Rapid Detection of Filoviruses by Real-time TaqMan Polymerase Chain Reaction Assays

Ebola virus （EBOV） and Marburg virus （MARV） are causative agents of severe hemorrhagic fever with high mortality rates in humans and non-human primates and there is currently no licensed vaccine or therapeutics. To date, there is no specific laboratory diagnostic test in China, while there is a national need to provide differential diagnosis during outbreaks and for instituting acceptable quarantine procedures. In this study, the TaqMan RT-PCR assays targeting the nucleoprotein genes of the Zaire Ebolavirus （ZEBOV） and MARV were developed and their sensitivities and specificities were investigated. Our results indicated that the assays were able to make reliable diagnosis over a wide range of virus copies from 103 to 109, corresponding to the threshold of a standard RNA transcript. The results showed that there were about 101 RNA copies per milliliter of virus culture supernatant, equivalent to 10,000 RNA molecules per infectious virion, suggesting the presence of many non-infectious particles. These data indicated that the TaqMan RT-PCR assays developed in this study will be suitable

The Lines of Lyman and the Titus-Bode Law Preferred Orbital of a Celestial Body

We have proposed, thanks to a new model of the hydrogen atom [1], some explanation of the lines observed by Lyman in the spectrographic analysis of this atom. The model is based on a prequantum physics, itself founded on classical mechanics completed by the existence of a universal cloud of tiny particles called U. This cloud induces simultaneously and similarly electromagnetic and gravitational effects. This common origin creates a narrow link between how planets are arranged in a solar system, say the Titus-Bode law, and how the electrons are arranged in an atom, say the lines of Lyman. We describe what this link is in the following text and, more generally, what is the preferred orbit of an isolated celestial body.

In this work, activated carbons (ACs) prepared by chemical activation of garcinia cola nut shell impregnated with H3PO4 (CBH2/1) and KOH (CBK1/1) were used to study the kinetics, equilibrium and thermodynamics of the adsorption of thymol blue from aqueous solution. The characterization of ACs showed the BET measurements gave surface area and total pore volume respectively of 328.407 m2·g-1 and 0.1032 cm3·g-1 for CBH2/1 and 25.962 m2·g-1 and 0.03 cm3·g-1for CBK1/1;elemental analysis showed a high percentage of carbon in both ACs. Influence of parameters such as initial pH, contact time, adsorbent mass, initial concentration, ionic strength and the effect of temperature on the removal of thymol blue from aqueous solution were studied in batch mode. The studies showed that equilibrium adsorption was attained after 60 minutes for the two ACs, adsorption capacity increased with increasing concentration of thymol blue, and maximum adsorption capacity was obtained at an acidic environment with pH 2. Avrami’s non-linear kinetic expression was the best suited for describing the adsorption kinetics of thymol blue onto ACs, while equilibrium data showed that the three-parameter isotherms better described the adsorption process since R2 > 0.96, and the error functions were lowest for all of them. Maximum adsorption capacity values obtained using the three-parameter Fritz-Schlunder equation were 32.147 mg·g-1 for CBH2/1 and 67.494 mg·g-1 for CBK1/1. The values of the model parameters g and mFS respectively, obtained using the Redlich-Peterson and Fritz-Schlunder III isotherms below 1, showed that the adsorption of thymol blue by the ACs occurred on heterogeneous surfaces. Thermodynamic analyses of the data of the adsorption of thymol blue onto ACs revealed that the adsorption process was temperature dependent, endothermic and spontaneous.

One case of intrahepatic cholangiocarcinoma amenable to resection after radioembolization

We report the case of a 57-year-old man who was diagnosed with a large unresectable cholangiocarcinoma associated with 2 satellite nodules and without clear margins with the right hepatic vein. Despite 4 cycles of GEMOX (stopped due to a hypertransaminasemia believed to be due to gemcitabine) and 4 cycles of FOLFIRINOX, the tumor remained stable and continued to be considered unresectable. Radioembolization (resin microspheres, SIRS-spheres®) targeting the left liver (474 MBq) and segment IV (440 MBq) was performed. This injection was very well tolerated, and 4 more cycles of FOLFIRINOX were given while waiting for radioembolization efficacy. On computed tomography scan, a partial response was observed; the tumor was far less hypervascularized, and a margin was observed between the tumor and the right hepatic vein. A left hepatectomy enlarged to segment VIII was performed. On pathological exam, most of the tumor was acellular, with dense fibrosis around visible microspheres. Viable cells were observed only at a distance from beads. Radioembolization can be useful in the treatment of cholangiocarcinoma, allowing in some cases a secondary resection.

Epidemiological, Diagnostic and Therapeutic Aspects of Locally Advanced and/or Metastatic Breast Cancers at the Chu-Mel of Cotonou

Introduction: Breast cancer mortality remains high in most developing countries. Benin Republic does not yet have a technical platform required for the diagnosis and effective management of breast cancer “according to the recommendations”. Objective: To study the epidemiological, diagnostic and therapeutic aspects of locally advanced and/or metastatic breast cancers at the Centre Hospitalier Universitaire-de la Mère et de l’Enfant Lagune (CHU-MEL) of Cotonou from 2018 to 2021. Study Methods: This was a retrospective cross-sectional study with a descriptive purpose, carried out from January 1, 2018 to September 30, 2021 in the Gynaecology-Obstetrics Section of the CHU-MEL of Cotonou. The study population was patients diagnosed with locally advanced and/or metastatic breast cancers. Results: At the end of the study, 63 cases of locally advanced and/or metastatic breast cancer were recorded in 80 cases of breast cancer;a 74.11% prevalence of locally advanced and metastatic breast cancer. The mean age of the patients was 42.6 years. The main reason for consultation was the discovery of a mass in the breast (65.1%). Clinically, an orange peel was noted in 39.6%. Mammography (47.6%) and breast ultrasonography (42.9%) were the reference imaging workups for breast cancer diagnosis. Histopathologically, infiltrating ductal carcinoma (94.1%) was the predominant type with Scarff-Bloom and Richardson (SBR) grade II found in 35.3%. Progesterone and estrogen receptors were present in 42.9%. Mastectomy with axillary node dissection was the surgical treatment performed (80%) after neoadjuvant chemotherapy (39.6%). Only one patient had received radiotherapy. No patient received hormone therapy. Conclusion: Locally advanced and metastatic cancers remain a real public health problem. It is essential that major investments be made in order to improve both their diagnosis and management.

The cumulative effects of periodic redox cycling on the mobility of As,Fe,and S from alluvial sediment to groundwater were investigated in bioreactor *** particular sediments from the alluvial floodplain of the Mekong Delta River were investigated: Matrix A (14 m deep) had a higher pyrite concentration than matrix B (7 m deep) *** present in matrix B but absent in matrix *** the reactors,the sediment suspensions were supplemented with As(III) and SO4^2-,and were subjected to three full-redox cycles entailing phases of nitrogen/CO2,compressed air sparging,and cellobiose *** differences in As concentration and speciation were observed upon redox *** support the fact that initial sediment composition is the main factor controlling arsenic release and its speciation during the redox ***,a high pyrite content associated with a low SO4^2- content resulted in an increase in dissolved As concentrations,mainly in the form of As(III),after anoxic half-cycles;whereas a decrease in As concentrations mainly in the form of As(V),was instead observed after oxic *** addition,oxic conditions were found to be responsible for pyrite and arsenian pyrite oxidation,increasing the As pool available for *** same processes seem to occur in sediment with the presence of gypsum,but,in this case,dissolved As were sequestered by biotic or abiotic redox reactions occurring in the FeeS system,and by specific physico-chemical condition (***).The contrasting results obtained for two sediments sampled from the same core show that many complexes and entangled factors are at work,and further refinement is needed to explain the spatial and temporal variability of As release to groundwater of the Mekong River Delta (Vietnam).
