreased infiltration in colorectal tumors,and were regulated by rs4246215 through causal pathways involving both FEN1 and DNA replication.In conclusion,this trans-omics incorporating GWAS data provides insights into a plausible pathway connecting DNA replication and immunity,expanding biological knowledge of colorectal tumorigenesis and therapeutic targets.

The water soluble coal tar pitches(WS-CTPs)were successfully prepared and used to construct the MnO_(2)@C composite materials by a hydrothermal method.It is interestingly observed that the structures and morphologies of MnO_(2)@C materials can be controlled by controlling the dosages of WS-CTPs and KMnO4.Meanwhile,it is aware that MnO_(2)exists in the MnO_(2)@C materials in an amorphous state.Compared with MnO_(2),MnO_(2)@C materials output a remarkable improvement in electrochemical performance.For instance,MnO_(2)@C-0.3 shows the storage capacity at 965.7 mA h g^(−1)after 300 cycles at a current density of 0.1 A g^(−1).In addition,after 600 cycles at a current density of 1.0 A g^(−1),the storage capacity of MnO_(2)@C-0.3 still keeps 450.3 mA h g^(−1),indicating that MnO_(2)@C-0.3 owns tremendous cycle stability at a high current density.In view of the fact that the coal tar pitches possess great cost advantages,the strategy of using WS-CTPs as a carbon source to cover the metal oxides is a competitive way to expand the application of metal oxides in the fabrication of electrodes of LIBs.

Difference among the quality indices,chemical composition and frying performance of galangal flavored sunflower oil prepared by three methods

Flavored vegetable oils have attracted more and more attentions from consumers and researchers due to their high oxidative stability and sensory quality.In the present study,the quality indices,chemical composition and frying performance of galangal flavored sunflower oil prepared by infusion method(GFSO-I),combination pressing method(GFSO-C)and direct addition method(GFSO-A)were investigated.The results displayed that the preparation time,the levels for the acid value(AV),peroxide value(PV),p-anisidine value(AnV),total oxidation value(TOTOX),and the values for total polar compounds(TPC),thiobarbituric acid(TBA),K232 and K268 of GFSO-C were prominently low(P<0.05),compared with that of GFSO-I and GFSO-A,while the fatty acid composition of them were similar.In the frying of Chinese Maye(tradional Chinese food,fried dough stick)at 180°C for 30 h,the values for TPC,TBA,K232 and K268,polymer,viscosity and b*of GFSO-C were obviously low(P<0.05),while the value for L*was obviously high(P<0.05),compared with that of GFSO-I and GFSO-A.In the sensory evaluation,the flavor,taste,crispness and overall acceptance of Chinese Maye fried by GFSO-C were better than that of Chinese Maye fried by GFSO-I and GFSO-A.Consequently,GFSO-C is more suitable for cooking,and the combination pressing method is optimal for industrial production of GFSO.

中文医疗问诊文本中,由于口语化的不规则表达和专业术语的频繁出现,药物名称等实体难以被精准地识别出来。为了充分利用中文句子词间关系的重要作用,提出了一种用于增强全局信息的医学命名实体识别模型。模型利用注意力机制增强了词嵌入表征,并在使用双向长短时记忆网络的序列处理能力获取上下文信息的基础上,同时从两个方面丰富了句子的全局信息表示。其一是根据句法关系获取词语之间额外依赖关系构建了图卷积网络层用于丰富词间的依赖;其二是构建了辅助任务用于预测词间句法依赖关系的类别。在中文医疗问诊数据集上的实验结果表明,模型具有很好的竞争力,F1值达到94.54%。与其他模型相比,在药物和症状等实体类别的识别上取得了明显提高。在微博公开数据集上的实验也表明,模型具有通用领域的应用价值。

Hydrogen-assisted fracture features of a high strength ferrite-pearlite steel

Up to now, the exact reason of hydrogen-induced fracture for ferrite-pearlite(FP) steel is still not fully understood. This study presents detail observations of the feature beneath the fracture surface with the aim to reveal the hydrogen-induced cracking initiation and propagation processes. Slow strain rate tensile(SSRT) testing shows that the FP steel is sensitive to hydrogen embrittlement(HE). Focused ion beam(FIB)was used to prepare samples for TEM observations after HE fracture. The corresponding fractographic morphologies of hydrogen charged specimen exhibit intergranular(IG) and quasi-cleavage(QC) fracture feature. Pearlite colony, ferrite/pearlite(F/P) boundary and the adjacent ferrite matrix are found to be responsible for the initial HE fracture and the subsequent propagation. With increasing of the stress intensity factor, fracture mode is found to change from mixed IG and QC to entire QC feature which only occurs at the ferrite matrix. No crack is observed at the ferrite/cementite(F/C) interface. This may be mainly due to the limited pearlite lamella size and relatively low interface energy.

Optimization of different intensities of exercise preconditioning in protecting exhausted exercise induced heart injury in rats

This study was to optimize the exercise preconditioning(EP)intensity in protecting from exhaustive exercise-induced cardiac injury(EECI).A total of 98 male Sprague-Dawley rats were divided into 7 groups(n=14):the control group(C),the exhaustive exercise group(EE)and the EP+EE groups,which include the V10(53.0%VO_(2max)),V15(58.4%VO_(2max)),V20(67.0%VO_(2max)),V26(74.0%VO_(2max))and V30(80.0%VO_(2max))groups.Except the C group,the other groups were subjected to treadmill running.The serum contents of N terminal pro B type natriuretic peptide(NT-proBNP)and cardiac troponin I(cTn-I)were detected by the enzyme-linked immunosorbent assay method,ECG was recorded,heart function was detected by pressure volume catheter and the activities of mitochondrial electron transfer pathway(ET pathway)complexesⅠ,ⅡandⅣwere measured by high-resolution respiration instrument.Compared to the EE group,the EP groups have shown decrease of NT-proBNP and cTn-I,improvement of mitochondrial respiratory function and car-diac function.Compared to other EP groups,the V26 group has shown significant decrease of myocardial enzymes and improvement of mitochondrial function.The correlation analysis showed the EP effect was proportional to EP intensity in the range of 53.0%VO_(2max)-74.0%VO_(2max).High intensity and long duration of exhaustive exercise caused cardiac injury and EP could decrease serum level of NT-proBNP and cTn-I,improve electrical derangement and the left ventricular function,and raise the activities of ET pathway complexesⅠ,ⅡandⅣ.The protection of EP on EECI was improved as the EP intensity was increased from 53.0%VO_(2max)to 74.0%VO_(2max)and when EP intensity was 74.0%VO_(2max),the effect was the most obvious among all the setting EP groups.

高质量学前教育体系是高质量教育体系建设的基础,学前教师是实现学前教育普及普惠与优质发展的关键力量。公办园非在编教师人事制度改革有助于为占学前教师近八成的非在编教师的人事制度改革提供重要参考。充分考虑多元利益相关者的诉求有助于减少改革的阻力,提升改革的可行性与有效性。本研究运用问卷法和访谈法,着重探讨公办园非在编教师、园长、举办者、教育行政部门四类核心与重要利益相关主体对公办园非在编教师人事制度改革的主要诉求,并基于以上诉求,从创新编制管理机制、健全待遇经费保障机制、建立同工同酬制度、保障专业发展机会等方面提出改革完善公办园非在编教师人事制度的对策建议。

Geophysical prediction of organic matter abundance in source rocks based on geochemical analysis:A case study of southwestern Bozhong Sag,Bohai Sea,China

The Bozhong Sag is the largest petroliferous sag in the Bohai Bay Basin,and the source rocks of Paleogene dongying and Shahejie Formations were buried deeply.Most of the drillings were located at the structural high,and there were few wells that met good quality source rocks,so it is difficult to evaluate the source rocks in the study area precisely by geochemical analysis only.Based on the Rock-Eval pyrolysis,total organic carbon(TOC)testing,the organic matter(OM)abundance of Paleogene source rocks in the southwestern Bozhong Sag were evaluated,including the lower of second member of dongying Formation(E_(3)d2L),the third member of dongying Formation(E_(3)d_(3)),the first and second members of Shahejie Formation(E_(2)s_(1+2)),the third member of Shahejie Formation(E_(2)s_(3)).The results indicate that the E_(2)s_(1+2)and E_(2)s_(3)have better hydrocarbon generative potentials with the highest OM abundance,the E_(3)d_(3)are of the second good quality,and the E_(3)d2L have poor to fair hydrocarbon generative potential.Furthermore,the well logs were applied to predict TOC and residual hydrocarbon generation potential(S_(2))based on the sedimentary facies classification,usingΔlogR,generalizedΔlogR,logging multiple linear regression and BP neural network methods.The various methods were compared,and the BP neural network method have relatively better prediction accuracy.Based on the pre-stack simultaneous inversion(P-wave impedance,P-wave velocity and density inversion results)and the post-stack seismic attributes,the three-dimensional(3D)seismic prediction of TOC and S_(2)was carried out.The results show that the seismic near well prediction results of TOC and S_(2)based on seismic multi-attributes analysis correspond well with the results of well logging methods,and the plane prediction results are identical with the sedimentary facies map in the study area.The TOC and S_(2)values of E_(2)s_(1+2)and E_(2)s_(3)are higher than those in E_(3)d_(3)and E_(3)d_(2)L,basically consistent with the geochemical analysis resul

Exposure to poor indoor air conditions poses significant risks to human health, increasing morbidity and mortality rates. Soft measurement modeling is suitable for stable and accurate monitoring of air pollutants and improving air quality. Based on partial least squares (PLS), we propose an indoor air quality prediction model that utilizes variational auto-encoder regression (VAER) algorithm. To reduce the negative effects of noise, latent variables in the original data are extracted by PLS in the first step. Then, the extracted variables are used as inputs to VAER, which improve the accuracy and robustness of the model. Through comparative analysis with traditional methods, we demonstrate the superior performance of our PLS-VAER model, which exhibits improved prediction performance and stability. The root mean square error (RMSE) of PLS-VAER is reduced by 14.71%, 26.47%, and 12.50% compared to single VAER, PLS-SVR, and PLS-ANN, respectively. Additionally, the coefficient of determination (R2) of PLS-VAER improves by 13.70%, 30.09%, and 11.25% compared to single VAER, PLS-SVR, and PLS-ANN, respectively. This research offers an innovative and environmentally-friendly approach to monitor and improve indoor air quality.

Paleoenvironmental Characteristics of Paleogene Lacustrine Source Rocks in the Western Bozhong Sag,Bohai Bay Basin,China:Evidence from Biomarkers,Major and Trace Elements

The organic matter(OM)enrichment mechanisms and depositional environment characteristics of lacustrine source rocks in the western Bozhong Sag,Bohai Bay Basin in Northeast China remain controversial.To address these issues,based on Rock-Eval pyrolysis,kerogen macerals,H/C and O/C ratios,GC-MS,major and trace elements,the dongying Formation Member(Mbr)3(E_(3)d_(3)),the Shahejie Formation mbrs 1 and 2(E_(2)s_(1+2)),and the Shahejie Mbr 3(E_(2)s_(3))source rocks in the western Bozhong Sag were studied.The above methods were used to reveal their geochemical properties,OM origins and depositional environments,all of which indicate that E_(2)s_(1+2)and E_(2)s_(3)are excellent source rocks,and that E_(3)d_(3)is of the second good quality.E_(3)d_(3)source rocks were formed under a warm and humid climate,mainly belong to fluvial/delta facies,the E_(3)d_(3)sediments formed under weakly oxidizing and freshwater conditions.Comparatively,the depositional environments of E_(2)s_(1+2)source rocks were arid and cold climate,representing saline or freshwater lacustrine facies,and the sediments of E_(2)s_(1+2)belong to anoxic or suboxic settings with large evaporation and salinity.During the period of E_(2)s_(3),the climate became warm and humid,indicating the freshwater lacustrine facies,and E_(2)s_(3)was characterized by freshwater and abundant algae.Moreover,compared with other intervals,the OM origin of E_(3)d_(3)source rocks has noticeable terrestrial input.The OM origin of the E_(2)s_(1+2)and E_(2)s_(3)are mainly plankton and bacteria.Tectonic subsidence and climate change have affected the changes of the depositional environment in the western Bozhong Sag,thus controlling the distribution of the source rocks,the geochemical characteristics in the three intervals of lacustrine source rocks have distinct differences.Overall,these factors are effective to evaluate the paleoenvironmental characteristics of source rocks by biomarkers,major and trace elements.The established models may have positive implications for research of l
