Approximately 140 million people worldwide are homozygous carriers of APOE4(ε4),a strong genetic risk factor for late onset familial and sporadic Alzheimer’s disease(AD),91%of whom will develop AD at earlier age than heterozygous carriers and noncarriers.Susceptibility to AD could be reduced by targeted editing of APOE4,but a technical basis for controlling the off-target effects of base editors is necessary to develop low-risk personalized gene therapies.Here,we first screened eight cytosine base editor variants at four injection stages(from 1-to 8-cell stage),and found that FNLS-YE1 variant in 8-cell embryos achieved the comparable base conversion rate(up to 100%)with the lowest bystander effects.In particular,80%of AD-susceptibleε4 allele copies were converted to the AD-neutralε3 allele in humanε4-carrying embryos.Stringent control measures combined with targeted deep sequencing,whole genome sequencing,and RNA sequencing showed no DNA or RNA off-target events in FNLS-YE1-treated human embryos or their derived stem cells.Furthermore,base editing with FNLS-YE1 showed no effects on embryo development to the blastocyst stage.Finally,we also demonstrated FNLS-YE1 could introduce known protective variants in human embryos to potentially reduce human susceptivity to systemic lupus erythematosus and familial hypercholesterolemia.Our study therefore suggests that base editing with FNLS-YE1 can efficiently and safely introduce known preventive variants in 8-cell human embryos,a potential approach for reducing human susceptibility to AD or other genetic diseases.

Sub-10-nm-sized Au@AuxIr1-x metal-core/alloy-shell nanoparticles as highly durable catalysts for acidic water splitting

The absence of efficient and durable catalysts for oxygen evolution reaction（OER） is the main obstacle to hydrogen production through water splitting in an acidic ele ctrolyte.Here,we report a controllable synthesis method of surface IrOxwith changing Au/Ir compositions by constructing a range of sub-10-nm-sized core-shell nanocatalysts composed of an Au core and AuxIr1-xalloy shell.In particular,Au@Au0.43Ir0.57exhibits 4.5 times higher intrinsic OER activity than that of the commercial Ir/C.Synchrotron X-ray-based spectroscopies,electron microscopy and density functional theory calculations revealed a balanced binding of reaction intermediates with enhanced activity.The water-splitting cell using a load of 0.02 mgIr/cm2of Au@Au0.43Ir0.57as both anode and cathode can reach 10 mA/cm2at 1.52 V and maintain activity for at least194 h,which is better than the cell using the commercial couple Ir/C‖Pt/C（1.63 V,0.2 h）.

The General Compromise Value for Cooperative Games With Transferable Utility

The authors introduce the general compromise value for cooperative games with transferable utility.With respect to a set of potential payoffs of which the maximal and minimal potential payoff vectors are regarded as the upper and lower bounds for players,the unique pre-imputation lying on the straight line segment with these two vectors as the extreme points is defined as the general compromise value.Potential-consistency and maximal proportional property are introduced to characterize the general compromise value.

从冬枣果实中分离芽孢杆菌,通过对峙培养法筛选对炭疽病菌有显著拮抗活性的菌株,利用冬枣果实检测拮抗菌对炭疽病的防治效果并对拮抗菌进行种属鉴定,采用抑菌圈法及液相色谱一质谱联用技术分别对拮抗菌合成抑菌物质进行抑菌活性检测和结构鉴定。获得1株对枣炭疽病菌有显著拮抗活性的芽孢杆菌Tj-19,经鉴定为解淀粉芽孢杆菌(Bacillus amyloliquefaciens)。Tj-19菌体及其发酵上清液对冬枣果实炭疽病的防治效果分别达到74.9%和75.8%,主要抑菌物质为伊枯草菌素A和伊枯草菌素B。

为了研究以D5(十甲基环五硅氧烷)为介质的聚酯染色机理以制定合理的染色工艺,探讨了聚酯(PET)在D5中微观结构的变化。通过构建不同比例聚酯、D5、1-甲基萘共存的分子模型,采用分子动力学模拟溶剂、促进剂、温度对聚酯微观特性参数的影响。结果表明:分子模拟构建的纯PET体系的玻璃化转变温度(Tg)与实验测得的PET的Tg相吻合;加入D5后,PET的Tg明显下降;继续加入1-甲基萘,Tg进一步降低。单纯D5的加入并没有显著改善PET分子链的运动性能,而1-甲基萘与D5共存时,PET分子链运动性能显著提高。D5与1-甲基萘的存在增加了PET的总自由体积。分子模拟聚酯微观结构发现,D5、1-甲基萘有利于提升聚酯上染速率,降低染色温度,改善染色效果。

Predicting rock cracking is important for assessing the stability of underground engineering.The effects of the intersecting angle a and the distribution orientation angleβof intersecting fissures on the uniaxial compressive strength and the failure characteristics of sandstone containing intersecting fissures are investigated through laboratory experiments and two-dimensional particle flow code(PFC2D).The relationship between the mechanical properties of sandstone and the intersecting angle a and the distribution orientation angleβis analysed.Crack initiation forms and the final failure modes are then categorised and determined via empirical methods.In addition,the cracking processes of intersecting fissures with different a andβvalues are discussed.The results show that variations in the peak stress,peak strain,average modulus,and crack initiation stress of sandstone containing intersecting fissures show a“moth”shape in the space of the a-β-mechanical parameters.Two crack initiation forms are identified:inner tip cracking(usually accompanied by one outer tip cracking)and only outer tips cracking.Two failure modes are observed:(1)the main fracture planes are created at the inner tip and one outer tip,and(2)the main fracture planes are formed at the two outer tips.Two main crack evolution processes of sandstone containing intersecting fissures under uniaxial compression are found.Approaches for quickly determining the crack initiation form and the failure mode are proposed.The combination of the determination equations for the crack initiation form and the failure mode can be used to predict the crack evolution.The approach for determining the crack evolution processes is hence proposed with acceptable precision.
