Multifunctional RGD coated a single-atom iron nanozyme:A highly selective approach to inducing ferroptosis and enhancing immunotherapy for pancreatic cancer

Nanozyme is a new promising approach to cancer therapy for its ability to induce ferroptosis by activating H_(2)O_(2)via a traditional radical pathway and enhance cancer immunotherapy.However,short half-life period of hydroxyl radical(·OH)results in unsatisfied effectiveness.Herein,we synthesized a single-atom iron nanozyme(Fe-SAzyme),which can activate H_(2)O_(2)via a non-radical pathway to generate Fe-based reactive oxygen species(ROS)(O=FeO_(3)=O)for promoting the ferroptosis of pancreatic cancer cells.This Fe-SAzyme could be specifically phagocytosed by pancreatic cancer cells,increasing ROS levels and inhibiting glutathione(GSH)synthesis,which activates ferroptosis.Tumor magnetic resonance imaging(MRI)showed decreased T2 signal after intravenous injection of RGD@Fe-AC(AC=activated carbon).Moreover,RGD@Fe-AC promoted dendritic cell(DC)maturation,overcame Treg-mediated immunosuppression,activated T cells to trigger adaptive immune responses,and enhanced the efficacy ofα-PD-L1 immunotherapy.Our research demonstrated that RGD@Fe-AC provided a straightforward,easily implemented,and selective approach for pancreatic cancer treatment and immunotherapy.

Optimization of Urban Multi-energy Flow Systems Considering Seasonal Peak Shaving of Natural Gas

In order to alleviate the shortage of natural gas supply in winter,relevant policies have been issued to promote the construction of gas peak-shaving and storage facilities.Largescale gas storage can transfer the supply-demand relationship of natural gas in time sequence,which has great potential in improving the economy and reliabillity of urban multi-energy flow systems.Addressing this issue,this paper proposes a mid-and long-term energy optimization method for urban multi-energy flow system that considers seasonal peak shaving of natural gas.First,the energy supply and demand features of different energy subsystems are analyzed.Then,a network model of the electricity-gas-heat multi-energy flow system is established.Considering the time-of-use electricity price mechanism and the seasonal fluctuations of the natural gas price,a mid-and long-term energy optimization model maximizing the annual economic revenue is established.The alternative direction multiplier method with Gaussian back substitution(ADMM-GBS)algorithm is used to solve the optimal dispatch model.Finally,the proposed method is verified by employing the actual data of the demonstration zone in Yangzhong City,China.The simulation results show that the proposed method is effective.

Clinical and microbiological characterization of Clostridium difficile infection in a tertiary care hospital in Shanghai, China

Background Over the last decade,Clostridium difficile infection （CDI） has emerged as a significant nosocomial infection,yet little has been reported from China.This study aimed to characterize the clinical and microbiological features of CDI from a hospital in Shanghai.Methods Patients with CDI seen between December 2010 and March 2013 were included in this study,of which clinical data were retrospectively collected.The microbiological features of corresponding isolates were analyzed including genotype by multi-locus sequence typing （MLST）,antimicrobial susceptibility,toxin production,sporulation capacity,biofilm formation,and motility.Results Ninety-four cases of CDI were included during this study period,12 of whom were severe cases.By reviewing the clinical data,all patients were treated empirically with proton pump inhibitor or antibiotics or both,and they were distributed widely across various wards,most frequently to the digestive ward （28/94,29.79％）.Comparing the severe with mild cases,no significant differences were found in the basic epidemiological data or the microbiological features.Among the 94 isolates,31 were toxin A-negative toxin B-positive all genotyped as ST37.They generated fewer toxins and spores,as well as similar amounts of biofilm and motility percentages,but exhibited highest drug resistance to cephalosporins,quinolones,macrolide-lincosamide and streptogramin （MLSB）,and tetracycline.Conclusions No specific clinical genotype or microbiological features were found in severe cases; antimicrobial resistance could be the primary reason for epidemic strains leading to the dissemination and persistence of CDI.

BACKGROUND Eltrombopag is an orally administered thrombopoietin receptor agonist linked to a heightened risk of treatment-related thromboembolism.Both venous and arterial thromboses have been documented in the medical literature.CASE SUMMARY In the absence of nephropathy,a 48-year-old patient receiving eltrombopag for immune thrombocytopenia(ITP)developed renal vein thrombosis and pulmonary embolism.The renal vein thrombus spontaneously resolved during subsequent anticoagulant treatment,restoring venous circulation.CONCLUSION A rapid upsurge in platelets,rather than their absolute number,may trigger thrombotic events in this setting.For patients at high thrombotic risk,individualized eltrombopag dosing and vigilance in platelet monitoring are perhaps needed during treatment of ITP.

In order to optimize the production process, improve the production efficiency and accelerate the development and application of the domestic dual-phase steel, the effects of heat treatment process parameters on microstructure and mechanical properties of DP440 cold rolled strip were studied by the CAS-300 simulated continuous annealing equipment. When the heating rate increased from 5 to 100 ℃/s, both the tensile strength and yield strength increased because of the decreased grain size. When the intercritical annealing temperature increased from 780 to 850 ℃, the martensite content decreased so that the tensile strength decreased first, then increased, and the yield strength increased. When the rapid cooling rate increased from 25 to 100 ℃/s, because the martensite content increased, the tensile strength increased, while the yield strength decreased. When the overaging temperature increased from 260 to 400 ℃, the tensile strength decreased, while the yield strength increased. When the overaging time increased from 240 to 480 s, the tensile strength decreased a little, while the yield strength increased a little.

Scaffolding proteins Gab1 is ubiquitously expressed and belongs to the GAB family(GABs),which integrates multiple signal events.This protein family has recently attracted interestthat genetic variants of human GAB1 are considered a risk factor for asthma.Our previousresearch has revealed a novel function of Gab2 in mediating mucin gene expression andGoblet cell hyperplasia(GCH),which is an important process in asthma.However,the functionof Gab1 in asthma remains unclear.In asthma exacerbated patients,we found a prominentelevated level of Gab1 in peripheral blood monouclear cells compared to that of convalescencepatients.Next,we generated conditional Gab1 deletion mice in myeloid cells(Gab1 KO mice)to determinate physiological the role of Gab1 in asthma.Lung histologyand mucus metaplasia indicated that disruption of Gab1 in dendritic cells(DCs)leads to alleviatedasthmatic inflammation.Future investigation showed that loss of Gab1results inimpaired DC migration to lymph node,while has no influences on DC maturation、antigenuptake and presentation.Mechanically,Gab1 is essentially involved in CCL19-mediatedAKT activation.Together,these finding has characterized a novel of Gab1 in mediating DCmigration and that deregulation of Gab1 signal in lungs predisposes asthmatic inflammation.

结合某热镀锌线的Vai C lec im动态气刀系统的特点,对热镀锌薄钢板边部过镀锌缺陷的产生原因进行了分析。通过采用加热装置对带钢边部进行热量补偿,有效解决了边部过镀锌缺陷问题,取得了较好的效果。

目的探讨核受体相互作用蛋白(NRIP3)在乳腺癌组织中的表达及与淋巴结阴性乳腺癌无远处转移生存的关系。方法利用BRB-Array Tools软件分析165例乳腺癌芯片中NRIP3基因表达水平,Kaplan-Meier法计算不同表达水平的无远处转移生存率,并进行Log-rank检验;应用Cox比例风险回归模型行多因素分析。结果 NRIP3基因低、中、高表达组中位无远处转移生存期(DMFS)分别为(3 980±331)d、(4 391±246)d、(4 518±147)d,Logrank检验生存曲线差异有统计学意义(χ2=7.847,P0.05)。结论 NRIP3基因高表达可抑制淋巴结阴性乳腺癌发生远处转移,其发挥作用与ER阳性表达以及可能的内分泌治疗关系值得进一步研究。
