High-resolution melting （HRM） analysis relies on the use of fluorescent dyes, such as LCGreen, ResoLight, and SYTO9, which bind in a saturated manner to the double-stranded DNAs. These dyes are expensive in use and may not be affordable when dealing with a large quantity of samples. EvaGreen is a much cheaper DNA helix intercalating dye and has been used in quantitative real-time polymerase chain reaction （PCR） and post-PCR DNA melt curve analysis. Here we report on the development of an EvaGreen-based HRM analysis and its performance, in comparison with the popular LCGreen-based HRM analysis, in detection of DNA polymorphism in plants. We found that various polymorphisms ranged from single nucleotide polymorphisms （SNPs） to Indels were equally detected by using EvaGreen- or LCGreen-based HRM. EvaGreen dye was sensitive enough in discovery of SNPs in fivefold pooled samples. Using this economical dye we successfully identified multiple novel mutant alleles of Gln1-3 gene, which produces a cytosolic glutamine synthetase isoenzyme （GS1）, in a maize ethyl methanesulfonate （EMS）-mutagenized library, and genotyped rice mapping populations with SNP markers. The current results suggest that EvaGreen is a promising dye for HRM analysis for its ease to use and cost effectiveness.

The presence/absence variants （PAVs） are a major source of genome structural variation and have profound effects on phenotypic and genomic variation in animals and humans. However, little is understood about PAVs in plant genomes. Our previous resequencing effort on three sorghum （Sorghum bicolour L.） genomes, each 12？ coverage, uncovered 5 364 PAVs. Here, we report a detailed characterization of 51 large-size （＆gt;30 kb） PAVs. These PAVs spanned a total size of 2.92 Mb of the sorghum genome containing 202 known and predicted genes, including 38 genes annotated to encode celldeath and stress response genes. The PAVs varied considerably for repeat sequences and mobile elements with DNA trans-posons as the major components. The frequency and distribution of these PAVs differed substantial y across 96＆amp;nbsp;sorghum inbred lines, and the low-and high frequency PAVs differed in their gene categories. This report shed new light on the occurrence and diversity of PAVs in sorghum genomes. Our research exemplifies a new perspective to explore genome structural variation for genetic improvement in plant breeding.

Sodium carbonate （Na2CO3） presents a huge challenge to plants by the combined damaging effects of Na＋, high pH, and CO32. Little is known about the cellular responses to Na2CO3 stress. In this study, the transcriptome of maize （Zea mays L. cv. B73） roots exposed to Na2CO3 stress for 5 h was compared with those of NaCI and NaOH stresses. The expression of 8,319 genes, representing over a quarter of the total number of genes in the maize genome, was altered by Na2CO3 stress, and the downregulated genes （5,232） outnumbered the upregulated genes （3,087）. The effects of Na2CO3 differed from those of NaCI and NaOH, primarily by downregulating different categories of genes. Pathways commonly altered by Na2CO3, NaCI, and NaOH were enriched in phenylpropanoid biosynthesis, oxidation of unsaturated fatty acids, ATP- binding cassette （ABC） transporters, as well as the metabolism of secondary metabolites. Genes for brassinosteroid biosynthesis were specifically upregulated by Na2CO3, while genes involved in ascorbate and aldarate metabolism, protein processing in the endoplasmic reticulum and by N-glycosylation, fatty acid biosynthesis, and the circadian rhythm were downregulated. This work provides the first holistic picture of early transcriptomic adaptation to Na2CO3 stress, and highlights potential molecular pathways that could be manipulated to improve tolerance in maize.

Harvesting bio-kinetic energy using a triboelectric nanogenerator(TENG) is one of the promising routes to solve the sustainable energy supply problem for wearable electronics. However, additional materials, complex fabricating processes or specific mechanical structures are needed for existing TENGs to harvest bio-kinetic energy. Besides, they need to be tightly attached to the human body, which may result in detachment and malfunction under tense human motion. Herein, an intrinsic epidermal electrode-based TENG(E-TENG) is proposed to harvest human walking energy. The wearing shoes and ground are used as tribo-materials, and the human epidermis is used as the back electrode of the E-TENG. Compared with the traditional TENGs,the E-TENG does not need any additional tribo-materials and complex mechanical structures. Under optimal conditions, the voltage output of E-TENG can reach 914 V. E-TENG has been used as a self-powered warning sensor and pedometer sensor for demonstration. Furthermore, E-TENG based self-powered wireless sensor system has been developed using a newly designed micro energy electronic switch(MEES). Ambient ultraviolet intensity, temperature and humidity information can be monitored and then transmitted to mobile phone every 3.5 min, demonstrating great potential for widespread wearable applications.

Deterministic inversion based on deep learning has been widely utilized in model parameters *** by logging data,seismic data,wavelet and modeling operator,deterministic inversion based on deep learning can establish nonlinear relationships between seismic data and model ***,seismic data lacks low-frequency and contains noise,which increases the non-uniqueness of the *** conventional inversion method based on deep learning can only establish the deterministic relationship between seismic data and parameters,and cannot quantify the uncertainty of *** order to quickly quantify the uncertainty,a physics-guided deep mixture density network(PG-DMDN)is established by combining the mixture density network(MDN)with the deep neural network(DNN).Compared with Bayesian neural network(BNN)and network dropout,PG-DMDN has lower computing cost and shorter training time.A low-frequency model is introduced in the training process of the network to help the network learn the nonlinear relationship between narrowband seismic data and low-frequency *** addition,the block constraints are added to the PG-DMDN framework to improve the horizontal continuity of the inversion *** illustrate the benefits of proposed method,the PG-DMDN is compared with existing semi-supervised inversion *** synthetic data examples of Marmousi II model are utilized to quantify the influence of forward modeling part,low-frequency model,noise and the pseudo-wells number on inversion results,and prove the feasibility and stability of the proposed *** addition,the robustness and generality of the proposed method are verified by the field seismic data.

Effect of the vortical structures on the hydrodynamic performance of a pitching hydrofoil

The objective is to study the vortical structural behaviors of a transient pitching hydrofoil and their effects on the hydrodynamic performance. The pitching motion of the hydrofoil is set to pitch up with an almost constant rate from 5° to 15° and then back to 5°, with the Reynolds number 4.4×10^(5) and the frequency 2 Hz. The results show that the main coherent structures around the pitching hydrofoil include small-scale laminar separation bubble (LSB), large-scale second vortex (SV) and trailing edge vortex (TEV) which are all vortical. The relationship between the vortical structure and the lift is investigated with the finite-domain impulse theory. It indicates that the major part of the lift is contributed by the LSB, whereas the shedding and the formation of the SV and TEV cause the fluctuation of the lift. The proper orthogonal decomposition (POD) method is applied to capture the most energetic modes, revealing that the LSB mode occupies a large amount of energy in the flow field. The dynamic mode decomposition (DMD) method accurately extracts the dominant frequency and modal characteristics, with the first mode corresponding to the mean flow, the second mode corresponding to the LSB structure and the third and fourth modes corresponding to the vortex shedding.

Let a and b be positive integers such that a≤b and a≡b(mod 2).We say that G has all(a,b)-parity factors if G has an h-factor for every function h:V(G)→{a,a+2,…,b-2,b} with b|V(G)| even and h(v)≡b(mod 2) for all v∈V(G).In this paper,we prove that every graph G with n≥2(b+1)(a+b) vertices has all(a,b)-parity factors if δ(G)≥(b^(2)-b)/a,and for any two nonadjacent vertices u,v ∈V(G),max{d_(G)(u),d_(G)(v)}≥bn/a+***,we show that this result is best possible in some sense.

This paper investigates the path-following control problem with obstacle avoidance of autonomous surface vehicles in the presence of actuator faults,uncertainty and external *** surface vehicles inevitably suffer from actuator faults in complex sea environments,which may cause existing obstacle avoidance strategies to *** reduce the influence of actuator faults,an improved artificial potential function is constructed by introducing the lower bound of actuator efficiency *** nonlinear state observer,which only depends on measurable position information of the autonomous surface vehicle,is used to address uncertainties and external *** using a backstepping technique and adaptive mechanism,a path-following control strategy with obstacle avoidance and fault tolerance is designed which can ensure that the tracking errors converge to a small neighborhood of *** with existing results,the proposed control strategy has the capability of obstacle avoidance and fault tolerance ***,the comparison results through simulations are given to verify the effectiveness of the proposed method.

The cylinder block/valve plate interface is one of the most critical frictional interfaces of the swashplate-type axial piston ***,the poor lubrication interface caused rapid wear and high friction loss in an elastohydrodynamic lubrication system,decreasing the pump *** resistant bronze coatings were fabricated on 38CrMoAl substrate by Physical Vapor Deposition(PVD)and Chemical Vapor Deposition(CVD),***-on-disc wear tests were performed to comparatively investigate the wear behaviors of the coatings and bulk ductile iron *** can be found that the PVD-bronze coating exhibited better wear resistance than the other two *** enhanced wear resistance was attributed to the unique composite microstructure and desired mechanical strength,which could resist to mechanical shear and spallation,decreasing friction *** appropriate hardness of(1.33±0.07)GPa could be beneficial for enhancing its wear *** PVD-bronze coating possessed a much lower and stable coefficient of friction(about 0.1)and wear rate(about 6000μm3.N-1.m-1)under the loading forces of about 100 N after 20 *** wear mechanism was the abrasive wear.
