CD4^(+)T cells can"help"or"license" conventional type 1 dendritic cells(cDC1s)to induce CD8^(+)cytotoxic T lymphocyte(CTl)anticancer responses,as proven in mouse *** recently identified cDC1s with a transcriptomic imprint of CD4^(+)T-cell help,specifically in T-cell-infiltrated human cancers,and these cells were associated with a good prognosis and response to pD-1-targeting ***,we delineate the mechanism of cDC1 licensing by CD4^(+)T cells in *** CD4^(+)T cells produce IFNβvia the STING pathway,which promotes MHC-I antigen(cross-)presentation by cDC1s and thereby improves their ability to induce CTl anticancer *** cooperation with CD40 ligand(l),IFNβalso optimizes the costimulatory and other functions of cDC1s required for CTl response ***-I-producing CD4^(+)T cells are present in diverse T-cell-infiltrated cancers and likely deliver“help”signals to CTls locally,according to their transcriptomic profile and colocalization with“helped/licensed”cDCs and tumor-reactive CD8^(+)T *** agreement with this scenario,the presence of IFN-I-producing CD4^(+)T cells in the TME is associated with overall survival and the response to pD-1 checkpoint blockade in cancer patients.

Differential modulation of crown allometry and stem growth at gap edges in five European tree species by drought conditions

The olfactory system of pieris brassicae caterpllars:from receptors to glomeruli

Tolfactory system of adult lepidopterans is among the best described neuronal ***,comparatively little is known about the organization of the olfactory system in the larval stage of these ***,we explore the expression of olfactory receptors and the organization of olfactory sensory neurons in caterpillars of pieris brassicae,a significant pest species in Europe and a well-studied species for its chemical *** describe the larval olfactory system in this species,we first analyzed the head transcriptome of third-instar larvae(l3)and identified 16 odorant receptors(ORs)including the OR coreceptor(Orco),13 ionotropic receptors(IRs),and 8 gustatory receptors(GRs).We then quantified the expression of these 16 ORs in different life stages,using qpCR,and found that the majority of ORs had significantly higher expression in the l4 stage than in the l3 and l5 stages,indicating that the larval olfactory system is not static throughout caterpillar *** an Orco-specific antibody,we identified all olfactory receptor neurons(ORNs)expressing the Orco protein in l3,l4,and l5 caterpillars and found a total of 34 Orco-positive ORNs,distributed among three sensilla on the *** number of Orco-positive ORNs did not differ among the three larval ***,we used retrograde axon tracing of the antennal nerve and identified a mean of 15 glomeruli in the larval antennal center(lAC),suggesting that the caterpillar olfactory system follows a similar design as the adult olfactory system,although with a lower numerical *** together,our results provide a detailed analysis of the larval olfactory neurons in p brassicae,highlighting both the differences as well as the commonalities with the adult olfactory *** findings contribute to a better understanding of the development of the olfactory system in insects and its life-stage-specific adaptations.

Background The exact influence of the COVID-19 pandemic on sexual intercourse and usage of contraception remains largely uncharted *** bridge this gap in knowledge,we conducted a comprehensive,cross-sectional examination of long-term trends in the prevalence of sexual intercourse and usage of contraception among South Korean adolescents from 2006 to *** In our research,we drew upon data encompassing 1,138,799 South Korean adolescents aged 12 to 18 years,derived from the Korean Youth Risk Behavior Web-based Survey(KYRBS)over a period spanning from 2006 to *** focused on the prevalence of sexual intercourse,contraception utilization,and the underlying associated factors among this *** KYRBS data was collected using a complex sampling strategy to determine the national prevalence estimates and shifts in prevalence before(2006-2019)and during(2020-2022)the COVID-19 pandemic *** During the pre-pandemic period,a decrease in adolescent sexual intercourse was observed(6.34%in 2006,5.53%in 2012,and 5.87%in 2019).However,in the post-pandemic period(2020-2022),there was a surge in sexual intercourse(4.55%in 2020 and 6.20%in 2022).This evident alteration in sexual intercourse trajectory between pre-and post-pandemic periods was statistically significant[βdiff,0.950;95%confidence interval(CI),0.756-1.144].Regarding contraceptive use among South Korean adolescents,there was an increase in the pre-COVID-19 pandemic phase across all demographic segments(14.61%in 2006,22.30%in 2012,and 47.69%in 2022)but a notable decline when compared with the pre-and post-pandemic periods(βdiff,-0.319;95%CI,-0.454 to-0.184).Additionally,during the study period,a decrease in sexual intercourse was observed in the pre-pandemic period(β,-0.129;95%CI,-0.148 to-0.110),followed by an increase in the post-pandemic period(β,0.821;95%CI,0.627 to 1.014).This shift is highlighted by an effect size of 0.96[weighted odds ratio(wOR);95%CI,0.92 to 1.00],indicating a subst

Genetic susceptibility to metabolic associated fatty liver disease(MAFlD)is complex and poorly *** characterization of the genetic background of hepatic fat content would provide insights into disease etiology and causality of risk *** performed genome-wide association study(GWAS)on two noninvasive definitions of hepatic fat content:magnetic resonance imaging proton density fat fraction(MRI-pDFF)in 16,050 participants and fatty liver index(FlI)in 388,701 participants from the United Kingdom(UK)Biobank(UKBB).Heritability,genetic overlap,and similarity between hepatic fat content phenotypes were analyzed,and replicated in 10,398 participants from the University Medical Center Groningen(UMCG)Genetics lifelines Initiative(UGlI).Meta-analysis of GWASs of MRI-pDFF in UKBB revealed five statistically significant loci,including two novel genomic loci harboring CREB3l1(rs72910057-T,p=5.40E−09)and GCM1(rs1491489378-T,p=3.16E−09),respectively,as well as three previously reported loci:pNplA3,TM6SF2,and *** of FlI in UKBB identified 196 genome-wide significant loci,of which 49 were replicated in UGlI,with top signals in ZpR1(p=3.35E−13)and FTO(p=2.11E−09).Statistically significant genetic correlation(rg)between MRI-pDFF(UKBB)and FlI(UGlI)GWAS results was found(rg=0.5276,p=1.45E−03).Novel MRI-pDFF genetic signals(CREB3l1 and GCM1)were replicated in the FlI *** identified two novel genes for MRI-pDFF and 49 replicable loci for *** a difference in hepatic fat content assessment between MRI-pDFF and FlI,a substantial similar genetic architecture was *** is identified as an easy and reliable approach to study hepatic fat content at the population level.

Brine shrimp(Artemia)has existed on Earth for 400 million years and has major ecological importance in hypersaline *** a crucial live food in aquaculture,brine shrimp cysts have become one of the most important aquatic products traded ***,our understanding of the biodiversity,prevalence and global connectedness of viruses in brine shrimp is still very limited.A total of 143 batches of brine shrimp(belonging to seven species)cysts were collected from six continents including 21 countries and more than 100 geographic locations worldwide during 1977–*** total,55 novel RNA viruses were identified,which could be assigned to 18 different viral families and related *** viruses were dsRNA viruses,16 were+ssRNA viruses,and 28 were−ssRNA *** analyses of the RNA-directed RNA polymerase(RdRp)showed that brine shrimp viruses were often grouped with viruses isolated from other invertebrates and ***,most brine shrimp viruses were related to those from different hosts that might feed on brine shrimp or share the same ecological niche.A notable case was the novel brine shrimp noda-like virus 3,which shared 79.25%(RdRp)and 63.88%(capsid proteins)amino acid identity with covert mortality nodavirus(CMNV)that may cause losses in *** addition,both virome composition and phylogenetic analyses revealed global connectedness in certain brine shrimp viruses,particularly among Asia and Northern *** highlights the incredible species diversity of viruses in these ancient species and provides essential data for the prevalence of RNA viruses in the global aquaculture *** broadly,these findings provide novel insights into the previously unrecognized RNA virosphere in hypersaline ecosystems worldwide and demonstrate that human activity might have driven the global connectedness of brine shrimp viruses.

Hepatitis C virus core antigen testing: Role in diagnosis, disease monitoring and treatment

While hepatitis B virus(HBV)screening relies on hepatitis B surface antigen to confirm HBV infection since the early days of hepatitis B disease management,hepatitis C virus(HCV)infection screening is based on anti-HCV testing which does not discriminate active from past *** to confirm infection HCV RNA testing has been required;recently a HCV core antigen assay became widely commercially available which could serve to confirm *** assay is less sensitive than current HCV RNA assays,but as more than 50%of anti-HCV positive persons will be HCV core antigen positive,HCV core antigen testing can be a cost effective and reflex test to confirm HCV infection in anti-HCV positive individuals and will be easier as it can be applied on the same *** treatment monitoring,more data need to be generated,but the early data available at present suggest that HCV core antigen may be an alternative to HCV RNA *** direct antivirals,HCV core antigen could even be superior to HCV RNA testing,as direct antivirals might already prevent virus formation when HCV core antigen is still produced and thereby correlates better with eventual viral clearance.
