Objective:PD-L1 and PD-L2 expression levels determine immune evasion and the therapeutic efficacy of immune checkpoint blockade.The factors that drive inducible PD-L1 expression have been extensively studied,but mechanisms that result in constitutive PD-L1 expression in cancer cells are largely unknown.Methods:DNA elements were deleted in cells by CRISPR/Cas9-mediated knockout.Protein function was inhibited by chemical inhibitors.Protein levels were examined by Western blot,mRNA levels were examined by real-time RT-PCR,and surface protein expression was determined by cellular immunofluorescence and flow cytometry.Immune evasion was examined by in vitro T cell-mediated killing.Results:We determined the core regions(chr9:5,496,378–5,499,663)of a previously identified PD-L1L2-super-enhancer(SE).Through systematic analysis,we found that the E26 transformation-specific(ETS)variant transcription factor(ETV4)bound to this core DNA region but not to DNA surrounding PD-L1L2SE.Genetic knockout of ETV4 dramatically reduced the expressions of both PD-L1 and PD-L2.ETV4 transcription was dependent on ERK activation,and BRAF/TAK1-induced ERK activation was dependent on extracellular signaling fromαvβ3 integrin,which profoundly affected ETV4 transcription and PD-L1/L2 expression.Genetic silencing or pharmacological inhibition of components of the PD-L1L2-SE-associated pathway rendered cancer cells susceptible to T cell-mediated killing.Conclusions:We identified a pathway originating from the extracellular matrix that signaled via integrin/BRAF/TAK1/ERK/ETV4 to PD-L1L2-SE to induce PD-L1-mediated immune evasion.These results provided new insights into PD-L1L2-SE activation and pathways associated with immune checkpoint regulation in cancer.

Feature Extraction to Polar Image

Some algorithms of feature extraction in existing literature studied for image processing was the gray image with one-dimensional parameter. However, some feature points’ extraction for three-dimensional color of polar image, such as the color edge extraction, inflection points, and so on, was urgently to be solved a polar color problem. For achieving quickly and accurately the color feature extraction to polar image, this paper proposed a similar region of color algorithm. The algorithm was compared to polar image, and the effect to color extraction was also described by the combination of the proposed and existing algorithms. Moreover, this paper gave the comparison of the proposed algorithm and an existing classical algorithm to extraction of color feature. These researches in this paper provided a powerful tool for polar image classification, color feature segmentation, precise recognition, and so on.

Application of FRS on Target Recognition

In order to let machine better imitate thinking method of people to perform recognition and classification for fuzzy and uncertain thing, this paper puts forward a fuzzy and rough association method to deal with the problem. However, the application of fuzzy rough sets (FRS) will be introduced mainly on pattern recognition. Some related theories on FRS would be discussed, and some fuzzy rough mathematical methods on pattern-recognition will be given. Then, concrete applications of FRS on image processing and recognition will be introduced. Simulation result signifies that this fuzzy and rough association method is not only fast but also closer to nature attribute of thing for processing and recognizing image by comparing with the single neural network and other recognition device. The recognition rate is about 95.78%.

建立及完善商业银行财务柔性维度的监管机制,对于防控银行业系统性风险具有重要意义.文章选取2010–2021年中国上市银行季度数据,构建面板回归模型对财务柔性对银行系统性风险的影响及其作用机制进行了实证分析.研究表明:1)财务柔性对银行系统性风险具有促进作用.相对于国有银行部门和经济下行期,财务柔性对银行系统性风险的促进作用仅存在于非国有银行部门和经济上行期.2)信贷过度扩张和表外业务在财务柔性与银行系统性风险的关系中承担着多重中介作用,财务柔性主要通过加剧信贷过度扩张与提高表外业务渠道来促进银行系统性风险,“财务柔性-信贷过度扩张/表外业务-银行系统性风险”的传导渠道均有效.3)资管新规与货币政策不确定性对两者关系具有非对称性调节作用.资管新规的实施会减弱财务柔性对银行系统性风险的促进作用,而货币政策不确定性提高则会加剧财务柔性对银行系统性风险的促进作用.4)区分财务柔性类型的进一步研究表明,现金柔性和债务柔性均对银行系统性风险具有促进作用.该成果将为提升银行财务柔性监管效率及防控中国银行业系统性风险提供重要的理论指导与决策参考.

为解决六角形三维编织血管支架的编织工艺复杂、开发周期长的问题,提出了基于MatLab编程的支架三维虚拟编织方法,以快速完成编织工艺开发。首先,基于六角形编织机底盘的结构特点,对底盘单元进行坐标数值化,从而可将支架编织的工艺和工序转换为不同时刻下携纱器在底盘上的XY平面坐标和运动信息以及纱线在芯棒上的高度坐标z值,并应用数值计算方法进行连接和拟合,建立纱线空间轨迹的数学模型,得到虚拟支架的三维基本形态。然后,将编织支架的三维形态展开为二维形态,分析二维平铺形态下单向支架沿螺旋方向缠绕的特点,应用线性方程组求解纱线的交织点坐标,并通过层内和层间的纱线交织关系判断得到各交织点的交织类型。最后,通过方程修正得到具有六角形编织特征的纱线波动方程,进一步建立了具有纱线立体交织特征的虚拟支架实体化数学模型。经不同编织工艺的支架虚拟仿真实验,将虚拟支架与其实物进行比较发现,虚拟支架均能准确直观地表达实物的几何特征和交织特征。由此验证了虚拟仿真建模的正确性,有利于六角形三维编织工艺的快速开发。

The application of indocyanine green in guiding prostate cancer treatment

Objective:Indocyanine green(ICG)with near-infrared fluorescence absorption is approved by the United States Food and Drug Administration for clinical applications in angiography,blood flow evaluation,and liver function assessment.It has strong optical absorption in the near-infrared region,where light can penetrate deepest into biological tissue.We sought to review its value in guiding prostate cancer treatment.Methods:All related literature at PubMed from January 2000 to December 2020 were reviewed.Results:Multiple preclinical studies have demonstrated the usefulness of ICG in identifying prostate cancer by using different engineering techniques.Clinical studies have demonstrated the usefulness of ICG in guiding sentinel node dissection during radical prostatectomy,and possible better preservation of neurovascular bundle by identifying landmark prostatic arteries.New techniques such as adding fluorescein in additional to ICG were tested in a limited number of patients with encouraging result.In addition,the use of the ICG was shown to be safe.Even though there are encouraging results,it does not carry sufficient sensitivity and specificity in replacing extended pelvic lymph node dissection during radical prostatectomy.Conclusion:Multiple preclinical and clinical studies have shown the usefulness of ICG in identifying and guiding treatment for prostate cancer.Larger randomized prospective studies are warranted to further test its usefulness and find new modified approaches.

A novel compression-induced twisting(CIT)-compliant mechanism was designed based on the freedom and constraint topology(FACT)method and manufactured by means of laser powder bed fusion(LPBF).The effects of LPBF printing parameters on the formability and compressive properties of the laserprinted CIT-compliant mechanism were studied.Within the range of optimized laser powers from 375 to 450 W and with the densification level of the samples maintained at above 98%,changes in the obtained relative densities of the LPBF-fabricated CIT-compliant mechanism with the applied laser powers were not apparent.Increased laser power led to the elimination of residual metallurgical pores within the inclined struts of the CIT mechanism.The highest dimensional accuracy of 0.2% and the lowest surface roughness of 20μm were achieved at a laser power of 450 W.The deformation behavior of the CIT-compliant mechanism fabricated by means of LPBF exhibited four typical stages:an elastic stage,a heterogeneous plastic deformation stage,a strength-destroying stage,and a deformation-destroying stage(or instable deformation stage).The accumulated compressive strain of the optimally printed CIT mechanism using a laser power of 450 W went up to 20% before fracturing,demonstrating a large deformation capacity.The twisting behavior and mechanical properties were investigated via a combination of finite-element simulation and experimental verification.An approximately linear relationship between the axial compressive strain and rotation angle was achieved before the strain reached 15% for the LPBF-processed CIT-compliant mechanism.

Influence of additive multilayer feature on thermodynamics, stress and microstructure development during laser 3D printing of aluminum-based material

A transient three dimensional model for describing the temperature behavior,thermo-capillary convection,microstructure evolution and the resultant mechanical properties during selective laser melting of Al N/Al Si10 Mg composite is proposed.The powder-solid transformation,temperature dependent physical properties and the preservation of the heat are taken into account.The effect of the additive manufacturing multilayer feature on the molten pool dynamics,cooling rate,crystal size,microstructure morphology,micro-hardness and types of the residual stress has been investigated.It shows that the operating temperature and the thermo-capillary convection obtained within the molten pool generally increases as the processing multilayers are successively added,while the thermal effect depth is negatively reduced.The preferential direction of the heat diffusion generally changes from a downward pattern,then to the slightly strengthened horizontal direction and finally to a typically horizontal one for various deposited layers being processed.Therefore,the microstructure of the solidified part along the building direction(Region I to Region V) undergoes an interesting transformation:directional columnar cellular microstructure,crosswise-extended cellular microstructure,refined cellular microstructure,fragmentation microstructure and the coarse cellular microstructure.The tensile stress and the compressive stress are comprehensively obtained within the finally solidified layers,significantly influencing the microhardness.

本文通过构建TVP-VAR模型就气候风险对跨境资本流动的时变冲击及其传导渠道进行实证分析。研究结果表明,气候风险会通过提升经济政策不确定性和引起大宗商品价格上涨而引发短期跨境资本外流,但随着气候风险影响减弱,资本外流现象会逐渐得到缓解并出现资本回流。进一步研究发现:近年来气候风险对财政政策不确定性和金属类大宗商品价格的正向冲击力度有所上升;贸易政策不确定性增强和家畜类商品价格上涨对跨境资本短期流出的冲击力度较大,货币与汇率政策不确定性增强和纺织品类商品价格上涨对跨境资本长期流入的冲击力度较大。

随着大量分布式光伏(distributed photovoltaic,DPV)并网,目前配电网中出现了日间过压和夜间低压并存的电压越限问题,并进一步影响了网络的经济运行。本工作基于此提出了两阶段光储系统协同运行的优化调度策略。该策略第一阶段通过计算节点的电压灵敏度来确定待调节的储能节点与充放功率,以及光伏可调节点;第二阶段建立了光储运行优化模型。该模型以储能的调度成本、购售电成本以及网损成本之和最小为目标,以网络潮流、节点电压、储能SOC(state of charge)、光伏的无功可调容量等作为约束,通过粒子群算法对该模型进行求解,可以得到光储系统日调度出力策略。最后,以某地区31节点的实际配电网作为算例,验证了本工作方法的有效性。算例结果表明,该策略可以通过光储的协同调度,有效治理电网中的电压越限问题,并且在保障配电网电压安全的同时,实现优化运行成本的目标。
