Acceptor-engineering tailored type-Ⅰ photosensitizer with aggregation-induced NIR-II emission for cancer multimodal phototheranostics

Exploration of single molecular species synchronously featured by long excitation/emission wavelength, accurate diagnosis, and effective therapy, remains supremely appealing to implement high-performance cancer phototheranostics. However, those previously established phototheranostic agents are undiversified and stereotyped in terms of structural skeleton, and generally exhibit insufficient phototheranostic outcomes. Herein, we innovatively utilized indanone-condensed thiadiazolo[3,4-g]quinoxaline(ITQ) as electron acceptor to construct novel photosensitizer with second near-infrared(NIR-II) emission. Experimental study and theoretical calculation demonstrated that comparing with the counterparts constituting by widely employed NIR-II building block benzobisthiadiazole(BBTD) and 6,7-diphenylthiadiazoloquinoxaline(DPTQ), ITQ-based photosensitizer(TITQ) showed superior aggregation-induced emission(AIE) characteristics, much stronger type-I reactive oxygen species(ROS) production, and prominent photothermal conversion capacity. Furthermore, TITQ nanoparticles with excellent biocompatibility were capable of effectively accumulating in the tumor site and visualizing tumor through fluorescence-photoacoustic-photothermal trimodal imaging with highly spatiotemporal resolution, and completely eliminating tumor by type-I photodynamic-photothermal therapy.

Petrology, geochemistry and Ar-Ar geochronology of eclogites in Jinshajiang orogenic belt, Gonjo area, eastern Tibet and restriction on Paleo-Tethyan evolution

As one of the important Paleo-Tethys suture zones in eastern Tibet,the Jinshajiang orogenic belt is of great significance to study the tectonic evolution of the main suture zone of *** this paper,eclogites developed in the Jinshajiang suture zone in Gonjo area,eastern Tibet,are selected as specific research objects,and petrological,geochemical and Ar-Ar geochronological analyses are carried *** major element data of the whole rock reveals that the eclogite samples have the characteristics of picritic basalt-basalt and belong to the oceanic low potassium *** results of rare earth elements and trace elements of the samples show that the protoliths of eclogites have characteristics similar to oceanic island basalt(OIB)or normal mid ocean ridge basalt(N-MORB).Muscovite(phengite)from two eclogite samples yield the Ar-Ar plateau ages of 247±2 Ma and 248±2 Ma respectively,representing the peak metamorphic age of eclogite facies and the timing of complete closure of the Jinshajiang Paleo-Tethys *** and biotite selected from the hosting rocks of eclogite yield the Ar-Ar plateau ages are 238±2 Ma and 225±2 Ma respectively,reflecting the exhumation age of eclogites and their hosting *** with the zircon U-Pb dating data(244 Ma)of eclogites obtained in previous work,it can be concluded that the Jinshajiang Paleo-Tethys ocean was completely closed and arc-continent collision was initiated at about 248-244 Ma(T21).Subsequently,due to the large-scale arc(continent)-collision orogeney between Deqin-Weixi continental margin arc and Zhongza block(T31-T32),the eclogites were rapidly uplifted to the shallow crust.

Rosiglitazone enhances fluorouracil-induced apoptosis of HT-29 cells by activating peroxisome proliferator-activated receptor γ

AIM： To examine whether and how rosiglitazone enhances apoptosis induced by fluorouracil in human colon cancer （HT-29） cells. METHODS： Human colon cancer HT-29 cells were cultured in vitro and treated with fluorouracil and/or rosiglitazone. Proliferation and growth of HT-29 cells were evaluated by MTF assay and trypan blue exclusion methods, respectively. The apoptosis of HT-29 cells was determined by acridine orange/ethidium bromide staining and flow cytometry using PI fluorescence staining. The expressions of peroxisome proliferator-activated receptor γ （PPARγ）, Bcl-2 and Bax in HT-29 cells were analyzed by Western blot. RESULTS： Although rosiglitazone at the concentration below 30 μmol/L for 72 h exerted almost no inhibitory effect on proliferation and growth of HT-29 cells, it could significantly enhance fluorouracil-induced HT-29 cell proliferation and growth inhibition. Furthermore, 10 μmol/L rosilitazone did not induce apoptosis of HT-29 cells but dramatically enhanced fluorouracil-induced apoptosis of HT-29 cells. However, rosiglitazone did not improve apoptosis induced by fluorouracil in HT-29 cells pretreated with GW9662, a PPARγ antagonist. Meanwhile, the expression of Bax and PPAR7 was upregulated, while the expression of Bcl-2 was down regulated in HT-29 cells treated with rosiglitazone in a time-dependent manner. However, the effect of rosiglitazone on Bcl-2 and Bax was blocked or diminished in the presence of GW9662. CONCLUSION： Rosiglitazone enhances fluorouracilinduced apoptosis of HT-29 cells by activating PPARγ.

Diffusion tensor imaging reveals brain abnormality patterns in a patient with semantic dementia

BACKGROUND： Previous studies regarding primary progressive aphasia （PPA） have focused on progressive, non-fluent aphasia. Little information is available with regard to the use of diffusion tensor imaging compared with conventional magnetic resonance imaging for the detection of subtle structural abnormalities. OBJECTIVE： To investigate and localize brain abnormalities in a Chinese patient with semantic dementia. DESIGN, TIME AND SETTING： A concurrent, non-randomized, case-controlled, neuroimaging, clinical trial was performed at the Department of Radiology, West China Hospital of Sichuan University in March 2009. PARTICIPANTS： One 75-year-old male patient, who was diagnosed with semantic dementia, and 21 age- and gender-matched healthy volunteers were recruited for the study. METHODS： Diffusion tensor imaging was used to determine mean diffusion （MD） and fractional anisotropy （FA） in the brains of the patient and the 21 healthy subjects. Voxel-based analysis of MD and FA values was performed using statistical parametric mapping. MAIN OUTCOME MEASURES： MD and FA value maps differences between patient and controls. RESULTS： MD was significantly increased in both cerebra, but was predominant on the left side and expanded to outside of the language-related region. Reduced MD was not detected in any of the brains. FA was shown to be decreased in the corpus callosum, but was increased in the basal ganglia. CONCLUSION： The present study provided clear in vivo magnetic imaging evidence of diffuse brain involvement in semantic dementia. Increases in MD were greater than in FA when brain diffusion alterations were detected, which suggested that MD could be a better marker of disease progression.

Prebiotic oligosaccharides change the concentrations of short-chain fatty acids and the microbial population of mouse bowel

The purpose of this study was to clarify effects of selected oligosaccharides on concentrations of cecal short-chain fatty acids （SCFAs）, total large bowel wet weight and wall weight, and cecal microbiota levels in mice. Mice were respectively given gavage of selected fructooligosaccharides （FOS）, galactooligosaccharides （GOS）, mannanoligosaccharides （MOS）, and ehitooligosaccharides （COS） [1000 mg/（kg body weightd）]. Control group was given physiological saline solution. After 14 d treatment, SCFAs and lactate in mice cecum were significantly increased （P〈0.05） by intake of oligosaccharides, especially FOS and GOS. Thus, providing these oligosaccharides as ingredients in nutritional formulas may benefit the gastrointestinal tract.

To predict the wave loads of a flexible trimaran in different wave fields,a one-way interaction numerical simulation method is proposed by integrating the fluid solver(Star-CCM+)and structural solver(Abaqus).Differing from the existing coupled CFD-FEA method for monohull ships in head waves,the presented method equates the mass and stiffness of the whole ship to the hull shell so that any transverse and longitudinal section stress of the hull in oblique waves can be ***,verification study and sensitivity analysis are carried out by comparing the trimaran motions using different mesh sizes and time step *** on the wave elevation of uni-and bi-directional waves is also carried *** a comprehensive analysis on the structural responses of the trimaran in different uni-directional regular wave and bi-directional cross sea conditions is carried out,***,the differences in structural response characteristics of trimaran in different wave fields are *** results show that the present method can reduce the computational burden of the two-way fluid-structure interaction simulations.
