Human-specific insertions play important roles in human phenotypes and diseases.Here we reported a 446-bp insertion(Insert-446)in intron 11 of the TBC1D8B gene,located on chromosome X,and traced its origin to a portion of intron 6 of the EBF1 gene on chromosome 5.Interestingly,Insert-446 was present in the human Neanderthal and Denisovans genomes,and was fixed in humans after human-chimpanzee divergence.We have demonstrated that Insert-446 acts as an enhancer through binding transcript factors that promotes a higher expression of human TBC1D8B gene as compared with orthologs in macaques.In addition,over-expression TBC1D8B promoted cell proliferation and migration through“a dual finger”catalytic mechanism(Arg538 and Gln573)in the TBC domain in vitro and knockdown of TBC1D8B attenuated tumorigenesis in vivo.Knockout of Insert-446 prevented cell proliferation and migration in cancer and normal cells.Our results reveal that the human-specific Insert-446 promotes cell proliferation and migration by upregulating the expression of TBC1D8B gene.These findings provide a significant insight into the effects of human-specific insertions on evolution.

1.Introduction Modern turbine engines can be substantially damaged when the airborne silicate-based particles(fly ash,desert sand,volcanic ash,runway debris,etc.)are ingested into jet engines at high temperatures(1150−1250℃)[1-4],leading to severe CaO-MgO-Al_(2)O_(3)-SiO_(2)(CMAS)corrosion during their service life.In order to extend the engine life,many efforts have been devoted to thermal barrier coatings(TBCs)and environmental barrier coatings(EBCs)to protect the substrate from corrosion by molten CMAS[5-7].To be specific,thermochemical and thermomechanical corrosion are the two main mechanisms of CMAS corrosion[8].The structure and composition of the coatings were changed due to the chemical reaction between the molten CMAS and coating,the detrimental phases were then precipitated[9,10].

基于数据挖掘探讨中医药治疗H型高血压病的用药规律

基于R语言数据挖掘技术探讨中医药治疗H型高血压病的用药规律,为中药治疗H型高血压病提供依据与思路。通过检索中国知网(CNKI)、万方(Wanfang)、维普(VIP)、中国生物医学文献服务系统(SinoMed),搜集中药治疗H型高血压病的临床研究,筛选数据并使用Excel建立数据库。运用Rstudio软件进行药物功效分类,及四气五味、归经、使用频次、相关性、关联规则和聚类分析,探讨中医药用药规律。共得到中药处方191首,主要证型有痰湿壅盛证、痰瘀互结型、阴虚阳亢证等,使用中药169种,频次1875次,其中单次使用频次大于20次的药物为高频药物,共26种,依次为天麻、茯苓、制半夏、牛膝、甘草等,高频药物功效分类以补虚药、活血化瘀药、平肝息风药、利水渗湿药为主,药物四气以温、平为主,五味以甘、苦为主,归经以肝经、脾经为多,关联规则分析共得到14组常用药物组合,相关性分析得到相关系数高的20种药物组合,聚类分析得到6个药物组方。对痰湿壅盛证H型高血压病进行关联规则分析得到8组药对,相关性分析得到相关系数高的7种药物组合,聚类分析得到3个药物组方。结果显示,H型高血压病病理因素以“痰”“瘀”“虚”为主,病位在肝,涉及脾、肺等,常用中药有天麻、茯苓、制半夏、牛膝等,常用方剂为半夏白术天麻汤、天麻钩藤饮与补阳还五汤。该研究展示的用药规律可为临床中药治疗H型高血压病提供一定思路。

Robust photo-assisted removal of NO at room temperature:Experimental and density functional theory calculation with optical carrier

Photo-assisted SCR(PSCR) offers a potential solution for removal of NO at room temperature. MnTiO_(x)as PSCR catalyst exhibits superior performance with NO removal of 100% at the room temperature. Electron paramagnetic resonance(EPR) analysis revealed the presence of numerous oxygen vacancies on MnTiO_(x). Optical carrier density functional theory(DFT) calculations showed that the threedimensional orbital hybridization of Mn and Ti is significantly enhanced under light irradiation. The MnTiO_(x)catalyst exhibited excellent electron–hole separation ability, which can adsorbe NH_(3)and dissociate to form NH_(2)fragments and H atoms. In-situ diffuse reflectance infrared fourier-transform spectroscopy(DRIFTS) indicated that the optical carrier enhanced NH_(3)adsorption on MnTiO_(x), which makes it possess excellent PSCR activity. This work provided an additional strategy to NO removal with PSCR catalysts and showed potential for use in photocatalysis.

A comprehensive review of heart rate variability as an indicator in the regulation of the autonomic nervous system by acupuncture:a bibliometric analysis

This study sought to conduct a bibliometric analysis of acupuncture studies focusing on heart rate variability(HRV)and to investigate the correlation between various acupoints and their effects on HRV by utilizing association rule mining and network analysis.A total of 536 publications on the topic of acupuncture studies based on HRV.The disease keyword analysis revealed that HRV-related acupuncture studies were mainly related to pain,inflammation,emotional disorders,gastrointestinal function,and hypertension.A separate analysis was conducted on acupuncture prescriptions,and Neiguan(PC6)and Zusanli(ST36)were the most frequently used acupoints.The core acupoints for HRV regulation were identified as PC6,ST36,Shenmen(HT7),Hegu(LI4),Sanyinjiao(SP6),Jianshi(PC5),Taichong(LR3),Quchi(LI11),Guanyuan(CV4),Baihui(GV20),and Taixi(KI3).Additionally,the research encompassed 46 reports on acupuncture animal experiments conducted on HRV,with ST36 being the most frequently utilized acupoint.The research presented in this study offers valuable insights into the global research trend and hotspots in acupuncture-based HRV studies,as well as identifying frequently used combinations of acupoints.The findings may be helpful for further research in this field and provide valuable information about the potential use of acupuncture for improving HRV in both humans and animals.

掺杂Li^(+)的NaGdF_(4)∶Yb^(3+)/Eu^(3+)上转换发光增强及防伪应用

稀土发光材料由于其良好的上转换发光性能已经逐渐成为荧光防伪领域的研究热点。首先采用水热法合成了一系列不同Li^(+)掺杂浓度的NaGdF_(4)∶Yb^(3+)/Eu^(3+)晶体,利用X射线衍射(XRD)、扫描电子显微镜(SEM)、上转换发射光谱测试等表征方法,对样品的形貌、尺寸和上转换发光性能进行了分析,并选择发光强度和效果最好的晶体应用于防伪识别。实验结果表明,NaGdF_(4)∶Yb^(3+)/Eu^(3+)/Li^(+)的XRD衍射图谱与标准六方相NaGdF4的衍射峰一致,衍射峰尖锐且没有杂峰,证明合成了高纯度高结晶度的NaGdF4∶Yb^(3+)/Eu^(3+)/Li^(+)晶体。晶体的SEM图可以看出生成的样品为纯六方相,分布均匀,无团聚现象。共掺Yb^(3+)/Eu^(3+)/Li^(+)对晶体的结构、形貌、尺寸几乎没有影响。从上转换发射光谱可以看出,15 mol%Li^(+)掺杂的NaGdF4∶Yb^(3+)/Eu^(3+)晶体绿色发光强度是未掺杂Li^(+)的样品的6倍。考察了980 nm激光器的功率变化对未掺杂Li^(+)以及掺杂15 mol%Li^(+)样品的上转换发光性能的影响,发现随着激发功率从0.8 W增加到2.2 W的过程中,上转换发光强度逐渐增强。拟合后得到样品上转换过程中的所需光子数n值接近于2,为双光子发光过程。对NaGdF4∶Yb/Eu/Li进行了荧光寿命测试,5D_(1)能级的荧光寿命是未掺杂的1.4倍。最后,将形貌均匀、发光强度强的NaGdF_(4)∶0.2Yb/0.02Eu/0.15Li制成防伪油墨,通过丝网印刷技术在纸张、玻璃、塑料等材料上印刷荧光防伪图案。在980 nm激光的泵浦下,防伪图案发出明亮的绿光。在自然环境下,纸张上的防伪图案有很好的隐蔽性。图案中的“safe”字样长度为5.5 mm,字母间距为0.5 mm,在980 nm激光的泵浦下,仍然可以分辨清楚。将印制有防伪图案的塑料暴露在室外自然环境下一个月,图案没有明显改变。表明NaGdF_(4)∶0.2Yb/0.02Eu/0.15Li制成的防伪图案分辨率高,易于识别,受环境影响小,在防伪识别方面具有良好的应用前景。

Enhanced comprehensive properties of stereolithography 3D printed alumina ceramic cores with high porosities by a powder gradation design

Ceramic cores with complex structures and optimized properties are critical for hollow turbine blades applied in aeroengines.Compared to traditional methods,additive manufacturing(AM)presents great advantages in forming complex ceramic cores,but how to balance the porosity and strength is an enormous challenge.In this work,alumina ceramic cores with high porosity,moderate strength,and low high-temperature deflection were prepared using stereolithography(SLA)3D printing by a novel powder gradation design strategy.The contradiction between porosity and flexural strength is well adjusted when the mass ratio of the coarse,medium,and fine particles is 2:1:1 and the sintering temperature is 1600℃.The fracture mode of coarse particles in sintered SLA 3D printing ceramic transforms from intergranular fracture to transgranular fracture with the increase of sintering temperature and the proportion of fine powders in powder system.The sintered porosity has a greater influence on the high-temperature deflection of SLA 3D printed ceramic cores than grain size.On this basis,a"non-skeleton"microstructure model of SLA 3D printed alumina ceramic cores is created to explain the relationship between the sintering process and properties.As a result,high porosity(36.4%),appropriate strength(50.1 MPa),and low high-temperature deflection(2.27 mm)were achieved by optimizing particle size gradation and sintering process,which provides an insight into the important enhancement of the comprehensive properties of SLA 3D printed ceramic cores.

Neuroprotective Effects of Brain-Gut Peptides: A Potential Therapy for Parkinson’s Disease

Parkinson's disease(PD) is the second most common neurodegenerative disease and is typically associated with progressive motor and non-motor dysfunctions.Currently, dopamine replacement therapy is mainly used to relieve the motor symptoms, while its long-term application can lead to various complications and does not cure the disease. Numerous studies have demonstrated that many brain-gut peptides have neuroprotective effects in vivo and in vitro, and may be a promising treatment for PD. In recent years, some progress has been made in studies on the neuroprotective effects of some newly-discovered braingut peptides, such as glucagon-like peptide 1, pituitary adenylate cyclase activating polypeptide, nesfatin-1, and ghrelin. However, there is still no systematic review on the neuroprotective effects common to these peptides. Thus,here we review the neuroprotective effects and the associated mechanisms of these four peptides, as well as other brain-gut peptides related to PD, in the hope of providing new ideas for the treatment of PD and related clinical research.

德阳市平原区浅层地下水水化学特征与健康风险评价

地下水是德阳市的重要供水水源,了解其水化学特征及生态风险状况对可持续发展至关重要.以德阳市平原区为研究对象,采集78个地下水样,分析其水化学特征及其影响因素,运用绝对主成分-多元线性回归模型(APCS-MLR)进行水化学组分来源解析,并综合运用熵权水质指数(EWQI)和人类健康风险评价模型(HHRA)进行地下水水质及健康风险评价.结果表明,地下水中Ca^(2+)、HCO_(3)^(-)、Mn^(2+)和TFe浓度均值超过国家Ⅲ类水质标准;Na^(+)、K^(+)、Cl^(-)、NO_(3)^(-)、Mn^(2+)和TFe变异系数介于1.11~3.72,含量起伏变化大,局部富集程度高;区内地下水化学类型以HCO_(3)·SO_(4)-Ca、HCO_(3)-Ca·Mg、HCO_(3)·SO_(4)-Ca·Mg和HCO_(3)-Ca型为主,分别占比33.33%、25.64%、19.23%和14.10%.APCS-MLR模型源解析结果表明,地下水水化学组分主要来源为:水岩作用源(32.87%)、农业活动源(12.75%)、生活污水和工业废水排放源(12.40%)、原生地质环境和水岩作用复合源(9.86%)、农业与工业复合源(9.44%)和未识别源(22.69%).熵权水质指数(EWQI)结果显示,地下水质量Ⅰ级和Ⅱ级占比分别为6.41%和74.36%,达到优良和良好程度;地下水质量为Ⅲ级占比11.54%,质量中等;地下水质量为Ⅳ级和Ⅴ级占比分别为6.41%和1.28%,地下水质量差或极差;影响地下水水质等级的主要指标依次为:TFe>Ca^(2+)>Mn^(2+)>HCO_(3)^(-).HHRA模型结果显示区内潜在非致癌风险较低,均未超过1;与皮肤接触相比,直接摄入是这些指标危害人类的主要途径;在相同环境下儿童比成人暴露更容易受到伤害.整体而言,区内地下水水质状况良好,研究区东南部存在一定的潜在风险,在地下水资源利用中应加强对该地区的地下水资源管理和污染防治.
